To a large extent, the mutual affinities of the mammalian orders continue to puzzle systematists, even though comparative anatomy and amino acid sequencing offer a massive data base from which these relationships could potentially be adduced. In the present paper the consistency index--the number of character states less the number of characters in a data set, divided by the total number of changes in the character states on a cladogram--was used to examine the relative resolving powers of recently published morphological and molecular- sequence data. Consistency indices were calculated for previously published alpha crystallin A chain and myoglobin amino acid-sequence cladograms and for four original amino acid-sequence cladograms (alpha crystallin A, myoglobin, and alpha and beta hemoglobin); these were found to be comparable to the consistency indices of morphologically based cladograms. Qualitative comparisons between the morphologically based and molecularly based trees were also made; only moderate congruence between the two was observed. Moreover, there was a general lack of congruence between the cladograms specified by each of the four proteins. Amino acid-sequence and morphological data agreed on the placement of edentates as an early eutherian offshoot and on the grouping of hyracoids, proboscideans, and sirenians. Otherwise there was only limited congruence: morphology strongly supported the grouping of lagomorphs and rodents and the alliance of pholidotes and edentates, but sequence analyses did not. The placement of tubulidentates differed widely among proteins. Morphology indicated the close association of sirenians with proboscideans; proteins suggested a pairing of sirenians with hyracoids. The sex ratio at hatching in broods of Tengmalms owl (Aegolius funereus) in northern Sweden was investigated for 3 years characterized by different phases of the vole and owl cycle. Previous work showed the sex ratio in this species to be male-biased for 1 year with a favourable food supply, and that in feeding experiments male nestlings (but not females) suffered higher mortality when food was limited, but not otherwise. Here we present data from a complete 3-year owl cycle, showing that mean brood sex ratio varied significantly among years, being male-biased (65% males) in the first year of high owl breeding density, unbiased (49%) in the second year of high owl breeding density, and female-biased (33% males) in the owls low year. Brood sex ratio did not vary significantly within years with laying date or parental age. Vole availability, and therefore the owls food supply, declined during the 3 years studied. Tengmalms owl parents thus appear to adaptively adjust the sex ratio of their broods according to the expected annual mortality risk of sons.  相似文献

The aim of the present study is to assess the possible protective effects of thymol and carvacrol against cisplatin (CP)‐induced nephrotoxicity. A single dose of CP {6 mg/kg, intraperitoneally (i.p.)} injected to male rats revealed significant increases in serum urea, creatinine, and tumor necrosis factor alpha levels. It also increased kidney contents of malondialdehyde and caspase‐3 activity with significant reduction in serum albumin, kidney content of reduced glutathione as well as catalase, and superoxide dismutase activity as compared to that of the control group. In contrast, administration of thymol {20 mg/kg, orally (p.o.)} and/or carvacrol (15 mg/kg, p.o.) for 14 days before CP injection and for 7 days after CP administration restored the kidney function and examined oxidative stress parameters. In conclusion, thymol was more effective nephroprotective than carvacrol. Moreover, a combination of thymol and carvacrol had a synergistic nephroprotective effect that might be attributed to antioxidant, anti‐inflammatory, and antiapoptotic activities.  相似文献

The influence of the phenothiazine trifluoperazine (Stellazine) on the rat tapeworm Hymenolepis diminuta was examined. The parasite was incubated in glucose-containing Krebs-Ringer media (pH 7.4) at 37 degrees C which included Ca2+ or EGTA and a range of trifluoperazine concentrations (0-2 mM). Release of soluble protein and lactate dehydrogenase activity were taken as measures of release of cytosolic components. The release of lactate dehydrogenase depended on drug concentration, maximum levels occurring at 2 mM trifluoperazine, this corresponded to 2% of the total lactate dehydrogenase present in the cestode. The effect of phenothiazines of differing lipophilicity were compared, and for trifluoperazine sulfoxide only minimal amounts of lactate dehydrogenase activity and protein were released. These values were similar to those obtained when H. diminuta was incubated in drug-free media. Our findings suggest that the integrity of the parasite is related to its calmodulin content. The potential cestocidal properties of trifluoperazine are considered.  相似文献

We previously showed that radioactive N-ethylmaleimide injected intramuscularly reacts with actomyosin and other muscle proteins and that a transfer of these modified proteins to lysosome-rich large granules was associated with their degradation (Gerard, K. W., and Schneider, D. L., (1979) J. Biol. Chem. 254, 11798-11805). We now show that muscle cells, when challenged by an increase in proteins modified with N-ethylmaleimide, can increase degradation by increasing the activities of enzymes involved in protein turnover. Cathepsin B activity increased 2-fold 36 h after injection of N-ethylmaleimide. In contrast, non-lysosomal proteolytic enzymes, calcium-dependent protease, and leucine aminopeptidase, did not significantly increase. Lysosomes are also involved in the degradation of normal muscle proteins labeled with [3H]leucine. Treatment with chloroquine, a known inhibitor of lysosome function, resulted in an inhibition of protein degradation, in an increase of the muscle protein content, and in the accumulation of radioactive proteins in lysosomal fractions. Chloroquine treatment for 2 days led to a 270% increase in cathepsin B and a 160% increase in lysosomal ATPase activities, but only a 30% increase in neutral proteinase activities. These results indicate a role for lysosomes in regulation of protein turnover in muscle.  相似文献

The tumour microenvironment and genetic alterations collectively influence drug efficacy in cancer, but current evidence is limited and systematic analyses are lacking. Using chronic lymphocytic leukaemia (CLL) as a model disease, we investigated the influence of 17 microenvironmental stimuli on 12 drugs in 192 genetically characterised patient samples. Based on microenvironmental response, we identified four subgroups with distinct clinical outcomes beyond known prognostic markers. Response to multiple microenvironmental stimuli was amplified in trisomy 12 samples. Trisomy 12 was associated with a distinct epigenetic signature. Bromodomain inhibition reversed this epigenetic profile and could be used to target microenvironmental signalling in trisomy 12 CLL. We quantified the impact of microenvironmental stimuli on drug response and their dependence on genetic alterations, identifying interleukin 4 (IL4) and Toll‐like receptor (TLR) stimulation as the strongest actuators of drug resistance. IL4 and TLR signalling activity was increased in CLL‐infiltrated lymph nodes compared with healthy samples. High IL4 activity correlated with faster disease progression. The publicly available dataset can facilitate the investigation of cell‐extrinsic mechanisms of drug resistance and disease progression.  相似文献
