Enterohemorrhagic and enteropathogenic Escherichia coli (EHEC and EPEC, respectively) strains are closely related human pathogens that are responsible for food-borne epidemics in many countries. Integration host factor (IHF) and the locus of enterocyte effacement-encoded regulator (Ler) are needed for the expression of virulence genes in EHEC and EPEC, including the elicitation of actin rearrangements for attaching and effacing lesions. We applied a proteomic approach, using two-dimensional polyacrylamide gel electrophoresis in combination with matrix-assisted laser desorption ionization-time of flight mass spectrometry and a protein database search, to analyze the extracellular protein profiles of EHEC EDL933, EPEC E2348/69, and their ihf and ler mutants. Fifty-nine major protein spots from the extracellular proteomes were identified, including six proteins of unknown function. Twenty-six of them were conserved between EHEC EDL933 and EPEC E2348/69, while some of them were strain-specific proteins. Four common extracellular proteins (EspA, EspB, EspD, and Tir) were regulated by both IHF and Ler in EHEC EDL933 and EPEC E2348/69. TagA in EHEC EDL933 and EspC and EspF in EPEC E2348/69 were present in the wild-type strains but absent from their respective ler and ihf mutants, while FliC was overexpressed in the ihf mutant of EPEC E2348/69. Two dominant forms of EspB were found in EHEC EDL933 and EPEC E2348/69, but the significance of this is unknown. These results show that proteomics is a powerful platform technology for accelerating the understanding of EPEC and EHEC pathogenesis and identifying markers for laboratory diagnoses of these pathogens.  相似文献

Molecular cloning and sequencing of salmon gonadotropin beta subunit   总被引:2,自引:0,他引:2  
Gonadotropin (GTH) was purified from the pituitaries of the Pacific chinook salmon using a combination of stepwise ethanol precipitation and concanavalin-A affinity chromatography. The alpha and beta subunits were dissociated and fractionated by C-18 reverse-phase high-performance liquid chromatography with a 0.1% trifluoroacetic acid/acetonitrile gradient. An enriched cDNA library was screened for the beta-GTH gene(s) using two synthetic oligonucleotides based on partial protein data. A positive, full-size clone (E3) was identified and sequenced. It contains 657 base pairs and codes for a 142-amino-acid precursor protein. The mature salmon beta-GTH (119 amino acids) is structurally homologous to human luteinizing hormone and chorionic gonadotropin. The effect of testosterone implantation on pituitary GTH and beta-GTH mRNA was examined with radioimmunoassay and Northern blot analysis. There was a corresponding increase in both the pituitary GTH and mRNA levels.  相似文献

The precursor proteins for winter flounder antifreeze polypeptide (AFP) were isolated from liver using gel filtration chromatography and reverse-phase high-performance liquid chromatography. Two major pro-antifreezes (Mr 5000), corresponding to the precursors for AFP-6 and AFP-8, were characterized by amino acid analyses and automated Edman degradation. These precursors showed significant antifreeze activity. The pro-antifreezes were synthesized in the liver seasonally as demonstrated by immunoblotting and in vitro liver incorporation studies. No mature AFP were detected in liver, thus indicating that the processing of pro-antifreezes, including amidation of the C-termini, occurred mainly in the serum. The function(s) of the prosequences, if any, remain unclear.  相似文献

Using synthetic analogs of an alpha-helical winter flounder antifreeze polypeptide (AFP) we investigated some important molecular details of the mechanism of action of this AFP. Of the seven peptides synthesized, all but one were amino-terminal deletions of the native AFP. Three of the seven synthetic analogs possessed the same antifreeze activity as the native polypeptide; the other analogs were devoid of antifreeze activity. The growth rates along the a and c axes of ice in solutions of varying concentrations of the three active AFP analogs were examined. The a axis growth rates of ice were inversely proportional to the concentration of the active peptides. The c-axis growth rates of ice were also dependent on peptide concentration. The active peptides enhanced c-axis growth at lower concentrations, while at higher concentrations they inhibited c axis growth. The ability of the peptides to develop antifreeze activity and to alter the a and c axis growth rates of ice depended on the presence of appropriately positioned amino acid residues with hydrogen bonding side chains. From these observations we propose that at low concentrations the AFP, through dipolar interactions and hydrogen bonding, interact with the prism faces of ice retarding a axis growth. At these concentrations, the electrical field of the AFP helix-dipole, like an externally applied field (Bartlett, J.T., van der Heuval, A.P., and Mason, B.J. (1963) Z. Angew. Math. Phys. 14, 599-610), can enhance ice c axis growth. At higher concentrations, the AFP interact with all ice crystal planes and retard both a and c axis growth.  相似文献

The ocean pout (Macrozoarces americanus) produces a set of antifreeze proteins that depresses the freezing point of its blood by binding to, and inhibiting the growth of, ice crystals. The amino acid sequences of all the major components of the ocean pout antifreeze proteins, including the immunologically distinct QAE component, have been derived by Edman degradation. In addition, sequences of several minor components were deduced from DNA sequencing of cDNA and genomic clones. Fifty percent of the amino acids are perfectly conserved in all these proteins as well as in two homologous sequences from the distantly related wolffish. Several of the conserved residues are threonines and asparagines, amino acids that have been implicated in ice binding in the structurally unrelated antifreeze protein of the righteye flounders. Aside from minor differences in post-translational modifications, heterogeneity in antifreeze protein components stems from amino acid differences encoded by multiple genes. Based on genomic Southern blots and library cloning statistics there are 150 copies of the 0.7-kilobase-long antifreeze protein gene in the Newfoundland ocean pout, the majority of which are closely linked but irregularly spaced. A more southerly population of ocean pout from New Brunswick in which the circulating antifreeze protein levels are considerably lower has approximately one-quater as many antifreeze protein genes. Thus, there appears to be a correlation between gene dosage and antifreeze protein levels, and hence the ability to survive in ice-laden seawater. Southern blot comparison of the two populations indicates that the differences in gene dosage were not generated by a simple set of deletions/duplications. They are more likely to be the result of differential amplification.  相似文献

14CO2 fixation was observed in orchid Dendrobium flowers; its rate decreased with the flower development. Chlorophyll (Chl) fluorescence in different developmental stages of flowers was compared to other green plant parts (leaf, inflorescence stalk, and fruit capsule). The photochemical efficiency of photosystem 2 (PS2) (Fv/Fm) of a leaf was 14-21 % higher than that of a mature flower perianth (sepal, petal, and labellum) which had a much lower total Chl content and Chl a/b ratio. A higher quantum yield of PS2 (ΦPS2) than in the mature flowers was observed in all green parts. Flower sepals had higher Chl content, Chl a/b ratio, and Fv/Fm values than the petal and labellum. During flower development the Chl content, Chl a/b ratio, Fv/Fm, and qN decreased while ΦPS2 and qP remained constant. An exposure of developing flowers to irradiances above 50 µmol m-2 s-1 resulted in a very drastic drop of ΦPS2 and qP, and a coherent increase of qN as compared to other green plant organs. A low saturation irradiance (PFD of 100 µmol m-2 s-1) and the increase in qN in the flower indicate that irradiation stress may occur since there is no further protection when the flower is exposed to irradiances above 100 µmol m-2 s-1. A low Chl/carotenoid ratio in mature flower perianth as a consequence of Chl content reduction in the course of flower development suggests a relief of irradiation stress via this mean.  相似文献

本文对兰属(Cymbidium)植物的七个种中的10个栽培种的光合途径进行了生理生化研究。结果表明,这些兰属植物叶片的磷酸烯醇式丙酮酸羧化酶(PEPCase)和丙酮酸磷酸双激酶(PPDK)的活性很低,乙醇酸氧化酶的活性较高。在光下用α-羟基吡啶甲烷磺酸(α-HPMS)处理叶片后,乙醇酸积累量明显高于对照。叶绿素a/b比值在1.24—2.69之间。光合速率最低为0.96—3.33μmlCO2m-2S-1。这些结果表明所研究的几种兰属植物均为C3植物。  相似文献

Research involving plant growth regulators (PGRs) and orchids in areas of orchid growth and development are reviewed. For all areas covered in the review — seed germination and seedling growth, lateral shoot production, root production, flower initiation and development, postharvest physiology, and photosynthate partitioning — it was concluded that further studies would assist in clarifying potential uses for PGRs in the orchid cut-flower industry. It is suggested that extra PGR research on orchids is justified at the present time because of favourable prospects facing the orchid cut-flower industry.  相似文献
