The apoplast of mature leaves of the tropical orchid OncidiumGoldiana was perfused with 0.5 mM p-chloromercuribenzenesulphonicacid (PCMBS) via the transpiration stream in order to test themode of phloem loading. The efficacy of introducing PCMBS byperfusion was shown by saffranin O dye movement in the veinsand leaf apoplast in control experiments. Photoassimilate exportas the result of phloem loading was measured by collection of14CO2-derived photoassimilates from the basal cut-ends of intactleaves. Phloem loading and translocation of photoassimilates was inhibitedby 89% in leaves perfused with PCMBS for 1 h. The effect ofPCMBS on leaf photosynthesis was minimal. The amount of radiocarbonfixed by PCMBS-treated leaves averaged 89% of control leavesperfused with distilled water. A negative correlation betweenthe total amount of photoassimilate exuded and the calculatedconcentration of PCMBS in the leaf apoplast was also observed.The results indicate that phloem loading in Oncidium Goldianaoccurs via the apoplastic pathway. The report that microvillar cores of isolated, demembranated brush borders retract into the terminal web in the presence of Ca(++) and ATP has been widely cited as an example of Ca(++)-regulated nonmuscle cell motility. Because of recent findings that microvillar core actin filaments are cross-linked by villin which, in the presence of micromolar Ca(++), fragments actin filaments, we used the techniques of video enhanced differential interference contrast, immunofluorescence, and phase contrast microscopy and thin-section electron microscopy (EM) to reexamine the question of contraction of isolated intestinal cell brush borders. Analysis of video enhanced light microscopic images of Triton- demembranated brush borders treated with a buffered Ca(++) solution shows the cores disintegrating with the terminal web remaining intact; membranated brush borders show the microvilli to vesiculate with Ca(++). Using Ca(++)/EGTA buffers, it is found that micromolar free Ca(++) causes core filament dissolution in membranated or demembranated brush borders, Ca(++) causes microvillar core solation followed by complete vesiculation of the microvillar membrane. The lengths of microvilli cores and rootlets were measured in thin sections of membranated and demembranated controls, in Ca(++)-, Ca(++) + ATP-, and in ATP-treated brush borders. Results of these measurements show that Ca(++) alone causes the complete solation of the microvillar cores, yet the rootlets in the terminal web region remain of normal length. These results show that microvilli do not retract into the terminal web in response to Ca(++) and ATP but rather that the microvillar cores disintegrate. NBD-phallicidin localization of actin and fluorescent antibodies to myosin reveal a circumferential band of actin and myosin in mildly permeabilized cells in the region of the junctional complex. The presence of these contractile proteins in this region, where other studies have shown a circumferential band of thin filaments, is consistent with the hypothesis that brush borders may be motile through the circumferential constriction of this “contractile ring,” and is also consistent with the observations that ATP-treated brush borders become cup shaped as if there had been a circumferential constriction. 相似文献

The first overview is presented of the paper contents of Kew's Economic Botany Collections (EBC). There are more than 350 specimens of paper representing nearly 50 plant families and more than a hundred genera. The dominant families numerically – and each for different reasons – are the Moraceae , Gramineae , Thymelaeaceae and Cyperaceae , examples from which are highlighted. Some emphasis is given to the historical context and geographical origin of papers, and to the people who collected or donated them. It is suggested that one focus of a future collecting strategy for the EBC should be the sorts of plant products, including papers, that are under threat due to losses of cultural diversity or biological diversity, or both. 相似文献

The amino acid sequences of the two major antifreeze polypeptides (AFP) from the shorthorn sculpin have been determined using an automatic protein sequencer and enzymic digestion. These two polypeptides, SS-3 and SS-8, consist of 33 and 45 amino acid residues respectively. The N-terminal methionyl residue is blocked in both the polypeptides. When aligned for maximum structural similarity these two AFP are 80% homologous, and there appears a deletion of 12 amino acid residues at the N-terminal portion of SS-3. Like the winter flounder AFP, both the sculpin AFP also contain the 11-amino-acid repeat sequences. The secondary structure of the sculpin AFP is mainly alpha-helical as deduced from circular dichroic spectral data. The helical content of SS-8 is high (73%), while that of SS-3 is moderate (about 45%). The latter exhibits a relatively weak antifreeze activity. Removal of the blocked N-terminal residue in SS-8 did not alter the helical content significantly but did reduce the antifreeze activity. Helical contents of proteolytically generated fragments of AFP are much lower, and they are devoid of activity. The alpha-helix in the SS-8 component is seen to be amphiphilic in character. The relevance of this feature to the mechanism of the antifreeze action is briefly discussed. 相似文献

The effect of growth temperature on the bioenergetics of photosynthetic algal cultures 总被引:4,自引:0,他引:4
Two photosynthetic algal cultures, one Chlorella vulgaris, and the other a Chlorogonium sp., were cultured under light limitations in chemostats. The effects of growth temperature on their energy yield and maintenance energy requirement were studied. It was observed that a lowering in temperature resulted in a lower maximum growth yield from the light energy, Y(G). This was attributed to two reasons. First, at low temperatures there was a change in the algal cell composition with more energy being expended to synthesize a higher biomass protein content. Secondly, at low temperatures, a cyanide-resistant respiratory pathway became operative which led to a decrease in the number of ATP being generated. The maintenance energy coefficient was a function of temperature increasing with decreasing temperature. This might reflect energy wastage by the cell at low temperatures. The maximum specific growth rate dropped with decreasing temperature, and can be described by an Arrhenius type rate-temperature model up to the optimal temperature for growth; i.e., activation energy remained constant. 相似文献

14CO2 fixation was observed in orchid Dendrobium flowers; its rate decreased with the flower development. Chlorophyll (Chl) fluorescence in different developmental stages of flowers was compared to other green plant parts (leaf, inflorescence stalk, and fruit capsule). The photochemical efficiency of photosystem 2 (PS2) (Fv/Fm) of a leaf was 14-21 % higher than that of a mature flower perianth (sepal, petal, and labellum) which had a much lower total Chl content and Chl a/b ratio. A higher quantum yield of PS2 (PS2) than in the mature flowers was observed in all green parts. Flower sepals had higher Chl content, Chl a/b ratio, and Fv/Fm values than the petal and labellum. During flower development the Chl content, Chl a/b ratio, Fv/Fm, and qN decreased while PS2 and qP remained constant. An exposure of developing flowers to irradiances above 50 µmol m-2 s-1 resulted in a very drastic drop of PS2 and qP, and a coherent increase of qN as compared to other green plant organs. A low saturation irradiance (PFD of 100 µmol m-2 s-1) and the increase in qN in the flower indicate that irradiation stress may occur since there is no further protection when the flower is exposed to irradiances above 100 µmol m-2 s-1. A low Chl/carotenoid ratio in mature flower perianth as a consequence of Chl content reduction in the course of flower development suggests a relief of irradiation stress via this mean. 相似文献

In mammals, the pituitary POU homeodomain protein, Pit-1, binds to proximal and distal 5'-flanking sequences of the PRL gene that dictate tissue-specific expression. These DNA sequences are highly conserved among mammals but are dramatically different from PRL 5' sequences in the teleost species, Oncorhynchus tschawytscha (chinook salmon). To analyze the molecular basis for pituitary-specific gene expression in a distantly related vertebrate, we transfected CAT reporter gene constructs containing 2.4 kilobases (kb) 5'-flanking sequence from the salmon PRL (sPRL) gene into various cell types. Expression of the sPRL gene was restricted to pituitary cells, but in rat pituitary GH4 cells levels of expression were at least 90-fold lower than those obtained with a -3 kb rat PRL (rPRL) construct. Conversely, in primary teleost pituitary cells, -2.4 kb sPRL/CAT was expressed at levels about 10-fold higher than -3 kb rPRL/CAT. To determine whether species-specific transactivation by Pit-1 was sufficient to explain these species differences in PRL gene expression, we isolated a cDNA clone encoding the salmon Pit-1 POU domain and constructed a rat Pit-1 expression vector that contained salmon Pit-1 POU domain sequences substituted in frame. The chimeric Pit-1 encoded 14 amino acids unique to salmon. Coexpression of rat Pit-1 with salmon or rat PRL/CAT in transfected HeLa cells resulted in specific and strikingly comparable levels of promoter activation. To develop an all-fish gene cassette suitable for gene transfer in aquaculture, the antifreeze protein (AFP) gene promoter from the ocean pout (Macrozoarces americanus) was analyzed for its ability to direct exogenous gene expression both in vitro and in vivo. The ocean pout AFP (opAFP) gene promoter fused to the bacterial chloramphenicol acetyltransferase (CAT) was functionally analyzed in two fish cell lines and in Japanese medaka embryos. The opAFP gene promoter was active in these systems, as demonstrated by the transient expression of CAT activity. These results suggest that the opAFP gene promoter is useful for many other gene transfer experiments. To facilitate use of the opAFP gene promoter as a common and versatile vehicle for fish gene transfers, an expression vector, opAFP-V, was constructed by linking the 2.1-kb opAFP gene promoter, the 63-bp opAFP gene 5' untranslated sequence, and the 1.2-kb opAFP gene 3' sequence by two unique restriction sites, Bg/II and HpaI, respectively. Thus, genes of interest can be inserted into either the Bg/II site or the HpaI site depending on the length of their 5' untranslated sequence. Acidity fluctuation, CO2 gas exchange, δ13 C value, PEP carboxylase and RuBP carboxylase activities in aerial roots of selected thick-leaved orchid hybrids ( Arachnis and Aranthera ) were studied. Both aerial roots and leaves showed acidity fluctuation over a 24 h period. Dark acidification in aerial roots was enhanced at low temperature (15°C). Aerial roots had δ13 C values close to those of leaves which have been previously demonstrated to possess crassulacean acid metabolism. Variation in δ13 C values along the length of the roots was observed; the root tip having a less negative δ13 C value (—13.34%‰) than the older portions of the roots (—14.55%‰). There was no net CO2 fixation by aerial root, although 1432 CO2 fixation was observed in light and in darkness. The pattern of fluctuation in activities of PEP carboxylase and RuBP carboxylase in aerial roots was similar to that obtained for the leaves. In both aerial roots and leaves, PEP carboxylase activity was several times higher than that of RuBP carboxylase. 相似文献
