Herein, we discuss the role of the native cysteines present in a major multidrug ABC transporter of Candida albicans, Cdr1p, and describe the construction of this transporter's functional cysteine-less (cysless) protein version for cross-linking studies. In the experiments in which all 23 cysteines were replaced individually, we observed that most of the cysteine replacements were tolerated by the protein, but the replacement of C1056, C1091, C1106, C1294 or C1336 resulted in an enhanced drug susceptibility together with an abrogated drug efflux. Notably, the ATPase activity was uncoupled, which largely remained unaffected in these variants. The substitution of the critical cysteines with serines restored the normal expression and functionality of Cdr1p because serine can effectively mimic the hydrogen bonding properties of cysteine. Finally, we constructed a functional cysless His-tagged Cdr1p in which all the cysteines of the native protein were replaced with alanines and the critical cysteines were replaced with serines. Notably, cysless GFP-tagged variant of Cdr1p was non-functional. The cysless His-tagged variant of Cdr1p is the first example of a cysless ABC transporter in yeast, and it will lead to a greater understanding of the architecture of this important protein and provide insight into the nature of drug binding and interdomain communication. 相似文献

In the present study D. discoideum has been used as a model organism to understand the role of poly (ADP-ribose) polymerase (PARP) in caspase independent paraptotic cell death pathways. D. discoideum lacks caspases and Bcl-2 family proteins; nevertheless it has 9 potential genes for PARP. PARP has been known to get activated in various cell death associated diseases. In this study kinetics of cell death induced by staurosporine (STS), a bacterial alkaloid, was established to unravel the role of PARP. It was found that STS induced cell death in D. discoideum did not involve PARP activation, however it involved cathepsin D. Results indicated that an alternative mechanism may be existing in D. discoideum that lacks Bcl-2 family proteins for STS induced cell death that evades Bax involvement. 相似文献

Enterobacter cloacae (DGE50) and Escherichia coli (DGE57) were used throughout the study. Samples were collected from contaminated marine water and soil to isolate bacterial
strains having tolerance against heavy metals and antimicrobial agents. We have demonstrated plasmid transfer, from Amp+Cu+Zn− strain (DGE50) to Amp−Cu−Zn+ strain (DGE57), producing Amp+Cu+Zn+ transconjugants (DGETC50→57) and Amp+Cu−Zn+ transformants (DGETF50→57). DGE57 did not carry any plasmid, therefore, it can be speculated that zinc tolerance gene in DGE57 is located on chromosome.
DGE50 was found to carry three plasmids, out of which two were transferred through conjugation into DGE57, and only one was
transferred through transformation. Plasmid transferred through transformation was one out of the two transferred through
conjugation. Through the results of transformation it was revealed that the genes of copper and ampicillin tolerance in DGE50
were located on separate plasmids, since only ampicillin tolerance genes were transferred through transformation as a result
of one plasmid transfer. By showing transfer of plasmids under lab conditions and monitoring retention of respective phenotype
via conjugation and transformation, it is very well demonstrated how multiple stress tolerant strains are generated in nature. 相似文献

Hypoxia is a common environmental stress that influences signaling pathways and cell function. Previous studies from our laboratory have identified significant differences in cellular responses to sustained or intermittent hypoxia with the latter proving more cytotoxic. We hypothesized that differences in susceptibility of neurons to intermittent (IH) and sustained hypoxia (SH) are mediated by altered Akt signaling. SH, but not IH, induced a significant increase in Akt activation in rat CA1 hippocampal region extracts compared with room air controls. Akt immunoprecipitations followed by proteomic analysis identified valosin-containing protein (VCP) as an Akt-binding protein. In addition, VCP expression and association with Akt was enhanced during SH, and this association was decreased upon phosphoinositide 3-kinase/Akt pathway blockade with LY294002. Active recombinant Akt phosphorylated recombinant VCP in vitro. Site-directed mutagenesis studies identified Ser352, Ser746, and Ser748 as Akt phosphorylation sites on VCP. In addition, rat CA1 hippocampal tissue exposed to SH exhibited an acidic pI shift of VCP. Protein phosphatase 2A treatment inhibited this acidic shift consistent with SH-induced phosphorylation of VCP in vivo. PC-12 cells transfected with active Akt, but not dominant negative Akt or vector, induced VCP expression and an acidic shift in VCP pI, which was inhibited by protein phosphatase 2A treatment. Furthermore, VCP association with ubiquitinated proteins was demonstrated in vector-transfected PC-12 cell lysates, whereas active Akt-transfected cells demonstrated a marked decrease in association of VCP with ubiquitinated proteins. We concluded that Akt phosphorylates VCP in vitro and in vivo, and VCP phosphorylation releases it from ubiquitinated substrate protein(s) possibly allowing ubiquitinated protein(s) to be degraded by the proteosome. 相似文献

We used gel filtration chromatography, anion-exchange chromatography and polyacrylamide gel electrophoresis to purify tributyltin-binding protein type 2 (TBT-bp 2) from plasma of Japanese flounder (Paralichthys olivaceus) injected intraperitoneally with TBT (5.0 mg/kg body weight). Sodium dodecyl sulfate-polyacrylamide gel electrophoresis indicated that the molecular mass of TBT-bp 2 was approximately 48 kDa, and isoelectric focusing-polyacrylamide gel electrophoresis indicated that the isoelectric point was approximately 3.0. TBT-bp 2 contained 40% N-glycan. The complete cDNA nucleotide sequence and the genome sequence of TBT-bp 2 were determined by means of rapid amplification of cDNA ends of liver tissue of Japanese flounder and a genome-walking technique, respectively. The 216 amino acid sequence of TBT-bp 2 showed 47% identity to the sequences of puffer fish (Takifugu pardalis) saxitoxin- and tetrodotoxin-binding protein but only 27% similarity to the sequence of TBT-bp 1. Analysis of the motif sequence of the amino acid sequence and the structure of the gene encoding TBT-bp 2 suggested that this protein belongs to the lipocalin superfamily. 相似文献

Effect of salinisation of soil on growth, water status and general nutrient accumulation in seedlings .of Delonix regia (Fabaceae) 下载免费PDF全文
Greenhouse experiments were conducted to assess the effect of salinisation of soil on emergence, growth, water content, proline content and mineral accumulation of seedlings of Delonix regia (Hook.) Raf. (Fabaceae). Sodium chloride (NaCl) was added to the soil and salinity was maintained at 0.3, 1.9, 3.9, 6.0 and 7.9 dS m?1. A negative relationship between seedling emergence and salt concentration was obtained. Salinity caused reduction in water content and water potential of tissues (leaves, stems, tap roots and lateral roots) that resulted in internal water deficit to plants. Consequently, shoot and root elongation, leaf expansion and dry matter accumulation in leaves, stems, tap roots and lateral root tissues of seedlings significantly decreased in response to increasing concentration of salt. Proline content in tissues was very low. There were no effective mechanisms to control net uptake of Na on root plasma membrane and subsequently its transport to shoot tissues. Potassium content significantly decreased in tissues in response to salinisation of soil. This tree species is a moderate salt-tolerant glycophytic plant. Nitrogen and calcium content in tissues significantly decreased as soil salinity increased. Phosphors content in tissues exhibited a declining trend with increase in soil salinity. Changes in tissues and whole-plant accumulation pattern of other elements tested, as well as possible mechanisms for avoidance of Na toxicity in this tree species in response to salinisation, are discussed. 相似文献
