Cancer stem cells (CSCs) are defined as a small population of cancer cells with the properties of high self-renewal, differentiation, and tumor-initiating functions. Recent studies have demonstrated that aldehyde dehydrogenase 1 (ALDH1) is a marker for CSCs in adult cancers. Although CSCs have been identified in some different types of pediatric solid tumors, there have been no studies regarding the efficacy of ALDH1 as a marker for CSCs. Therefore, in order to elucidate whether ALDH1 can be used as a marker for CSCs of pediatric sarcoma, we examined the characteristics of a population of cells with a high ALDH1 activity (ALDH1high cells) in rhabdomyosarcoma (RMS), the most common soft tissue sarcoma in children. We used the human embryonal RMS (eRMS) cell lines RD and KYM-1, and sorted the cells into two subpopulations of ALDH1high cells and cells with a low ALDH1 activity (ALDH1low cells). Consequently, we found that the ALDH1high cells comprised 3.9% and 8.2% of the total cell population, respectively, and showed a higher capacity for self-renewal and tumor formation than the ALDH1low cells. With regard to chemoresistance, the survival rate of the ALDH1high cells was found to be higher than that of the ALDH1low cells following treatment with chemotherapeutic agents for RMS. Furthermore, the ALDH1high cells exhibited a higher degree of pluripotency and gene expression of Sox2, which is one of the stem cell markers. Taken together, the ALDH1high cells possessed characteristics of CSCs, including colony formation, chemoresistance, differentiation and tumor initiation abilities. These results suggest that ALDH1 is a potentially useful marker of CSCs in eRMS.  相似文献

Summary The posterior part of the median eminence of the albino mouse (CF # 1-JCL) contains a cluster of myelinated axons beneath the tanycyte layer. Among them, small Herring bodies surrounded by myelin sheaths are revealed by electron microscopy. These structures contain electron-dense bodies, lamellar bodies, autophagic bodies, autophagic vacuoles, and neurofilaments. A few neurosecretory granules and mitochondria are also present. Summary The morphological features during development of diapause and non-diapause eggs of the silkworm,Bombyx mori, were investigated by means of light and electron microscopy, with special reference to eggs up to 24 h after oviposition.The blastoderm and yolk cells began to be formed about 6 and 24 h after oviposition, respectively, in both the diapause and non-diapause eggs, indicating that the diapause and non-diapause eggs develop at similar rates at least until 24 h after oviposition.Specific changes in the distribution of yolk granules were observed during early development of the diapause egg. Its yolk granules gradually aggregated into clusters from the periphery toward the inside of the egg during the period of blastoderm formation. Aggregation of yolk granules was most noticeable about 12 h after oviposition and then they dispersed again before yolk cell formation. On the other hand, yolk granules of the non-diapause eggs remained dispersed during development.  相似文献

Toxoplasma gondii is an intracellular parasite that invades nucleated cells, causing toxoplasmosis in humans and animals worldwide. The extremely wide range of hosts susceptible to T. gondii is thought to be the result of interactions between T. gondii ligands and receptors on its target cells. In this study, a host cell-binding protein from T. gondii was characterized, and one of its receptors was identified. P104 (GenBank Access. No. CAJ20677) is 991 amino acids in length, containing a putative 26 amino acid signal peptide and 10 PAN/apple domains, and shows low homology to other identified PAN/apple domain-containing molecules. A 104-kDa host cell-binding protein was detected in the T. gondii lysate. Immunofluorescence assays detected P104 at the apical end of extracellular T. gondii. An Fc-fusion protein of the P104 N-terminus, which contains two PAN/apple domains, showed strong affinity for the mammalian and insect cells evaluated. This binding was not related to protein-protein or protein-lipid interactions, but to a protein-glycosaminoglycan (GAG) interaction. Chondroitin sulfate (CS), a kind of GAG, was shown to be involved in adhesion of the Fc-P104 N-terminus fusion protein to host cells. These results suggest that P104, expressed at the apical end of the extracellular parasite, may function as a ligand in the attachment of T. gondii to CS or other receptors on the host cell, facilitating invasion by the parasite.  相似文献

Summary

In this work we have studied the possibility that cytoskeletal systems are involved in energid migration and blastoderm formation of the silkworm, Bombyx mori. The results obtained suggest: (1) energid migration is mainly controlled by microtubules and perhaps secondarily by microfilaments, (2) morphological changes of microprojections covering the egg surface are regulated by microfilaments and (3) the changes of microprojections are independent of energid migration.  相似文献

Almost all mammalian alphaherpesviruses can grow in cells derived from several types of animals in vitro . However, FHV-1 can only infect feline cell lines. For this reason, FHV-1 should be a good model to investigate species barriers to herpesviruses in vivo . To apply bacterial mutagenesis of FHV-1, we cloned the FHV-1 genome as a BAC. Using λ and flp recombinations, we introduced a monomeric red fluorescence protein into the C-terminus of glycoprotein D. Although GFP in the constructed recombinant FHV-1, a transfectant of the bacmid of FHV-1 that possessed the GFP, acted in non-feline cell lines, the virus could not enter non-feline cell lines, demonstrating that the host specificity of FHV-1 was restricted in an early step of infection. The host range of canine herpesvirus is limited to dogs in vitro and in vivo ; it cannot enter non-canine cell lines as a result of infection but the GFP is active by transfection, revealing the same result that the restriction step is at an early stage of infection. These results suggest the possibility of breaking species barriers of FHV-1 and CHV by modifying the gene(s) that act at the early stage of infection.  相似文献

Us3 is a serine-threonine protein kinase encoded by herpes simplex virus 1 (HSV-1). As reported here, we attempted to identify the previously unreported physiological substrate of Us3 in HSV-1-infected cells. Our results were as follows. (i) Bioinformatics analysis predicted two putative Us3 phosphorylation sites in the viral envelope glycoprotein B (gB) at codons 557 to 562 (RRVSAR) and codons 884 to 889 (RRNTNY). (ii) In in vitro kinase assays, the threonine residue at position 887 (Thr-887) in the gB domain was specifically phosphorylated by Us3, while the serine residue at position 560 was not. (iii) The phosphorylation of gB Thr-887 in Vero cells infected with wild-type HSV-1 was specifically detected using an antibody that recognized phosphorylated serine or threonine residues with arginine at the −3 and −2 positions. (iv) The phosphorylation of gB Thr-887 in infected cells was dependent on the kinase activity of Us3. (v) The replacement of Thr-887 with alanine markedly upregulated the cell surface expression of gB in infected cells, whereas replacement with aspartic acid, which sometimes mimics constitutive phosphorylation, restored the wild-type phenotype. The upregulation of gB expression on the cell surface also was observed in cells infected with a recombinant HSV-1 encoding catalytically inactive Us3. These results supported the hypothesis that Us3 phosphorylates gB and downregulates the cell surface expression of gB in HSV-1-infected cells.  相似文献

Foot-and-mouth disease caused by Foot-and-mouth disease virus (FMDV) is a severe and acute vesicular disease of cloven-hoofed animals including cattle, pigs, sheep and goats. As FMDV is highly contagious and causes productivity losses among infected animals, outbreaks of the disease are a primary animal health concern worldwide. In April, 2010, the disease reoccurred in Miyazaki prefecture in 10 years. Compared to the outbreak in 2000 in which no infection among pigs was observed, a total of 292 infected farms have been involved in this outbreak, and infected animals (37,412 cattle, 42 water buffalos, 174,132 pigs, 14 goats, and 8 sheep) were culled and buried. Vaccination was decided to reduce the speed of virus spreading. Finally a total of 76,756 heads of vaccinated animals were also slaughtered. The outbreak has continued for 2.5 months, and the ban on animal movements have been eased 3 months after the first occurrence. As several factors for disease spreading have been rumored, I would like to note this point and discuss future preventive measures.  相似文献
