minced beef were characterized in detail.Debaryomyces hansenii was the most commonly isolated yeast from most samples followed byCandida zeylanoides andPichia membranaefaciens. The presence of sulphite in sausages did not appear to affect the numbers and range of yeasts present but did affect their
relative proportions. A survey of one factory showed that meat intended for sausage production and equipment harboured the

Western blotting is a significant tool employed for the detection of cell proteins. High-molecular-weight proteins have proven a challenge to detect by western blotting, but proteins even of 100 KDa can still present difficulties in detection. This work reports the development of a heat transfer method that is suitable for both low- and high-molecular-weight proteins. The procedure involves the use of a constant temperature at 78 °C in a dedicated heat transfer module. Through the use of this protocol the neuronal adaptor protein X11α (120 KDa), which prior to this methodology was undetectable endogenously in the neuroblastoma cell line (N2a), was successfully detected in the N2a cell line. The procedure provides a reproducible protocol that can be adapted for other high-molecular-weight proteins, and it provides the advantage that low-molecular-weight proteins are not sacrificed by the methodology. 相似文献

Animal personality is defined as individual variation in behaviour that is consistent over time and/or across contexts. Animal personality is now considered a fundamental aspect in the fields of animal behaviour and behavioural ecology, yet the majority of studies assess repeatability of behaviours over only relatively short time periods (e.g. a week) using just two measures. Understanding whether behavioural traits are repeatable over longer periods is important for the assessment of individual differences in behaviour. Using zebrafish (Danio rerio), we investigated the repeatability of activity and exploratory behaviours, including distance travelled, time spent in the bottom of the arena, stationary time and overall exploration of the novel arena over a 28-week period, using five intervals. All measures were repeatable over 28 weeks, but the repeatability estimates were much lower when comparing the initial week one and week two behaviours. There were clear sex differences in aspects of activity, with males more active than females. Importantly, our behavioural assays suggest that zebrafish require an initial “tank experience,” prior to the main phenotyping session, to ensure that behaviours being measured are repeatable—these effects are often not considered, but have implications for the many studies that measure behaviour at a single time point only. 相似文献

The cetacean tail fluke blades are not supported by any vertebral elements. Instead, the majority of the blades are composed of a densely packed collagenous fiber matrix known as the core layer. Fluke blades from six species of odontocete cetaceans were examined to compare the morphology and orientation of fibers at different locations along the spanwise and chordwise fluke blade axes. The general fiber morphology was consistent with a three‐dimensional structure comprised of two‐dimensional sheets of fibers aligned tightly in a laminated configuration along the spanwise axis. The laminated configuration of the fluke blades helps to maintain spanwise rigidity while allowing partial flexibility during swimming. When viewing the chordwise sectional face at the leading edge and mid‐chord regions, fibers displayed a crossing pattern. This configuration relates to bending and structural support of the fluke blade. The trailing edge core was found to have parallel fibers arranged more dorso‐ventrally. The fiber morphology of the fluke blades was dorso‐ventrally symmetrical and similar in all species except the pygmy sperm whale (Kogia breviceps), which was found to have additional core layer fiber bundles running along the span of the fluke blade. These additional fibers may increase stiffness of the structure by resisting tension along their long spanwise axis. 相似文献

Parental behavior and infant development of black and white ruffed lemurs (Varecia variegata) were studied on Nosy Mangabe Island in northeast Madagascar. Ruffed lemur females produced twins, prepared nests for neonates in the trees, transported infants by mouth, and parked them in the trees. During two consecutive birth seasons, the average birth rate for nine females was 0.58. Two females reproduced in one social group. Lactating females spent most of their time resting with their infants or foraging for food. Infants developed rapidly and were fully mobile by 3–4 months. No female reproduced successfully in 2 consecutive years. In 1988, infant mortality within 3 months of birth was very high (64%). Accidental falls may have been one major cause. Adults of both sexes, including a reproductive female, exhibited alloparental behavior such as guarding infants and nonmaternal nursing. Alloparental care may increase the likelihood of infant survival. Some of these observations are not compatible with the idea that ruffed lemurs live in small pair-bonded groups, as other researchers have suggested. 相似文献
