目的:探索硫氧还蛋白(Trx)抗体柱对Trx融合蛋白纯化的可行性。方法与结果:对含有Trx基因的质粒表达载体pTrxFus进行改造,在Trx读框之后加入6×His序列,并在大肠杆菌中表达C端带有6×His标签的Trx,经Ni2+柱亲和纯化后制备多克隆抗体;把经蛋白A纯化后的抗体偶联在溴化氰活化的琼脂糖凝胶上,制成Trx抗体柱;用此抗体柱纯化与Trx融合表达的豇豆胰蛋白酶抑制剂(CpTI),SDS-PAGE结果显示获得了纯度较高的Trx-CpTI。结论:用Trx抗体制成的免疫亲和层析柱可以有效纯化Trx融合蛋白。 相似文献

Peptides were derived from the large chondroitin sulfate proteoglycan from chick cartilage by clostripain digestion. Using differential chondroitinase ABC and keratanase treatment and direct carbohydrate analysis, three major peptides of 86, 75, and 27 kDa were shown to bear only chondroitin sulfate chains. Another major peptide of 65 kDa was shown to contain both chondroitin sulfate and keratan sulfate chains, allowing it to be separated from the peptides derived from the chondroitin sulfate domain by DEAE-cellulose chromatography. An additional new peptide (100 kDa) containing keratan sulfate chains was found only in clostripain digests of proteoglycan-hyaluronate-link protein aggregates. Unlike any of the other peptides derived from clostripain digestion of proteoglycan monomer or aggregate, this peptide had the properties of a functional hyaluronate binding region. All of these peptides were purified to apparent homogeneity by preparative electroelution from sodium dodecyl sulfate-polyacrylamide gel electrophoresis and deglycosylated with anhydrous hydrogen fluoride. Automated Edman degradation of the two largest chondroitin sulfate peptides revealed that they had unique N termini and several unrecognized residues, which were all subsequently revealed to be modified serine residues following deglycosylation. The keratan sulfate-bearing peptide also had a unique N terminus, which contained a single unrecognized residue, even after HF deglycosylation. Finally, the N terminus of the hyaluronate binding region was blocked. These studies allow estimates of core peptide masses in the absence of carbohydrate as well as provide primary amino acid sequence for O-xylosylated serine residues in the multiply substituted proteoglycans. 相似文献

目的:探讨患者创伤后发生急性肾损伤的患者发病率、临床特点以及发病危险因素,以便有效预防和及早治疗。方法:回顾性分析我院重症监护室2004年1月至2010年12月收治的创伤患者相关临床资料,分析创伤后急性肾损伤的发病率以及发病危险因素。结果:共有106例患者纳入我们的研究,其中47例患者创伤后并发急性肾损伤。在发生急性肾损伤患者中,平均年龄为31±19岁,84.6%为男性;其中25例为脓毒血症引起,18例是因为低血压导致急性肾功能损伤。所有患者中,24例患者出现了少尿的症状,19例患者进行了透析治疗。腹部外伤[(OR)=3.66,P=0.027]和应用呋塞米[(OR=4.10,P=0.026)]是发生急性肾损伤的危险因素。结论:急性肾损伤时创伤后的严重并发症之一,死亡率高。只有找到创伤后发生急性肾损伤的危险因素,才能有效预防和及早治疗。 相似文献

Two new species of Eimeria were observed in the feces of captive white-tailed deer fawns, Odocoileus virginianus, from Alabama. The first new species was easily recognized because of its small size. Sporulated oocysts are spherical, average 10.2 by 10.0 microm, and lack a micropyle and oocyst residuum. Oocysts contain a polar granule and elongate-ellipsoidal sporocysts that measure 6.7 by 3.1 microm. A Stieda body is present on the sporocysts. Oocysts were observed in the feces, and gamonts and oocysts were observed in the jejunum of a month-old fawn from Minnesota that died from enteritis due to this species. Oocysts of this small species were present in 5 of the 6 white-tailed deer fawns examined. Oocysts of a second new species are ellipsoidal and average 29.5 by 24.6 microm. The oocyst encloses an oocyst residuum, polar granule, and elongate-ellipsoidal sporocysts that average 16.0 by 9.0 microm. A Stieda body and substieda body are present on the sporocysts. Oocysts of the second new species were present in 4 of the 6 white-tailed deer fawns examined. Oocysts of E. ivensae are ovoid or flask-like and average 32.0 by 20.8 microm. The oocyst wall is rough, contains a micropyle, and encloses elongate-ellipsoidal sporocysts that average 16.5 by 7.8 microm. A Stieda body is present on the sporocysts. Oocysts of E. ivensae were present in 4 of the 6 white-tailed deer fawns. Oocysts of E. odocoilei are spherical or slightly subspherical and measure 24.7 by 21.5 microm. They enclose ovoid sporocysts that average 12.7 by 8.8 microm. A Stieda and substieda body are present on the sporocyst. The global AIDS pandemic continues to expand and in some regions of the world, such as southern Africa, the prevalence of HIV-1 infection exceeds 20%. The devastating spread of the virus in young women in these countries appears disproportional to overall risk of infection. Regions with high prevalence of HIV-1 are often also highly endemic for other pathogenic viruses including HSV, CMV and HTLV. We propose that acquisition by HIV-1 of the envelope glycoproteins of other viruses, in a process we call “natural pseudotyping,” expands the cellular tropism of HIV-1, enabling it to infect female genital epithelial cells directly and thereby dramatically increasing risk of infection during sexual intercourse. In this proof-of-concept study, we demonstrate that when HIV-1 co-infects T cells along with the gammaretrovirus xenotropic murine leukemia virus-related virus (XMRV), progeny HIV-1 particles are produced capable of infecting primary vaginal, ectocervical and endocervical epithelial cells. These cell types are normally resistant to HIV-1 infection. Infection of primary genital cells was neutralized by antisera against the XMRV glycoprotein, confirming that infection was mediated by the XMRV glycoprotein acquired through pseudotyping of HIV. Inhibition by AZT showed that active replication of HIV-1 occurred in these cells and ruled out non-specific endocytic uptake of the virus. These results demonstrate that natural pseudotyping can expand the tropism of HIV-1 to include genital epithelial cells and have potential implications for sexual transmission of the virus. 相似文献

Analysis of the life cycle of the soil saprophyte Bacillus cereus in liquid soil extract and in soil
Bacillus is commonly isolated from soils, with organisms of Bacillus cereus sensu lato being prevalent. Knowledge of the ecology of B. cereus and other Bacillus species in soil is far from complete. While the older literature favors a model of growth on soil-associated organic matter, the current paradigm is that B. cereus sensu lato germinates and grows in association with animals or plants, resulting in either symbiotic or pathogenic interactions. An in terra approach to study soil-associated bacteria is described, using filter-sterilized soil-extracted soluble organic matter (SESOM) and artificial soil microcosms (ASM) saturated with SESOM. B. cereus ATCC 14579 displayed a life cycle, with the ability to germinate, grow, and subsequently sporulate in both the liquid SESOM extract and in ASM inserted into wells in agar medium. Cells grew in liquid SESOM without separating, forming multicellular structures that coalesced to form clumps and encasing the ensuing spores in an extracellular matrix. Bacillus was able to translocate from the point of inoculation through soil microcosms as shown by the emergence of outgrowths on the surrounding agar surface. Microscopic inspection revealed bundles of parallel chains inside the soil. The motility inhibitor L-ethionine failed to suppress outgrowth, ruling out translocation by a flagellar-mediated mechanism such as swimming or swarming. Bacillus subtilis subsp. subtilis Marburg and four Bacillus isolates taken at random from soils also displayed a life cycle in SESOM and ASM and were all able to translocate through ASM, even in presence of L-ethionine. These data indicate that B. cereus is a saprophytic bacterium that is able to grow in soil and furthermore that it is adapted to translocate by employing a multicellular mode of growth. 相似文献

本文比较研究了短尾猴和日本猴的雄性性行为。短尾猴属单次爬跨射精型种类,每次交配平均持续23.2秒,日本猴属多次爬跨射精型种类,每次交配平均持续8.2分,交配期间雄性平均爬跨雌性lO.6次才能达到射精。短尾猴第1顺位雄性是群中的主要交配者,它占有交配总数的70.9 %;而日本猴第5顺位以下的雄性占交配总数的64%。交配中,两种猴雄性间相互打搅行为的发生频率大致相当。短尾猴高顺位雄性的打搅行为能使低顺位雄性的交配中断,但日本猴高顺位的打搅行为只能使低顺位雄性交配的54.3%中断。 相似文献

目的:探讨夏枯草对人甲状腺癌细胞系SW579细胞生长的抑制作用及其对细胞增殖周期和凋亡的影响。方法:采用甲基噻唑(MTT)比色法和生长曲线测定不同浓度夏枯草在不同作用时间内对在体外培养SW579细胞增殖的影响,同时应用流式细胞术检测细胞增殖周期及凋亡率的变化。结果:夏枯草可在G0/G1期阻滞人甲状腺癌细胞系SW579的增殖,使S期细胞比率降低;在一定范围内,夏枯草的浓度越高、作用时间越长,对肿瘤细胞生长的抑制作用越强,凋亡率也越高。结论:夏枯草能抑制人甲状腺癌细胞系SW579细胞生长,并诱导细胞凋亡而阻止细胞周期。 相似文献
