The anti-malarial agent atovaquone specifically targets the cytochrome bc1 complex and inhibits the parasite respiration. Resistance to this drug, a coenzyme Q analogue, is associated with mutations in the mitochondrial cytochrome b gene. We previously reported atovaquone resistant mutations in Plasmodium berghei, in the first quinone binding domain (Qo1) of the cytochrome b gene (M133I and L144S) with V284F in the sixth transmembrane domain. However, in P. falciparum the most common mutations are found in the Qo2 region. To obtain a better model for biochemical and genetic studies, we have now extended our study to isolate a wider range of P. berghei resistant strains, in particular those in the Qo2. Here we report four new mutations (Y268N, Y268C, L271V and K272R), all in the Qo2 domain. Two of these mutations are convergent to codon 268 (nt802–804) drug-induced mutation in P. falciparum. 相似文献

Summary In order to examine its clinical efficacy, recombinant human interferon- (rIFN-) was instilled intravesically into 51 patients with superficial bladder cancer. Ten patients, who received intermittent intravesical instillation at a dose of (3–36) × 106 U rIFN- on days 1–3 every week, showed no response. Thirty-two patients received intravesical instillation at a dose of (3–36) × 106 U every day for 10–20 days. Eight patients showed partial response, indicating an efficacy rate of 25%. Nine patients received divided doses of 18 × 106 U twice a day every day for 10–20 days. Six patients showed partial response, indicating an efficacy rate of 67%. This value was significantly higher than that obtained by administering divided doses. The response to intravesical instillation therapy with rIFN- varies with treatment protocol. Frequent and longer exposure to rIFN- may induce better regression of superficial bladder cancer. Six incidences of side-effects were found in five cases (9.8%): pollakiuria in one, pain on micturition in two, fever in two, and eruption in one case. All of these side-effects were slight and reversible after drug withdrawal. Laboratory tests showed only a few changes with low severity. The virulent phenotype of Shigella requires loci on the chromosome as well as on the large virulence plasmid, and is regulated via a complex web of interactions amongst various chromosomal and large plasmid genes. To further investigate the role of chromosomal loci in virulence, we performed random Tn 10 mutagenesis in Shigella flexneri YSH6000T, and isolated an avirulent mutant (V3404) incapable of spreading throughout an epithelial cell monolayer. Although V3404 initially spread intercellularly at the same rate as the wild-type, it gradually slowed down and ceased spreading as a result of increasing defects in cell division, leading to the formation of long filamentous bacteria lacking septa, trapped within cells. In addition, the mutation affected the ability of V3404 to polymerize actin, a prerequisite for intra- and inter-cellular spreading ability. Sequencing of Tn 10 -flanking DNA revealed that the mutated gene, designated ispA (intracellular septation), was equivalent to a previously sequenced but uncharacterised gene of Escherichia coli located between trp and tonB . Using E. coli sequence data, we cloned the ispA gene from the YSH6000T chromosome and found that it complemented the V3404 mutation. Nucleotide sequencing and in vitro expression experiments revealed that ispA coded for a small (21 kDa), very hydrophobic protein. Prefoldin is a jellyfish-shaped hexameric chaperone that captures a protein-folding intermediate and transfers it to the group II chaperonin for correct folding. In this work, we characterized the organic solvent tolerance of Escherichia coli cells that overexpress prefoldin and group II chaperonin from a hyperthermophilic archeaum, Pyrococcus horikoshii OT3. The colony-forming efficiency of E. coli cells overexpressing prefoldin increased by 1,000-fold and decreased the accumulation of intracellular organic solvent. The effect was impaired by deletions of the region responsible for the chaperone function of prefoldin. Therefore, we concluded that prefoldin endows E. coli cells by preventing accumulation of intracellular organic solvent through its molecular chaperone activity. 相似文献

Cycling of soil carbon in the first year after a clear-felling was compared with that before the felling in a Japanese red
pine forest in Hiroshima Prefecture, west Japan. The daily mean temperature at the soil surface in summer was increased after
the felling in comparison to that before felling, and the water content of both the A0 layer and the surface mineral soil was decreased due to the loss of the forest canopy. The rate of weight loss of the A0 layer was reduced after felling. However, accumulation of the A0 layer rapidly decreased because of the lack of litter supply to the forest floor. Low soil respiration after felling was
mainly caused by the cessation of root respiration. Analysis of annual soil carbon cycling was then conducted using a compartment
model. The relative decomposition rate of the A0 layer decreased whereas that of humus and dead roots in mineral soil increased to some extent after felling. The accumulation

Aqueous phase diagrams were constructed for two new alkylglucosides with isoprenoid-type hydrophobic chains, viz. 1-O-beta-(3,7-dimethyloctyl)-D-glucopyranoside, beta-Glc(Ger), and 1-O-beta-(3,7,11,15-tetramethylhexadecyl)-D-glucopyranoside, beta-Glc(Phyt). In a low concentration regime, from 0.17 to 34 wt.% beta-Glc(Ger), the beta-Glc(Ger)/water system exhibits two phase, a dilute (L1dil) and a concentrated isotropic phase (L1con), coexistence region. Above about 62 wt.% beta-Glc(Ger), an Lalpha phase is formed. The extent of the L1dil + L1conc two-phase region decreases as temperature increases and totally disappears above 130 degrees C, exhibiting an upper critical temperature. The beta-Glc(Phyt)/water system exhibits an Lalpha phase above 78 wt.% surfactant below which, an Lalpha + water two-phase region appears. One notable feature of these compounds is their low values of Krafft-eutectic temperature, TK, e.g. the value of TK for beta-Glc(Phyt) is below 0 degrees C although the total number of carbon atoms in the hydrophobic chain is as large as 20. 相似文献

In Drosophila, the 255kDa catalytic subunit (dpolεp255) and the 58kDa subunit of DNA polymerase ε (dpolεp58) have been identified. The N-terminus of dpolεp255 carries well-conserved six DNA polymerase subdomains and five 3'→5' exonuclease motifs as observed with Polε in other species. We here examined roles of dpolεp255 during Drosophila development using transgenic fly lines expressing double stranded RNA (dsRNA). Expression of dpolεp255 dsRNA in eye discs induced a small eye phenotype and inhibited DNA synthesis, indicating a role in the G1-S transition and/or S-phase progression of the mitotic cycle. Similarly, expression of dpolεp255 dsRNA in the salivary glands resulted in small size and endoreplication defects, demonstrating a critical role in endocycle progression. In the eye disc, defects induced by knockdown of dpolεp255 were rescued by overexpression of the C-terminal region of dpolεp255, indicating that the function of this non-catalytic domain is conserved between yeast and Drosophila. However, this was not the case for the salivary gland, suggesting that the catalytic N-terminal region is crucial for endoreplication and its defect cannot be complemented by other DNA polymerases. In addition, several genetic interactants with dpolεp255 including genes related to DNA replication such as RFC, DNA primase, DNA polη, Mcm10 and Psf2 and chromatin remodeling such as Iswi were also identified. 相似文献
