A subcellular fractionation study was carried out onStreptococcus mitis ATCC 9811 to determine the location of the arginine deiminase pathway. Arginine deiminase activity was detected in the cell wall fraction, ornithine carbamoyltransferase activity was recovered in both the cell wall and cytoplasmic fractions, and carbamate kinase activity was detected in the cytoplasmic fraction.  相似文献

The effectiveness of a 70-MeV proton beam in the induction of chromosome aberrations was studied. We employed peripheral lymphocytes and analyzed the frequencies of dicentrics and rings after irradiation at doses ranging from 0.1 to 8.0 Gy at various depths within a Lucite phantom. The frequency of chromosome aberrations after irradiation with an unmodulated proton beam at 5 mm showed a dose-response relationship similar to that of 60Co gamma rays. However, irradiation at greater depths with the spread-out Bragg peak induced higher aberration frequencies at doses lower than those with gamma rays. Furthermore, the distribution curve of chromosome aberration frequencies as a function of depth was found to be slightly different from the physically measured depth-dose curve. With the spread-out Bragg peak the biological effects were more marked at greater depths, resulting in a distribution of relative biological effectiveness values. The results obtained from chromosome aberration analysis may not be related directly to those for the relationship between dose and cell killing. Slight differences in values for relative biological effectiveness due to the change of dose and site of proton beam irradiation may not be important for practical proton beam therapy, but may be important in the prevention of late radiation injuries.  相似文献

The objective of the present study was to investigate the effects of rhythmic arm swing on ipsilateral and contralateral soleus motoneuron pool excitability. Ten healthy human subjects participated in this study. Soleus H-reflexes were recorded from the ipsilateral and contralateral soleus muscles while the subject swung the right arm anteroposteriorly as if during gait. The soleus H-reflex was depressed throughout the whole arm swing cycle except in the ipsilateral leg during the onset of the backward arm swing, and in the contralateral leg during the last half of the backward arm swing and the onset of the forward arm swing. The depression was cyclically modulated in accordance with the time course of the arm swing periods, and the pattern of the modulation was reciprocal between the ipsilateral and contralateral legs. This cyclical and reciprocal modulation may be related to the regulation of soleus motoneuron pool excitability during gait.  相似文献

Reinforcement learning (RL) for a linear family of tasks is described in this paper. The key of our discussion is nonlinearity of the optimal solution even if the task family is linear; we cannot obtain the optimal policy using a naive approach. Although an algorithm exists for calculating the equivalent result to Q-learning for each task simultaneously, it presents the problem of explosion of set sizes. The green macroalga Ulva L. is well known as having an alternation of isomorphic biphases: gametophyte and sporophyte. However, an examination of the temporal alternation in phase dominance has not been carried out. By inducing reproduction of thallus samples in the laboratory, this study reports the temporal changes in the two phases and sex ratios in a natural population of Ulva pertusa Kjellm. in the Seto Inland Sea, Japan, over a period of >3 years. The results showed that a temporal alternation in phase dominance occurred after 11–20 months; seasonal changes in phase dominance were not observed in the Ulva population. The sex ratio of male and female gametophytes remained ~1:1 throughout the investigated period, although skewed sex ratios were observed in several collections.  相似文献

In this study, three fluoroquinolones, pazufloxacin, ciprofloxacin and levofloxacin, were simultaneously determined in spiked human serum by high-performance liquid chromatography (HPLC) method with fluorescence detection. Chromatography was performed using a C8 column with an isocratic mobile phase consisting of 1% triethylamine (pH 3.0)/acetonitrile (86/14, v/v). Protein precipitation was conducted using perchloric acid and methanol. The calibration curves for the three fluoroquinolones were linear over concentrations ranging from 0.1 to 20.0 μg/mL. The within-day and between-day coefficients of variation obtained from three fluoroquinolones were less than 7%, and relative errors ranged from −1.6% to 9.3%. Mean recoveries of pazufloxacin, ciprofloxacin, and levofloxacin from spiked human serum were 97%, 88%, and 90%, respectively. The proposed method proved to be simple and reliable for the determination of three fluoroquinolones.  相似文献

Although the endothelial expression of various adhesion molecules substantially differs between pulmonary microvessels, their importance for neutrophil and lymphocyte sequestration in ventilator-induced lung injury (VILI) has not been systematically analyzed. We investigated the kinetics of polymorphonuclear cells (PMN) and mononuclear cells (MN) in the acinar microcirculation of the isolated rat lung with VILI by real-time confocal laser fluorescence microscopy, with or without inhibition of ICAM-1, VCAM-1, or P-selectin by monoclonal antibodies (MAb). Adhesion molecules in each microvessel were estimated by intravital fluorescence microscopy or immunohistochemical staining. In high tidal volume-ventilated lungs, 1) ICAM-1, VCAM-1, and P-selectin were differently upregulated in venules, arterioles, and capillaries; 2) venular PMN rolling was improved by inhibition of ICAM-1, VCAM-1, or P-selectin, whereas arteriolar PMN rolling was improved by ICAM-1 or VCAM-1 inhibition; 3) capillary PMN entrapment was ameliorated only by anti-ICAM-1 MAb; and 4) MN rolling in venules and arterioles and MN entrapment in capillaries were improved by ICAM-1 and VCAM-1 inhibition. In conclusion, the contribution of endothelial adhesion molecules to abnormal leukocyte behavior in VILI-injured microcirculation is microvessel and leukocyte specific. ICAM-1- and VCAM-1-dependent, but P-selectin-independent, arteriolar PMN rolling, which is expected to reflect the initial stage of tissue injury, should be taken as a phenomenon unique to ventilator-associated lung injury.  相似文献
