We have demonstrated that ischemic neuronal death (apoptosis) of rat CA1 region of the hippocampus was prevented by infusing pituitary adenylate cyclase-activating polypeptide (PACAP) either intracerebroventricularly or intravenously. We have also demonstrated that the activity of mitogen-activated protein (MAP) kinase family members, including ERK (extracellular signal-regulated kinase), Jun N-terminal kinase (JNK)/stress-activated protein kinase (SAPK) and p38, was increased in the hippocampus within 1-6 h after brain ischemia. The molecular mechanisms underlying the PACAP anti-apoptotic effect were demonstrated in this study. Ischemic stress had a strong influence on MAP kinase family, especially on JNK/SAPK and p38. PACAP inhibited the activation of JNK/SAPK and p38 after ischemic stress, while ERK is not suppressed. These findings suggest that PACAP inhibits the JNK/SAPK and p38 signaling pathways, thereby protecting neurons against apoptosis.  相似文献

The blood–brain barrier (BBB) controls the exchange of peptides and regulatory proteins between the central nervous system (CNS) and the blood. Transport across the BBB of such regulatory substances is altered in animal models of Alzheimer’s disease. These alterations could lead to cognitive impairments or diminish their therapeutic potential. Here, we measured the transport rate of radioactively labeled pituitary adenylate cyclase-activating polypeptide (PACAP) from blood into whole brain and into 11 brain regions in three groups of mice: young (2 months old) ICR, young (2 months old) SAMP8, and aged (12 months old) SAMP8 mice. The SAMP8 is a strain which develops impaired learning and memory with aging that correlates with an age-related increase in brain levels of amyloid β protein (AβP). PACAP is a powerful neurotrophin that may have a therapeutic role in neurodegenerative diseases. We found that I-PACAP crossed the BBB fastest at the hypothalamus and the hippocampus in all three groups. Slower transport rates into the whole brain, the olfactory bulb, the hypothalamus, and the hippocampus for aged SAMP8 mice was likely related to differences both from strain and expression of AβP with aging.  相似文献

A deoxycytidine deaminase that was extremely thermostable in the presence of dithiothreitol was found in a mesophilic bacterium isolated from soil. The bacterium was classified as a Nocardioides sp. The enzyme was purified to a homogeneous protein by treatment at 100 degrees C, ammonium sulfate precipitation, and chromatography on DEAE-Toyopearl, hydroxyapatite, and then Sephacryl S-100. Twenty micrograms of the pure enzyme was obtained from 811 mg of the starting crude protein. After treatment at 50 degrees C for 15 min in the absence of dithiothreitol, enzyme activity was 44% of the starting activity; after treatment at 100 degrees C for 2 h in the presence of 50 mm dithiothreitol, activity was 56% of the starting activity. Dithiothreitol greatly stabilized the enzyme. Activity was maximum at 99 degrees C. The Km values for deoxycytidine, cytidine, and methyl-deoxycytidine were 55.2, 140, and 130 microM, respectively. The molecular mass was estimated to be 52 kDa by gel permeation chromatography. The enzyme molecule was dissociated into two subunits each of 18 kDa subunit when reduced with mercaptoethanol.  相似文献

Collagen XIX is an extremely rare extracellular matrix component that localizes to basement membrane zones and is transiently expressed by differentiating muscle cells. Characterization of mice harboring null and structural mutations of the collagen XIX (Col19a1) gene has revealed the critical contribution of this matrix protein to muscle physiology and differentiation. The phenotype includes smooth muscle motor dysfunction and hypertensive sphincter resulting from impaired swallowing-induced, nitric oxide-dependent relaxation of the sphincteric muscle. Muscle dysfunction was correlated with a disorganized matrix and a normal complement of enteric neurons and interstitial cells of Cajal. Mice without collagen XIX exhibit an additional defect, namely impaired smooth-to-skeletal muscle cell conversion in the abdominal segment of the esophagus. This developmental abnormality was accounted for by failed activation of myogenic regulatory factors that normally drive esophageal muscle transdifferentiation. Therefore, these findings identify collagen XIX as the first structural determinant of sphincteric muscle function, and as the first extrinsic factor of skeletal myogenesis in the murine esophagus.  相似文献

A 48-h microstructure experiment captured the variation of turbulence in Lake Biwa, Japan, during a strong stratification period, 1 week after a typhoon event. A free-fall microstructure profiler (TurboMAP) and a fine-scale profiler (F-probe) were deployed. An array of five ADCPs positioned close to the experiment site provided current measurements associated with the internal-wave field for a period of one week. Strong winds related to the passage of a typhoon close to the lake generated two low-frequency internal waves: a Kelvin wave and an inertial wave. Both waves were in the first baroclinic mode. This study pictures the superposition of two large-scale internal waves and a stronger current resulting from the two waves being in phase. The synchronization of the waves provoked enhanced shear in the hypolimnion, more than 10 m above the bottom, as well as high dissipation (10?7 W kg?1) and vertical diffusivity reaching 10?4 m2 s?1. The enhanced shear event seems to be related to the current amplitude since it occurred when the current amplitude was increased by the diel wind. Assuming the same turbulence intensity for the enhanced hypolimnetic shear events, this superposition may affect nutrient transfer in the hypolimnion. Also, we witnessed sediment resuspension, consisting of high turbidity and high fluorescence intensity, likely due to a combination of bottom stress and enhanced turbulence.  相似文献

The coupling reaction of glucoamylase and halogenacetyl cellulose (HAC) without pretreating with organic solvent led to form large particles of immobilized glucoamylase and the activity and the specific activity of the preparation were very low. However, the coupling reaction with HAC pretreated with organic solvents allowed to form very fine particles and the activity was increased by five times. The latter contained 3~6% of enzyme protein and the specific activity to maltose reached to 80~90% of native glucoamylase. Since the specific activity of the preparation was presumed to be much influenced by the particle size, the specific activity and general properties of different particle sizes were compared with those of native enzyme. The specific activities of particles of 0~15μ, 15~55μ, 70~190μ and 130~270μ showed 82%, 33%, 27% and only 7% of native enzyme, respectively. Km values of native form, 0~15μ, 15~55μ and 70~190μ particles were 0.90×10?3m, 1.35×10?3m, 1.60×10?3m and 2.1×10?3m in the case of maltose as substrate, respectively. The other properties of particles of 0~15μ were almost identical to those of native enzyme except for the effect of temperature on the reaction rate. However, pH activity, pH stability and urea stability of particles of 70~190μ were much inferior to those of native enzyme and particles of 0~15μ.  相似文献

A cell suspension culture of a Vitis hybrid converted quercetin to six glucosides. Their structures were identified as quercetin 3-O-β-d-glucopyranoside, quercetin 3,4′-di-O-β-d-glucopyranoside, quercetin 3,7-di-O-β-d-glucopyranoside, isorhamnetin 3-O-β-d-glucopyranoside, isorhamnetin 3,4′-di-O-β-d-glucopyranoside, and isorhamnetin 3,7-di-O-β-d-glucopyranoside by UV, FD-MS, 1H-NMR, 13C-NMR spectroscopy and TLC analysis.

The course of conversion was also investigated and it was shown that quercetin 3-O-glucoside reached the maximum yield of 31% in 24 hr and then gradually disappeared accompanied by the production of quercetin 3,4′- and 3,7-di-O-glucosides. Although the same rise and fall relationship was observed between isorhamnetin 3-O-glucoside and isorhamnetin 3,4′- or 3,7-di-O-glucoside, their conversion ratios were much lower than those of quercetin glucosides.  相似文献

The murine preosteoblastic cell line, MC3T3-E1, is widely used to study bone formation and differentiation in vitro. However, this cell line is unstable in culture. The current study was designed to establish a stable osteoblastic cell line. A mammalian expression vector carrying the SV 40 large T antigen was introduced into a primary culture of cells isolated from the calvaria of newborn mice. Among isolated cell lines, the MN16 cell line was selected for further characterization. The MN16 cell line was cultured for 28 days, and compared with the MC3T3-E1 cell line with or without induction. The expression of bone-related genes was examined using the real-time RT-PCR technique. Alizarin red and von Kossa staining were used to detect mineralization of nodules in the cultures. The cell line showed the characteristics of osteoblastic cells in term of gene expression patterns of various molecular markers and calcium deposition in the cell layer after induction. Furthermore, the MN16 cells showed strong adhesion to the basic domain of collagen, a result that is specific for bone-derived cells. The MN16 cell line was found to be stably differentiated into bone formation cells in vitro and should be useful for studying bone biology.  相似文献

Cannabinoid receptor type 1 (CB1) is mainly expressed in the brain, as well as being expressed in functional relevant concentrations in various peripheral tissues. 1-(4-Chlorophenyl)-3-(3-(6-(pyrrolidin-1-yl)pyridin-2-yl)phenyl)urea (PSNCBAM-1, 1) was developed as a potent allosteric antagonist for CB1 and its oral administration led to reductions in the appetite and body weight of rats. Several analogs of 1 (compounds 2 and 3) were recently identified through a series of structure-activity relationship studies. Herein, we report the synthesis of radiolabeled analogs of these compounds using [11C]COCl2 and an evaluation of their potential as PET ligands for CB1 imaging using in vitro and in vivo techniques. [11C]2 and [11C]3 were successfully synthesized in two steps using [11C]COCl2. The radiochemical yields of [11C]2 and [11C]3 were 17 ± 8% and 20 ± 9% (decay-corrected to the end of bombardment, based on [11C]CO2). The specific activities of [11C]2 and [11C]3 were 42 ± 36 and 37 ± 13 GBq/μmol, respectively. The results of an in vitro binding assay using brown adipose tissue (BAT) homogenate showed that the binding affinity of 2 for CB1 (KD = 15.3 µM) was much higher than that of 3 (KD = 26.0 µM). PET studies with [11C]2 showed a high uptake of radioactivity in BAT, which decreased in animals pretreated with AM281 (a selective antagonist for CB1). In conclusion, [11C]2 may be a useful PET ligand for imaging peripheral CB1 in BAT.  相似文献
