Immobilized invertase was prepared by ionically binding the enzyme to diethylaminoacetyl cellulose (DEAA-cellulose). DEAA-cellulose-invertase complex was quite stable to electrolyte in the range of pH 5–7. Bound invertase was less active than the native enzyme, and approximately 55–70% of the enzyme activity was lost on binding. The complex was stable for 9 days' continuous inversion in a column system at 30°C, but was rather unstable at 40°C. Heat stability and the effect of temperature on the reaction rate of the complex were almost identical with those of the native enzyme.  相似文献

Immobilized β-galactosidase gel was prepared using poly(vinylpyrrolidone) (PVP) under β-ray irradiation. In contrast to the gelation of N-vinylpyrrolidone monomer–enzyme solution, the gelation of PVP-β-galactosidase solution (PVP content: 10%) was almost completely uneffected by the dose rate and amount of phosphate present. PVP-enzyme solution was gelled by irradiation with 3.0 Mrad. The expressed activity of the PVP-enzyme gel was about 30% of the initial activity and added activity was almost totally entrapped. No leakage of enzyme from these gels could be detected. Leakage was, however, detected in the case of the gelation of PVP-enzyme solution containing more than 1% of enzyme protein. When the general properties of the gel were compared with those of the native enzyme, the gel proved to be slightly inferior to the native enzyme with respect to optimum temperature, heat stability, pH activity, and pH stability. Continuous hydrolysis of lactose in acid whey could be carried out at 50°C using a column packed with the gel and sawdust and the degree of hydrolysis was found to be almost, constant for 12 days. The merits of using PVP in the immobilization of enzymes include the simplicity of the procedure and the fact that the PVP-enzyme gel can be used in the food industry without anxiety because of its high degree of compatibility with living organisms.  相似文献

We report that dermatopontin (DP), an abundant dermal extracellular matrix protein, is found in the fibrin clot and in the wound fluid, which comprise the provisional matrix at the initial stage of wound healing. DP was also found in the serum but at a lower concentration than that in wound fluid. DP co-localized with both fibrin and fibronectin on fibrin fibers and interacted with both proteins. Both normal fibroblast and HT1080 cell adhesion to the fibrin-fibronectin matrix were dose-dependently enhanced by DP, and the adhesion was mediated by α5β1 integrin. The cytoskeleton was more organized in the cells that adhered to the fibrin-fibronectin-DP complex. When incubated with DP, fibronectin formed an insoluble complex of fibronectin fibrils as visualized by electron microscopy. The interacting sites of fibronectin with DP were the first, thirteenth, and fourteenth type III repeats (III(1), III(13), and III(14)), with III(13) and III(14) assumed to be the major sites. The interaction between III(2-3) and III(12-14) was inhibited by DP, whereas the interaction between I(1-5) and III(12-14) was specifically and strongly enhanced by DP. Because the interaction between III(2-3) and III(12-14) is involved in forming a globular conformation of fibronectin, and that between I(1-5) and III(12-14) is required for forming fibronectin fibrils, DP promotes fibronectin fibril formation probably by changing the fibronectin conformation. These results suggest that DP has an accelerating role in fibroblast cell adhesion to the provisional matrix in the initial stage of wound healing.  相似文献

Bacteriophages (phages) belonging to the family Podoviridae genus N4‐like viruses have been used as therapeutic agent in phage therapy against Pseudomonas aeruginosa infections. P. aeruginosa phage KPP21 was isolated in Japan, and phylogenetically investigated the phages belonging to this viral genus. Morphological and genetic analyses confirmed that phage KPP21 belongs to the family Podoviridae genus N4‐like viruses. Moreover, phylogenetic analyses based on putative DNA polymerase and major virion protein showed that P. aeruginosa phages belonging to the genus N4‐like viruses are separated into two lineages and that phage KPP21 is in the same clade as phage LUZ7.  相似文献

Background: Tumor necrosis factor (TNF)‐α‐inducing protein (Tipα) is a newly identified carcinogenic factor present in Helicobacter pylori. Tipα has the unique function of inducing TNF‐α production by gastric cells in vitro and is assumed to be related with the development of gastritis and gastric cancer. We investigated the effects of vaccination with Tipα against H. pylori infection and analyzed the immune responses. Methods: C57BL/6 mice were immunized via the intranasal route with CpG, recombinant Tipα + CpG, and recombinant del‐Tipα (a mutant of Tipα) + CpG. Eight weeks after the mice were infected with H. pylori (5 × 107 CFU), the number of colonizing bacteria in the stomach was calculated, and the histological severity of gastritis was evaluated. Levels of Tipα‐specific IgG and IgA antibodies in mouse serum were measured by an enzyme‐linked immunosorbent assay (ELISA). Local production of cytokines including Interleukin (IL)‐10, TNF‐α and Interferon (IFN)‐γ in gastric mucosa was also measured by real time‐PCR. Results: Levels of Tipα‐specific antibodies were significantly higher in Tipα‐immunized and del‐Tipα‐immunized mice than in the infection control group. The numbers of colonizing bacteria were significantly reduced in Tipα‐immunized mice (4.29 × 105 CFU/g) and del‐Tipα immunized mice (2.5 × 105 CFU/g) compared with infection control mice (5.7 × 106 CFU/g). The levels of IFN‐γ and IL‐10 were significantly higher in del‐Tipα‐immunized mice than the infection control group. Conclusion: Vaccinations with Tipα and del‐Tipα were effective against H. pylori infection. The inhibition of H. pylori colonization is associated mainly with Th1 cell‐mediated immunity.  相似文献

Haury et al., (Deep Sea Res., 37, 447–461, 1990) describedhow turbulence and vertical shear generated by the passage ofa storm affected the vertical distributions and community structureof plankton in Monterey Bay, CA. The data presented came froma larger set of five plankton recorder collections of zooplankton,taken together with temperature, salinity, turbulent kineticenergy dissipation rate, and vertical current shear data onthe research submarine Dolphin. Here we report on the analysisof the whole data set, which represents a wide range of physicaldynamic conditions. While the overall community structure remainedrelatively constant over the sampling period, the relationshipsbetween taxonomic categories and between categories and thephysical dynamics were complex and variable; factors relatedto depth were the most important. During periods of weak winds(low kinetic energy conditions) some of the biological variabilityappeared to be related to the level of turbulent dissipation.Other relationships were not detected due to the limitationsof the plankton sampler and its interactions with the highlyvariable biological and physical conditions. In view of thesedifficulties, we recommend that future field investigationsof the relationships between physical parameters and zooplanktondistributions make greater use of acoustic and other remote-sensingtechnologies, preferably in combination with numerical simulationstudies to help guide and interpret the field work.  相似文献
