Development of a highly sensitive in vitro assay method for biological activity of endotoxin contamination in biological products.

The pyrogen test in rabbits has been replaced by the bacterial endotoxin test. The endotoxin test, however, showed a considerable discrepancy with pyrogenicity and was, therefore, assumed to have an efficacy limitation in directly predicting harmful biological effects of endotoxin. We developed a sensitive in vitro assay method by making use of tumour necrosis factor alpha (TNF-alpha) induction in RAW264.7 cells, which showed a fine correlation with pyrogenicity in rabbits. RAW264.7 cells maintained by serial subculture under an endotoxin-free condition have gained the similar level of sensitivity as the endotoxin test to allow extensive dilutions of a drug for eliminating adverse effects on the cells. The in vitro TNF-alpha induction assay was shown to be capable to detect quantitatively a synergistic effect of a drug and endotoxin. The synergy is assumed necessary to be taken into consideration to define the limit value for the endotoxin test for guaranteeing the similar level of safety as by the pyrogen test.     

Detection of Corynebacterium kutscheri from the oral cavity of rats.

A simple and useful method for the detection of C. kutscheri from the oral cavity of living rats was devised. In 10 sacrificed rats from two naturally and subclinically infected conventional colonies, 10(4.28) or 10(3.84) CFU/ml C. kutscheri were isolated from upper incisor swab extractions, while 10(1.38) or 10(1.58) and < 10 or 10(1.56) CFU/ml from the upper soft palate and pharynx, respectively. In another survey with 26 living animals, which were reared on the same rack, organisms were detected from the upper incisor and gingival swabs in 15 of 26 rats (57.7%). The results were reproducible at a second survey 10 days later. No organisms were isolated from any sites of the orally negative rats. These results indicated that culture of swab specimens from the upper incisors and gingivae of incisors is useful for the detection of C. kutscheri infection in living rats.     

Further Evidence for Inactivation of Fructose-1,6-bisphosphatase at the Beginning of SO2 Fumigation. Increase in Fructose-1,6-bisphosphate and Decrease in Fructose-6-phosphate in SO2-Fumigated Spinach Leaves

The substrate level of the photosynthetic reductive pentosephosphate cycle in spinach leaves during SO₂ fumigation wassurveyed. At the beginning of SO₂ fumigation, fructose-1,6-bisphosphateincreased and fructose-6-phosphate decreased, while ribulose-1,5-bisphosphateremained unchanged and 3-phosphoglyceric acid rapidly decreased.These results suggested that the inhibition of photosynthesisin spinach leaves with SO₂ might be due to inactivation of fructose-1,6-bisphosphatase. An improved intrafemoral injection with minimized leakage as an orthotopic mouse model of osteosarcoma

Osteosarcoma, the most common type of primary bone cancer, is the second highest cause of cancer-related death in pediatric patients. To understand the mechanisms behind osteosarcoma progression and to discover novel therapeutic strategies for this disease, a reliable and appropriate mouse model is essential. For this purpose, osteosarcoma cells need to be injected into the bone marrow. Previously, the intratibial and intrafemoral injection methods were reported; however, the major drawback of these methods is the potential leakage of tumor cells from the injection site during or after these procedures. To overcome this, we have established an improved method to minimize leakage in an orthotopic mouse model of osteosarcoma. By taking advantage of the anatomical benefits of the femur with less bowing and larger medullary cavity than those of the tibia, osteosarcoma cells are injected directly into the femoral cavity following reaming of its intramedullary space. To prevent potential leakage of tumor cells during and after the surgery, the injection site is sealed with bone wax. This method requires a minor surgery of approximately 15 min under anesthesia. Our established orthotopic osteosarcoma model could serve as a valuable and reliable tool for examining progression of various types of bone tumors.     

Effects of different light intensities during the forenoon on the afternoon thermal sensation in mild cold

1. 1. The study aimed at knowing whether thermal sensation during afternoon cool exposure could be influenced by bright light (4000 lx) or dim light (200 lx) in the forenoon.

2. 2. The subjects felt cooler after exposure to dim light than to bright light.

3. 3. Melatonin in the urine was significantly higher in bright light than in dim light at 10:30 h and at noon.
