Helicobacter pylori is a pathogen that colonizes the stomach and causes chronic gastritis. Helicobacter pylori can colonize deep inside gastric glands, triggering increased R‐spondin 3 (Rspo3) signaling. This causes an expansion of the “gland base module,” which consists of self‐renewing stem cells and antimicrobial secretory cells and results in gland hyperplasia. The contribution of Rspo3 receptors Lgr4 and Lgr5 is not well explored. Here, we identified that Lgr4 regulates Lgr5 expression and is required for H. pylori‐induced hyperplasia and inflammation, while Lgr5 alone is not. Using conditional knockout mice, we reveal that R‐spondin signaling via Lgr4 drives proliferation of stem cells and also induces NF‐κB activity in the proliferative stem cells. Upon exposure to H. pylori, the Lgr4‐driven NF‐κB activation is responsible for the expansion of the gland base module and simultaneously enables chemokine expression in stem cells, resulting in gland hyperplasia and neutrophil recruitment. This demonstrates a connection between R‐spondin‐Lgr and NF‐κB signaling that links epithelial stem cell behavior and inflammatory responses to gland‐invading H. pylori.  相似文献

Biological traits analysis (BTA) links community structure to both ecological functions and response to environmental drivers through species’ attributes. In consequence, it has become a popular approach in marine benthic studies. However, BTA will reach a dead end if the scientific community does not acknowledge its current shortcomings and limitations: (a) uncertainties related to data origins and a lack of standardized reporting of trait information; (b) knowledge gaps on the role of multiple interacting traits on driving the organisms’ responses to environmental variability; (c) knowledge gaps regarding the mechanistic links between traits and functions; (d) a weak focus on the spatial and temporal variability that is inherent to the trait expression of species; and, last but not least, (e) the large reliance on expert knowledge due to an enormous knowledge gap on the basic ecology of many benthic species. BTA will only reach its full potential if the scientific community is able to standardize and unify the reporting and storage of traits data and reconsider the importance of baseline observational and experimental studies to fill knowledge gaps on the mechanistic links between biological traits, functions, and environmental variability. This challenge could be assisted by embracing new technological advances in marine monitoring, such as underwater camera technology and artificial intelligence, and making use of advanced statistical approaches that consider the interactive nature and spatio‐temporal variability of biological systems. The scientific community has to abandon some dead ends and explore new paths that will improve our understanding of individual species, traits, and the functioning of benthic ecosystems.  相似文献

Mycobacterium avium subsp. paratuberculosis (MAP) is a vigorous microorganism which causes incurable chronic enteritis, Johne’s disease (JD) in cattle. A target of control programmes for JD is to accurately detect MAP-infected cattle early to reduce disease transmission. The present study evaluated the efficacy of two different cultural procedures and a TaqMan real-time PCR assay for detection of subclinical paratuberculosis in dairy herds. Therefore, sixty-one faecal samples were collected from two Dutch dairy herds (n = 40 and n = 21, respectively) which were known to be MAP-ELISA positive. All individual samples were assessed using two different cultural protocols in two different laboratories. The first cultural protocol (first laboratory) included a decontamination step with 0.75% hexadecylpyridinium chloride (HPC) followed by inoculation on Herrold’s egg yolk media (HEYM). The second protocol (second laboratory) comprised of a decontamination step using 4% NaOH and malachite green-oxalic acid followed by inoculation on two media, HEYM and in parallel on modified Löwenstein-Jensen media (mLJ). For the TaqMan real-time PCR assay, all faecal samples were tested in two different laboratories using TaqMan® MAP (Johne’s) reagents (Life Technologies). The cultural procedures revealed positive reactions in 1.64% of the samples for cultivation protocol 1 and 6.56 and 8.20% of the samples for cultivation protocol 2, respectively. The results of the TaqMan real-time PCR performed in two different laboratories yielded 13.11 and 19.76% positive reaction. The kappa test showed proportional agreement 0.54 between the mLJ media (second laboratory) and TaqMan® real-time PCR method (second laboratory). In conclusion, the TaqMan real-time PCR could be a strongly useful and efficient assay for the detection of subclinical paratuberculosis in dairy cattle leading to an improvement in the efficiency of MAP control strategies.  相似文献

Water-level fluctuations (WLF) of lakes have temporal scales ranging from seconds to hundreds of years. Fluctuations in the lake level generated by an unbalanced water budget resulting from meteorological and hydrological processes, such as precipitation, evaporation and inflow and outflow conditions usually have long temporal scales (days to years) and are here classified as long-term WLF. In contrast, WLF generated by hydrodynamic processes, e.g. basin-scale oscillations and travelling surface waves, have periods in the order of seconds to hours and are classified as short-term WLF. The impact of WLF on abiotic and biotic conditions depends on the temporal scale under consideration and is exemplified using data from Lake Issyk-Kul, the Caspian Sea and Lake Constance. Long-term WLF induce a slow shore line displacement of metres to kilometres, but immediate physical stress due to currents associated with long-term WLF is negligible. Large-scale shore line displacements change the habitat availability for organisms adapted to terrestrial and aquatic conditions over long time scales. Short-term WLF, in contrast, do not significantly displace the boundary between the aquatic and the terrestrial habitat, but impose short-term physical stress on organisms living in the littoral zone and on organic and inorganic particles deposited in the top sediment layers. The interaction of WLF acting on different time scales amplifies their overall impact on the ecosystem, because long-term WLF change the habitat exposed to the physical stress resulting from short-term WLF. Specifically, shore morphology and sediment grain size distribution are the result of a continuous interplay between short- and long-term WLF, the former providing the energy for erosion the latter determining the section of the shore exposed to the erosive power.  相似文献

Consequences of internal wave motion for phytoplankton and in particular for the distribution and production of the harmful and buoyant cyanobacterium Planktothrix rubescens were investigated based on data from two field campaigns conducted in Lake Ammer during summer 2009 and 2011. In both years, P. rubescens dominated the phytoplankton community and formed a deep chlorophyll maximum (DCM) in the metalimnion. Internal wave motions caused vertical displacement of P. rubescens of up to 6 m and 10 m, respectively. Vertical displacements of isotherms and of iso-concentration lines of P. rubescens from the same depth range coincided, suggesting that P. rubescens did not or could not regulate its buoyancy to prevent wave-induced vertical displacements. Diatoms dominated the phytoplankton community in the epilimnion and were vertically separated from P. rubescens. The thickness of the diatom layer, but not the diatom concentrations within the layer, changed in phase with the changes in the thickness of the epilimnion caused by internal wave motions. Seiche induced vertical displacements of P. rubescens caused fluctuations in the light intensity available at the depth of the P. rubescens layer. The interplay between seiche induced vertical displacements of the P. rubescens layer and the daily cycle of incident light lead to differences in the daily mean available light intensity between lake ends by up to a factor of ∼3. As a consequence, the daily mean specific oxygen production rate of P. rubescens differed by up to a factor of ∼7 between lake ends. The horizontal differences in the specific oxygen production rate of P. rubescens were persistent over several days suggesting that the associated production of P. rubescens biomass may lead to phytoplankton patchiness. The effect of internal seiches on the spatial heterogeneity and the persistence of horizontal differences in production, however, depend on the timing and the synchronization between internal wave motion and the daily course of incident light intensity. Vertical displacements caused by internal waves could be distinguished from other factors influencing the distribution of P. rubescens (e.g. active buoyancy control, production, vertical mixing) by a temperature-based data transformation. This technique may be of general use for separating wave-induced transport from other processes (e.g. sedimentation, vertical mixing) that affect the distributions of dissolved substances and suspended particles.  相似文献

The limits to optical resolution and the challenge of identifying specific protein populations in transmission electron microscopy have been obstacles in cell biology. Many phenomena cannot be explained by in vitro analysis in simplified systems and need additional structural information in situ, particularly in the range between 1 nm and 0.1 µm, in order to be fully understood. Here, electron spectroscopic imaging, a transmission electron microscopy technique that allows simultaneous mapping of the distribution of proteins and nucleic acids, and an expression tag, miniSOG, are combined to study the structure and organization of DNA double-strand break repair foci.  相似文献
