Bacillus megaterium strain B6 producing extracellular β-amylase was isolated and grown in a medium supplemented with waste potato starch. It showed highest enzyme synthesis in the early stationary phase. The partially purified β-amylase had a temperature optimum at 60°C and a pH optimum at 6.9 and was not affected by Schardinger dextrins. These properties would allow its application in sugar industry.  相似文献

The garnet-type phase Li7La3Zr2O12 (LLZO) attracts significant attention as an oxide solid electrolyte to enable safe and robust solid-state batteries (SSBs) with potentially high energy density. However, while significant progress has been made in demonstrating compatibility with Li metal, integrating LLZO into composite cathodes remains a challenge. The current perspective focuses on the critical issues that need to be addressed to achieve the ultimate goal of an all-solid-state LLZO-based battery that delivers safety, durability, and pack-level performance characteristics that are unobtainable with state-of-the-art Li-ion batteries. This perspective complements existing reviews of solid/solid interfaces with more emphasis on understanding numerous homo- and heteroionic interfaces in a pure oxide-based SSB and the various phenomena that accompany the evolution of the chemical, electrochemical, structural, morphological, and mechanical properties of those interfaces during processing and operation. Finally, the insights gained from a comprehensive literature survey of LLZO–cathode interfaces are used to guide efforts for the development of LLZO-based SSBs.  相似文献

The mechanism of cytoplasmic male sterility was investigated in maize by isolating mitochondria from seedlings and various anther stages and analyzing cytochrome oxidase and succinic dehydrogenase biochemically and electrophoretically. Sterile anthers exhibited a lack of biochemical activity and fewer isozymatic bands for cytochrome oxidase. No apparent differences were detected biochemically or electrophoretically between fertile and sterile anthers for succinate dehydrogenase.  相似文献

Germinability of the conidia of B. oryzae, after an efflux of substances — both electrolytes and non-electrolytes — was tested in sterilized solutions of plasmolytica and osmotica, ranging from molarity to 10?12 M. Maximum percent germination was recorded at 10?8 M concentration regardless of the exogenous supplement in the order KCl >D-glucose >Mannitol >NaCl. Germination was very poor or non-existent in other, higher or lower molarities tested except at 10?7 M wíth c. 50% germination. In nature the conidia, though nutrient independent, are subject to leaching and likely to lose viability; hence the possible role of the proper osmoticum, basing on the observed fact, in maintenance of membrane equilibrium and germinability of the leached conidia has been discussed.  相似文献

The multiple sex chromosome system, X1X2Y /X1X1X2X2, in the small Indian mongoose, Herpestes auropunctatus, results from a translocation of a part of Y chromosome to an autosome. It is not possible to distinguish the autosome which harbours the Y chromosome element in the somatic complement. By employing the surface-spreading technique to prophase I meiocytes we have identified the region to which the Y chromosome has been translocated as the short arm of chromosome 9 which is a subtelocentric chromosome. This Y chromosome component lacks heterochromatin and no sex vesicle is organised during meiotic prophase. This suggests to us that Y heterochromatin in mammals may be required for the production of a sex vesicle.We take great pleasure in dedicating this paper to our revered teacher Prof. S.P. Enhancement of the productivity of xylanase and beta-xy-losidase of Aspergillus ochraceus was investigated by multistep mutagenesis. The spores of the wild strain were subjected to UV and N-methyl-N-nitro-N-nitro-soguanidine (NTG). The hyperxylanolytic mutant (NG-13), which showed good clearing on the surface of the xylan-agar plate, secretes xylanase and beta-xylosidase at high levels during growth on commercial xylan and on agricultural wastes. Both liquid and solid state cultures were employed in the study for enzyme production. The xylanase from NG-13 was purified to homogeneity by ammonium sulfate precipitation and gel filtration. This purified enzyme showed a pH optimum of 6.0 and was stable in the range of pH 5 to 10. Prolonged stability of the enzyme was observed at 45 degrees C though its activity was maximal at 50 degrees C. The molecular weight of the enzyme was estimated to be 4.3 x 10(4) by SDS-polyacrylamide gel electrophoresis and 5 x 10(4) by gel filtration on Sephadex G-75. The kinetic data showed that the K(m) and V(max) values for xylan were 1 x 10(-3)M and 19.6 mumol/ min/mg protein, respectively. The enzyme was both more active and thermostable in the presence of K(+)and was inactivated by thiol reagents such as Hg(2+), p-hydroxymercuribenzoate (PHMB), 3', 5'-dithiobis (2'-nitrobenzoic acid) (DTNB), and N-ethylmaleimide (NEM).  相似文献

Summary Triple test cross progenies resulting from the crossing of three testers (Kloka, UP 368 and an f1 intermediate between them) and 24 varieties of bread wheat have been studied for plant height (cm), peduncle length (cm), ear length (cm), number of spikelets per spike and harvest index (ratio between economic and total yield). Epistasis was not significant for any of the characters studied. The testers were inadequate for plant height and for peduncle length although the testers varied considerably for these traits. Additive variance played a significant role in the inheritance of all the characters except number of spikelets per spike. The dominance variance was important for plant height, ear length and harvest index. The degree of dominance was in the over-dominance range for plant height. Complete dominance was operative for ear length, number of spikelets per spike and harvest index whereas for peduncle length only partial dominance was observed. The possibility of the isolation of the recombinants with high harvest index has been stressed.  相似文献

The ancestry of the cells in the hermaphrodite and male gonadal somatic structures of C. elegans has been traced from the two gonadal somatic progenitor cells (Z1 and Z4) that are present in the newly hatched larvae of both sexes. The lineages of Z1 and Z4 are essentially invariant. In hermaphrodites, they give rise to a symmetrical group of structures consisting of 143 cells, and in males, they give rise to an asymmetrical group of structures consisting of 56 cells. The male gonad can be distinguished from the hermaphrodite gonad soon after the first division of Z1 and Z4. However, the development of Z1 and Z4 in hermaphrodites shares several features in common with their development in males suggesting that the two programs are controlled by similar mechanisms. In the hermaphrodite lineage, a variability in the positions of two cells is correlated with a variability in the lineages of four cells. This variability suggests that cell-cell interaction may play a more significant role in organisms that develop by invariant lineages than has hitherto been considered. None of the somatic structures (e.g., uterus, spermatheca, vas deferens) develops as a clone of a single cell. Instead, cells that arise early in the Z1–Z4 lineage generally contribute descendants to more than one structure, and individual structures consist of descendants of more than one lineage.  相似文献
