Sequence analysis of a 237 kb genomic fragment from the central region of the MHC has revealed that the HLA-B and HLA-C genes are contained within duplicated segments peri-B (53 kb) and peri-C (48 kb), respectively, and separated by an intervening sequence (IF) of 30 kb. The peri-B and peri-C segments share at least 90% sequence homology except when interrupted by insertions/deletions including Alu, L1, an endogenous retrovirus, and pseudogenes. The sequences of peri-B, IF, and peri-C were searched for the presence of Alu elements to use as markers of evolution, chromosomal rearrangements, and polymorphism. Of 29 Alu elements, 14 were identified in peri-B, 11 in peri-C, and 4 in IF. The Alu elements in peri-B and peri-C clustered phylogenetically into two clades which were classified as ``preduplication' and ``postduplication' clades. Four Alu J elements that are shared by peri-B and peri-C and are flanked by homologous sequences in their paralogous locations, respectively, clustered into a ``preduplication' clade. By contrast, the majority of Alu elements, which are unique to either peri-B or peri-C, clustered into a postduplication clade together with the Alu consensus subfamily members ranging from platyrrhine-specific (Spqxcg) to catarrhine-specific Alu sequences (Y). The insertion of platyrrhine-specific Alu elements in postduplication locations of peri-B and peri-C implies that these two segments are the products of a duplication which occurred in primates prior to the divergence of the New World primate from the human lineage (35–44 mya). Examination of the paralogous Alu integration sites revealed that 9 of 14 postduplication Alu sequences have produced microsatellites of different length and sequence within the Alu 3′-poly A tail. The present analysis supports the hypothesis that HLA-B and HLA-C genes are products of an extended segmental duplication between 44 and 81 million years ago (mya), and that subsequent diversification of both genomic segments occurred because of the mobility and mutation of retroelements such as Alu repeats. Dendrocacalia crepidifolia Nakai (Compositae, Senecioneae), the only species of this arboreal genus endemic to Haha Island in the Bonin Islands, was found to be dioecious. Male flowers differ from female ones in having a stunted style (style in female exserted from corolla and deeply bifurcating) and anthers filled with fertile pollen (anthers in the female lacking pollen). The size of the corolla and number of florets per head were similar between male and female flowers. The crown area of this arboreal species was also similar in male and female plants. The sex ratio was 0.55 male, not significantly different from 0.5. Both sexes produced nectar of similar sugar concentration (ca. 50%). The flowers are pollinated by feral honeybees (Apis mellifera), but they are thought to have been pollinated by small, lesshairy, endemic solitary bees before honeybees were introduced and subsequently became the dominant bee species on the island. The evolution of dioecy ofDendrocacalia on the island is thought to stem from the deleterious effects of inbreeding that are inherent in plants with geitonogamy. The increased geitonogamy on the island has resulted from increased woodiness (i.e., increased number of flowers per plant) and the original dependence on endemic bee pollinators, which are now endangered.  相似文献

For identification of microsomal cytochrome P-450 (P-450) enzymes which catalyze 2- or 4-hydroxylations of estrogens in the rat liver, estradiol (E2) and estradiol 17-sulfate (E2-17-S) were selected as the substrates and incubated with various kinds of purified P-450 enzymes: PB-1, PB-2, PB-4 and PB-5 obtained from phenobarbital-treated male rats (Sprague-Dawley); MC-1 and MC-5 from 3-methylcholanthrene-treated male rats; and UT-1, UT-2, UT-4 and UT-5 from untreated animals. The reactions were carried out under the P-450-reconstructed system, and the resulting products were determined by HPLC using electrochemical detection. All the enzymes tested were shown to have varying degrees of catalytic activities for 2-hydroxylation of the two substrates; UT-1 and UT-2 had the highest activity. Of the induced P-450 enzymes, PB-2 and MC-1 showed fairly high catalytic activity for 4-hydroxylation of E2. The P-450 enzymes obtained from the untreated male rats, especially UT-4, showed the highest catalytic activity for 4-hydroxylation of the two substrates. From these results and also from kinetic experiments, the P-450 enzymes which catalyze 2- and 4-hydroxylations of estrogen were considered to be different species. A part of E2 was converted to such metabolites as estrone and those having a hydroxyl group at positions 6β, 15 or 16, each production of which was estimated to be catalyzed by single or multiple P-450s.  相似文献

Taking advantage of five mouse genomic or cDNA probes [KE5(probe 14), KE4 (probe11), KE3 (probe7), KE2 (probe5), and SET] mapped on the H-2K region in mouse, we have identified and localized homologues of these five genes in the human major histocompatibility complex region (HKE5, HKE4, HKE3, HKE2, and HSET, respectively). Cosmid cloning and pulsed field gel electrophoresis analyses indicated that a human homologous gene, HKE5, is located 10 kilobases (kb) centromeric of the 2(XI) collagen (COL11A2) gene followed by HKE4. HKE3, closely linked to HKE2, is located 170 kb centromeric of HKE4. Furthermore, HSET is located 50 kb centromeric of HKE2. This gene organization outside the DP subregion is completely identical to that of the mouse H-2K region centromeric of I-Pb 3, a mouse homologue of the DPB gene, except the lack of genes corresponding to the H-2K and -K2 genes in human.  相似文献

When polyethyleneimine (PEI), a water-soluble cationic polymer, was added to solutions of glucose dehydrogenase (GDH) from Bacillus megaterium IWG3 at a molar ratio of PEI to GDH greater than 10, the thermal stability of GDH markedly increased. By addition of PEI, the rate of GDH-catalysed oxidation of -d-glucose increased in a low concentration range of NAD+ and NADP+ and the Michaelis constants and inhibition constants for both NAD+ and NADP+ decreased. These results suggested that negatively charged GDH interacts with cationic water-soluble polymers to form conjugates by electrostatic attraction, and also that negatively charged coenzymes are adsorbed by the polymers, resulting in enrichment of the coenzymes in the vicinity of GDH. Being sessile organisms, plants show a high degree of developmental plasticity to cope with a constantly changing environment. While plasticity in plants is largely controlled genetically, recent studies have demonstrated the importance of epigenetic mechanisms, especially DNA methylation, for gene regulation and phenotypic plasticity in response to internal and external stimuli. Induced epigenetic changes can be a source of phenotypic variations in natural plant populations that can be inherited by progeny for multiple generations. Whether epigenetic phenotypic changes are advantageous in a given environment, and whether they are subject to natural selection is of great interest, and their roles in adaptation and evolution are an area of active research in plant ecology. This review is focused on the role of heritable epigenetic variation induced by environmental changes, and its potential influence on adaptation and evolution in plants.  相似文献
