We have investigated the effects of endogenous angiotensin II (ANG II) on hepatic angiotensinogen mRNA levels in rats. Changes in endogenous ANG II were induced by various sodium intakes (standard-, low-, and high-sodium) or by enalapril treatment. In a low sodium state for 2 weeks, angiotensinogen mRNA levels and plasma ANG II concentration increased 1.3-fold and 1.6-fold compared to those in standard sodium state, respectively. In a high sodium state, angiotensinogen mRNA levels and plasma ANG II concentration decreased by 42% and 56% compared to the standard sodierm state, respectively. Four hours after treatment with enalapril (3 mg/kg), angiotensinogen mRNA level and plasma ANG II concentration decreased by 25% and 12% compared to the standard sodium state, respectively. There was a close correlation between angiotensinogen mRNA level and plasma ANG II concentration (r = 0.79, P less than 0.01). These results suggest that endogenous ANG II may play an important role in the regulation of hepatic angiotensinogen synthesis.  相似文献

The haloacetate dehalogenase gene,dehH2, cloned fromMoraxella sp. strain B could be used as a selection marker gene for vectors inEscherichia coli andPseudomonas putida. Haloacetates, especially iodoacetate, inhibit the growth of some microorganisms. ThedehH2 gene introduced into the cells conferred iodoacetate resistance on them. An Epstein-Barr virus (EBV)-infected fibroblast line, designated DSEK, was spontaneously established from synovial tissue of a patient with rheumatoid arthritis (RA). DSEK cells expressed EBV nuclear antigens EBNA-1 and EBNA-2 and latent membrane protein LMP-1. Cell surface markers of DSEK cells were similar to those of EBV-negative fibroblast clones derived from synoviocytes and were negative for lymphocyte and macrophage markers. DSEK cells expressed CD44, CD58, and HLA-DR antigens and spontaneously produced interleukin-10 basic fibroblast growth factor and transforming growth factor beta1. These results indicate that rheumatoid synoviocytes can be a target for EBV infection and suggest that EBV may play a role in the pathogenesis of RA.  相似文献

We have developed a method for the analysis of 5,6-dihydroxyeicosatrienoate-1,5-lactone (5,6-DiHETriE-δ-lactone) in tissue homogenates, supplemented with NADPH and arachidonic acid [20:4(n−6)] as a substrate. During the incubation and the extraction, most of the 5,6-epoxyeicosatrienoic acid (5,6-EpETriE) was converted to 5,6-dihydroxyeicosatrienoic acid (5,6-DiHETriE), and most of the 5,6-DiHETriE was converted to 5,6-DiHETriE-δ-lactone. Consequently, the chief degradation product of 5,6-EpETriE and 5,6-DiHETriE in the incubation mixture was 5,6-DiHETriE-δ-lactone. 5,6-DiHETriE-δ-lactone, corresponding to [20:4(n−6)], was shown to be characterized by a high intensity of quasimolecular ions (MH+ and MNH4+), using ion analysis obtained by reversed-phase HPLC-thermospray MS. On selected-ion monitoring (SIM) chromatograms of 5,6-DiHETriE-δ-lactone and with deuterium-labeled 15(s)-hydroxyeicosatetraenoic acid as the internal standard, the regression equation of the peak-area ratio and the amount of 5,6-DiHETriE-δ-lactone was y = 12.2x + 0.7 (r = 0.9996). 5,6-Epoxygenase activity was represented as the sum of the amount of 5,6-DiHETriE-δ-lactone, 5,6-EpETriE and 5,6-DiHETriE per mg protein, after 30 min in an incubation mixture. The activity from rat brain homogenate decreased considerably with growth of the rat.  相似文献

A hypothetical biogeographical history of northeastern Asiatic soricine shrews in the late Quaternary was developed by integrating their present distributions, fossil records, a hypothetical phylogeny, and geological investigations. First, a biological area cladogram of the northeastern Asiatic region was constructed by applying the vicariance hypothesis to the phylogeny of thecaecutiens/shinto group, a monophyletic group proposed by Ohdachi et al. (1997). Comparing the biological area cladogram with a geological hypothesis by Ohshima (1990,1991,1992), we hypothesized a geographical history of northeastern Asia. Species were then located on the dendrogram of the geographical history, referring to the present distributions, fossil records, and phylogeny of shrews. According to our hypothesis, higher species diversity of the northern region of northeastern Asia (Hokkaido, Sakhalin, and Eastern Siberia) was achieved by several series of colonizations and habitat expansion. On the other hand, the shrew communities of the southern region (Honshu, Sado, Shikoku, and Kyushu) were created by extinction and isolation followed by speciation.  相似文献

Using an isogenic line of rice having lazy gene (la), we studied the correlation between the agravitropic response at the young seedling stage and the lazy habit (prostrate growth of tillers) at the more advanced stage of growth. In this study, it was found that both agravitropism and lazy habit were controlled by the single recessivela gene. That is, F2 segregants of Kamenoo×lazy-Kamenoo, which had an agravitropic response at their young seedling stage, showed a lazy habit of growth in the more advanced stage of vegetative growth. On the other hand, seedlings that showed normal gravitropic curvature at their early stage of growth had an upright growth in the mature stage.  相似文献
