When totally etiolated pea epicotyls were cut into segments and incubated with potassium phosphate buffer, pH 6.0, in the
dark at 25 C, an instantaneous loss of photoreversible absorbance change, Δ (ΔA) between 660 and 730 nm, was observed after
the first irradiation with actinic red light in the spectrophotometric measurement of phytochromein vivo. The shorter the epicotyl segments, and the longer the period of dark incubation, the greater was the loss detected in the
measurement.
A remarkable decline of Δ(ΔA) in the far-red region was seen inin vivo difference spectra for phytochrome, after the epicotyl segments were incubated in the dark at 25 C. As the period of dark
incubation was prolonged, the ratio of the maximal change of Δ(ΔA) in the far-red region to that in the red region was reduced.
It decreased to ca. one third of the initial value after incubation for 8 hr.
The evidence indicates that Pfr killer activity and P* denaturation, both of which have so far been known onlyin vitro, can also occur in segments of etiolated pea epicotyls. 相似文献

Persimmon waste was chemically modified with dimethylamine (DMA) to obtain a tertiary-amine-type gel, named DMA persimmon waste gel (DMA-PW). It was found to be effective for the adsorption of Au(III), Pd(II), and Pt(IV) in hydrochloric acid medium. In contrast, base metals such as Cu(II), Zn(II), Fe(III), and Ni(II) were not practically adsorbed. The formation of ion pairs of the metal chloro complex anions with the protonated adsorption gels was proposed as the main adsorption process. The gel exhibited selectivity only for precious metals with a remarkably high capacity for Au(III), i.e., 5.63 mol/kg dry gel and comparable capacities, i.e., 0.42 and 0.28 mol/kg for Pd(II) and Pt(IV), respectively. According to the kinetic and electrochemical studies, the adsorption rate of Au(III) was greatly enhanced by the chemical modification. Also, its excellent adsorption characteristics for the precious metals were confirmed by adsorption and elution tests using a column packed with the DMA-PW gel. 相似文献

When inappropriate DNA structures arise, they are sensed by DNA structure-dependent checkpoint pathways and subsequently repaired. Recruitment of checkpoint proteins to such structures precedes recruitment of proteins involved in DNA metabolism. Thus, checkpoints can regulate DNA metabolism. We show that fission yeast Rad9, a 9-1-1 heterotrimeric checkpoint-clamp component, is phosphorylated by Hsk1(Cdc7), the Schizosaccharomyces pombe?Dbf4-dependent kinase (DDK) homolog, in response to replication-induced DNA damage. Phosphorylation of Rad9 disrupts its interaction with replication protein A (RPA) and is dependent on 9-1-1 chromatin loading, the Rad9-associated protein Rad4/Cut5(TopBP1), and prior phosphorylation by Rad3(ATR). rad9 mutants defective in DDK phosphorylation show wild-type checkpoint responses but abnormal DNA repair protein foci and decreased viability after replication stress. We propose that Rad9 phosphorylation by DDK releases Rad9 from DNA damage sites to facilitate DNA repair. 相似文献

The Mu phage virion contains tail-spike proteins beneath the baseplate, which it uses to adsorb to the outer membrane of Escherichia coli during the infection process. The tail spikes are composed of gene product 45 (gp45), which contains 197 amino acid residues. In this study, we purified and characterized both the full-length and the C-terminal domains of recombinant gp45 to identify the functional and structural domains. Limited proteolysis resulted in a Ser64-Gln197 sequence, which was composed of a stable C-terminal domain. Analytical ultracentrifugation of the recombinant C-terminal domain (gp45-C) indicated that the molecular weight of gp45-C was about 58 kDa and formed a trimeric protomer in solution. Coprecipitation experiments and a quartz crystal microbalance (QCM) demonstrated that gp45-C irreversibly binds to the E. coli membrane. These results indicate that gp45 shows behaviors similar to tail-spike proteins of other phages; however, gp45 did not show significant sequence homology with the other phage tail-spike structures that have been identified. 相似文献

The origin of transfer (oriT) of a bacterial plasmid plays a key role in both the initiation and termination of conjugative DNA transfer. We have previously shown that a conjugation-dependent recombination between the tandem R64 oriT sequences cloned into pHSG398 occurred, resulting in the deletion of the intervening sequence during DNA transfer. In this study, we tandemly cloned two oriT sequences of IncI1 plasmid R64 into pUC18. Specific recombination between the two oriT sequences in pUC18 was observed within Escherichia coli cells harboring mini-R64. This recombination was found to be independent of both the recA gene and conjugative DNA transfer. The R64 genes nikA and nikB, required for conjugal DNA processing, were essential for this recombination. Although a fully active 92-bp oriT sequence was required at one site for the recombination, the 44-bp oriT core sequence was sufficient at the other site. Furthermore, when two oriT sequences were tandemly cloned into the single-stranded phage vector M13 and propagated within E. coli cells, recombination between the two oriT sequences was observed, depending on the nikB gene. These results suggest that the R64 relaxase protein NikB can execute cleavage and rejoining of single-stranded oriT DNA within E. coli cells, whereas such a reaction in double-stranded oriT DNA requires collaboration of the two relaxosome proteins, NikA and NikB. 相似文献

Recent studies on global climate change report that increase in seawater temperature leads to coastal ecosystem change, including
coral bleaching in the tropic. In order to assess the effect of increased seawater temperature on a temperate coastal ecosystem,
we studied the inter-annual variation in productivity of Laminaria japonica using long-term oceanographic observations for the Uwa Sea, southern Japan. The annual productivity estimates for L. japonica were 2.7 ± 2.5 (mean ± SD) kg wet wt. m−1 (length of rope) (2003/2004), 1.0 ± 0.6 kg wet wt. m−1 (2004/2005) and 12.1 ± 12.5 kg wet wt. m−1 (2005/2006). Our previous study using the same methodology at the same locality reported that the productivity was estimated
for the 2001/2002 (33.3 ± 15.2 kg wet wt. m−1) and 2002/2003 (34.0 ± 8.7 kg wet wt. m−1) seasons. Productivity in 2003/2004 and 2004/2005 was significantly lower than in years 2001/2002, 2002/2003 and 2005/2006.
A comparison of oceanographic conditions among the 5 years revealed the presence of threshold seawater temperature effects.
When the average seawater temperature during the first 45 days of each experiment exceeded 15.5°C, productivity was reduced
to about 10 % of that in cooler years. Moreover the analysis of growth and erosion rates indicates that when the seawater
temperature was over 17.5°C, erosion rate exceeded growth rate. Thus, an increase of seawater temperature of just 1°C during

Recently, statins have been being studied for their proapoptic and antimetastatic effects. However, the exact mechanisms of their anticancer action are still unclear. Dolichyl phosphate is a nonsterol isoprenoid derivative in the mevalonate pathway that affects the expression of the Insulin-like growth factor 1 receptor (IGF-1R). IGF-1R activation is required for prostate cell proliferation; therefore, IGF-1R inhibitory agents may be of preventive and/or therapeutic value. In this study, the effects of simvastatin on IGF-1R signaling in prostate cancer PC-3 cells were examined. Simvastatin suppressed proliferation and induced apoptosis of PC-3, and the expression of IGF-1R was suppressed by simvastatin. Knockdown of IGF-1R by siRNA led to inhibition of proliferation of PC-3. Simvastatin also inhibited IGF-1-induced activation of both ERK and Akt signaling and IGF-1-induced PC-3 cell proliferation. Our results suggest statins are potent inhibitors of the IGF-1/IGF-1R system in prostate cancer cells and may be beneficial in prostate cancer treatment. 相似文献

Three O-methyltransferases which catalyze S-adenosyl-L-methionine (SAM)-dependent O-methylation of licodione (LMT), flavone/flavonol (FMT), and caffeic acid (CMT) were separated from the callus culture of Glycyrrhiza echinata, and characteristic differences between their pH optima and Mg2+ requirement for activity were demonstrated. The activity of LMT, which is involved in retrochalcone (echinatin) biosynthesis, but not of FMT or CMT, was found to be stimulated when suspension-cultured G. echinata cells were treated with yeast extract (YE), which causes rapid production of echinatin in the cells. Cycloheximide suppressed both the YE-induced echinatin formation and LMT enhancement. The results indicate a selective induction of retrochalcone pathway in Glycyrrhiza cells in response to stress.Abbreviations SAM

Members of the family Vibrionaceae are generally found in marine and brackish environments, playing important roles in nutrient cycling. The Rumoiensis clade is an unconventional group in the genus Vibrio, currently comprising six species from different origins including two species isolated from non-marine environments. In this study, we performed comparative genome analysis of all six species in the clade using their complete genome sequences. We found that two non-marine species, Vibrio casei and Vibrio gangliei, lacked the genes responsible for algal polysaccharide degradation, while a number of glycoside hydrolase genes were enriched in these two species. Expansion of insertion sequences was observed in V. casei and Vibrio rumoiensis, which suggests ongoing genomic changes associated with niche adaptations. The genes responsible for the metabolism of glucosylglycerate, a compound known to play a role as compatible solutes under nitrogen limitation, were conserved across the clade. These characteristics, along with genes encoding species-specific functions, may reflect the habit expansion which has led to the current distribution of Rumoiensis clade species. Genome analysis of all species in a single clade give us valuable insights into the genomic background of the Rumoiensis clade species and emphasize the genomic diversity and versatility of Vibrionaceae. 相似文献
