Background
Fibroadenoma is the most common benign solid breast lesion type and a very common cause for histologic assessment. To justify a conservative therapy, a highly specific discrimination between fibroadenomas and other breast lesions is crucial. Phase-contrast imaging offers improved soft-tissue contrast and differentiability of fine structures combined with the potential of 3-dimensional imaging. In this study we assessed the potential of grating-based phase-contrast CT imaging for visualizing diagnostically relevant features of fibroadenomas.Materials and Methods
Grating-based phase-contrast CT was performed on six ex-vivo formalin-fixed breast specimens containing a fibroadenoma and three samples containing benign changes that resemble fibroadenomas using Talbot Lau interferometry and a polychromatic X-ray source. Phase-contrast and simultaneously acquired absorption-based 3D-datasets were manually matched with corresponding histological slices. The visibility of diagnostically valuable features was assessed in comparison with histology as the gold-standard.Results
In all cases, matching of grating-based phase-contrast CT images and histology was successfully completed. Grating-based phase-contrast CT showed greatly improved differentiation of fine structures and provided accurate depiction of strands of fibrous tissue within the fibroadenomas as well as of the diagnostically valuable dilated, branched ductuli of the fibroadenomas. A clear demarcation of tumor boundaries in all cases was provided by phase- but not absorption-contrast CT.Conclusions
Pending successful translation of the technology to a clinical setting and considerable reduction of the required dose, the data presented here suggest that grating-based phase-contrast CT may be used as a supplementary non-invasive diagnostic tool in breast diagnostics. Phase-contrast CT may thus contribute to the reduction of false positive findings and reduce the recall and core biopsy rate in population-based screening. Phase-contrast CT may further be used to assist during histopathological workup, offering a 3D view of the tumor and helping to identify diagnostically valuable tissue sections within large tumors. 相似文献53.

Trekking poles are used by hikers for improved stability and lowered leg fatigue due to increased upper body muscle involvement. However, the weight of the poles and exaggerated upper body movement when using poles may increase total energy expenditure at a given walking speed. Few studies have investigated the physiological responses of hiking with trekking poles outside the laboratory setting. The purposes of this study were to determine if trekking poles altered physiological responses to hiking on varied terrain, and whether responses between trials were dependent on the grade of the terrain. Fourteen recreational hikers completed four hiking trials over a course that included sustained sections of flat (0 +/- 1% grade), steep uphill (>10% grade), gradual uphill (5% grade), gradual downhill (-5% grade) and steep downhill (<-10% grade) terrain. Subjects walked at a self-selected speed that was matched across trials using time-splits and a metronome. Two trials were conducted with hiking poles and two without poles. [latin capital V with dot above]O2 was significantly elevated (p <0.05) during the pole trials (1502.9 +/- 510.7 ml/min) compared to the no-pole trials (1362.4 +/- 473.2 ml/min). Similarly, ventilatory efficiency ([latin capital V with dot above]E) (43.1 +/- 9.6; 38.3 +/- 10.1 L/min) and heart rate (HR) (112.1 +/- 9.7; 105.7 +/- 10.4 bt/min) were significantly higher during the pole trials than the no-pole trials. However, ratings of perceived exertion (RPE) was not altered by pole condition (8.5 +/- 0.7; 8.4 +/- 0.8). Comparisons within each grade revealed significantly higher physiological responses for [latin capital V with dot above]O2, [latin capital V with dot above]E and HR in the pole-condition at all grades, with no significant variable*grade interactions. RPE measures were not significantly different between pole trials at any grade. These data suggest that trekking poles may be a beneficial tool for increasing caloric expenditure, as energy production increased during exercise without increased perceptions of effort. 相似文献

SIVQ-LCM is a new methodology that automates and streamlines the more traditional, user-dependent laser dissection process. It aims to create an advanced, rapidly customizable laser dissection platform technology. In this report, we describe the integration of the image analysis software Spatially Invariant Vector Quantization (SIVQ) onto the ArcturusXT instrument. The ArcturusXT system contains both an infrared (IR) and ultraviolet (UV) laser, allowing for specific cell or large area dissections. The principal goal is to improve the speed, accuracy, and reproducibility of the laser dissection to increase sample throughput. Affinity cross-linking of 125I-labeled recombinant human interleukin-6 (IL-6) to human hepatoma cells (HepG2) allowed the detection of three IL-6-containing complexes with molecular masses of 100 kDa, 120 kDa and 200 kDa. Treatment of HepG2 cells with dexamethasone led to a time- and dose-dependent up-regulation of IL-6-receptor mRNA levels. By the use of cross-linking this effect was also seen at the protein level, where all three IL-6-binding complexes increased upon incubation of HepG2 cells with dexamethasone. Under conditions of IL-6-receptor up-regulation by dexamethasone, gamma-fibrinogen mRNA induction by IL-6 is stronger and occurs earlier than without dexamethasone. We propose therefore that the expression of the IL-6 receptor might be a rate-limiting step in acute-phase-protein induction. 相似文献

The megadiverse genus Carex (c. 2000 species, Cyperaceae) has a nearly cosmopolitan distribution, displaying an inverted latitudinal richness gradient with higher species diversity in cold‐temperate areas of the Northern Hemisphere. Despite great expansion in our knowledge of the phylogenetic history of the genus and many molecular studies focusing on the biogeography of particular groups during the last few decades, a global analysis of Carex biogeography and diversification is still lacking. For this purpose, we built the hitherto most comprehensive Carex‐dated phylogeny based on three markers (ETS–ITS–matK), using a previous phylogenomic Hyb‐Seq framework, and a sampling of two‐thirds of its species and all recognized sections. Ancestral area reconstruction, biogeographic stochastic mapping, and diversification rate analyses were conducted to elucidate macroevolutionary biogeographic and diversification patterns. Our results reveal that Carex originated in the late Eocene in E Asia, where it probably remained until the synchronous diversification of its main subgeneric lineages during the late Oligocene. E Asia is supported as the cradle of Carex diversification, as well as a “museum” of extant species diversity. Subsequent “out‐of‐Asia” colonization patterns feature multiple asymmetric dispersals clustered toward present times among the Northern Hemisphere regions, with major regions acting both as source and sink (especially Asia and North America), as well as several independent colonization events of the Southern Hemisphere. We detected 13 notable diversification rate shifts during the last 10 My, including remarkable radiations in North America and New Zealand, which occurred concurrently with the late Neogene global cooling, which suggests that diversification involved the colonization of new areas and expansion into novel areas of niche space. 相似文献

Leaf epidermal features are important taxonomic features in Quercus. We studied leaf epidermal features of 91 species and one forma of Quercus subgenus Cyclobalanopsis using light and scanning electron microscopy. Trichome terminology in oaks was assessed and clarified, aiming to score the epidermal features accurately for genus‐wide comparison. Nine trichome types, anomocytic and cyclocytic stomatal apparatus, smooth layer and platelet epicuticular wax flakes, and two trichome base types were found in subgenus Cyclobalanopsis. The epidermal features revealed three main groups in subgenus Cyclobalanopsis. The epidermal features of Quercus s.l. were uniform, supporting recognition of Cyclobalanopsis as a subgenus of Quercus rather than as a separate genus. Most leaf epidermal features show a mosaic pattern, although their numerous variations offered valuable resources for species identification. There is often an inverse relationship between the diversity of a plant community and the invasibility of that community by non-native plants. Native herbivores that colonize novel plants may contribute to diversity–invasibility relationships by limiting the relative success of non-native plants. Here, we show that, in large collections of non-native oak trees at sites across the USA, non-native oaks introduced to regions with greater oak species richness accumulated greater leaf damage than in regions with low oak richness. Underlying this trend was the ability of herbivores to exploit non-native plants that were close relatives to their native host. In diverse oak communities, non-native trees were on average more closely related to native trees and received greater leaf damage than those in depauperate oak communities. Because insect herbivores colonize non-native plants that are similar to their native hosts, in communities with greater native plant diversity, non-natives experience greater herbivory. 相似文献
