The effects of a submerged macrophyte (Potamogeton pectinatus), turbidity (produced by 50 mg l–1 kaolin), and their combination on the feeding rate of young razor fish (Pelecus cultratus (L.)) were studied in the laboratory. Young razor fish showed preference for Copepoda in most of the simulated habitats. Razor fish fed more efficiently in the absence of any environmental structure, but the more structured habitats, 3 g wet wt l–1 of simulated submerged macrophyte and turbidity combined with vegetation, had a negative impact on food consumption rates.  相似文献

Mitochondrial import of pyruvate by the mitochondrial pyruvate carrier (MPC) is a central step which links cytosolic and mitochondrial intermediary metabolism. To investigate the role of the MPC in mammalian physiology and development, we generated a mouse strain with complete loss of MPC1 expression. This resulted in embryonic lethality at around E13.5. Mouse embryonic fibroblasts (MEFs) derived from mutant mice displayed defective pyruvate-driven respiration as well as perturbed metabolic profiles, and both defects could be restored by reexpression of MPC1. Labeling experiments using 13C-labeled glucose and glutamine demonstrated that MPC deficiency causes increased glutaminolysis and reduced contribution of glucose-derived pyruvate to the TCA cycle. Morphological defects were observed in mutant embryonic brains, together with major alterations of their metabolome including lactic acidosis, diminished TCA cycle intermediates, energy deficit and a perturbed balance of neurotransmitters. Strikingly, these changes were reversed when the pregnant dams were fed a ketogenic diet, which provides acetyl-CoA directly to the TCA cycle and bypasses the need for a functional MPC. This allowed the normal gestation and development of MPC deficient pups, even though they all died within a few minutes post-delivery. This study establishes the MPC as a key player in regulating the metabolic state necessary for embryonic development, neurotransmitter balance and post-natal survival.  相似文献

The device performance of organic polymer:fullerene bulk heterojunction solar cells strongly depends on the interpenetrating network of the involved donor and acceptor materials in the active layer. Since morphology formation depends on the conditions of film preparation, the final morphology varies for different deposition methods. In order to understand and optimize industrial coating processes and, therefore, the performance of the solar cells produced, a deeper understanding of structure formation is important. In situ measurements of slot‐die printed polymer:fullerene active layers are presented that reveal insights into the evolution of the structure. Polymer crystallization and ordering is monitored by in situ grazing incidence wide angle X‐ray scattering (GIWAXS), and in situ grazing incidence small‐angle X‐ray scattering (GISAXS). The development of the morphology exhibits five stages independent of the drying conditions. Two growth rates are observed, an initial slow formation of poly(3‐hexylthiophene‐2,5‐diyl) crystallites in well‐aligned edge‐on orientation followed by a rapid crystal growth. By combining the GIWAXS and GISAXS measurements, a five‐stage growth and assembly process is found and described in detail along with a proposed model of the structural evolution. The findings are an important step in tailoring the assembly process.  相似文献

We have developed chemical affinity reagents for the hepatic galactosyl receptor. Asialoorosomucoid (ASOR) was derivatized with five homobifunctional N-hydroxysuccinimide (NHS) ester cross-linkers. NHS/ASOR derivatives were synthesized, purified, and applied within 10 min to isolated rat hepatocytes at 4 degrees C. Specific binding of these 125I-labeled derivatives was approximately 90% in the presence of either EGTA or excess ASOR. Specific cross-linking assessed by the resistance of specifically bound NHS/125I-ASOR to release by EGTA, was 50-75% of the specifically bound ligand. The extent of specific cross-linking correlated with the average number of NHS groups per ASOR and was controlled by varying the molar ratio of cross-linker to ASOR during the synthesis. Cross-linking proceeded rapidly at 4 degrees C as a first-order process (k = 0.25 min-1, t1/2 = 2.8 min). After being cross-linked with any of the NHS/125I-ASOR derivatives, cells were washed with EGTA, solubilized in Triton X-100, and analyzed by SDS-PAGE and autoradiography. Major bands were observed at Mr congruent to 84K, 93K, and 105K corresponding to the expected size of 1:1 adducts between NHS/ASOR (Mr congruent to 41.3K) and the three subunits of the receptor, Mr congruent to 43K, 50K, and 60K. The three subunits, rat hepatic lectin (RHL) 1, 2, and 3, were labeled in the ratio of about 1.0:1.2:1.0, respectively. After cross-linking, a polyclonal goat antibody to the receptor immunoprecipitated up to 100% of the specifically cross-linked NHS/125I-ASOR. Preimmune IgG immunoprecipitated less than 1% of the radiolabeled ligand. Cell surface receptors were cross-linked to NHS-ASOR, extracted with Triton X-100, immunoprecipitated with anti-orosomucoid-Sepharose, and subjected to Western blot analysis. By use of anti-sera specific for RHL 1 or RHL 2/3 (from K. Drickamer), cross-linked complexes of Mr congruent to 85K or approximately 90-115K, respectively, were detected as were un-cross-linked native subunits. The ratio of free to cross-linked subunits was approximately 10:1 for RHL 1 and approximately 0.5:1 for RHL 2/3. We conclude that all three receptor subunits can cross-link to ligand. We propose a model in which the native receptor is a heterohexamer composed of four subunits of RHL 1 and two subunits of RHL 2 and/or RHL 3.  相似文献

In Neusiedler See, a shallow alkaline lake with fluctuating water level and salinity, four species of Hexarthra occur: H. mira, H. fennica, H. jenkinae (occasional) and H. polyodonta. The analysis of longterm data reveals a general phenological pattern which does not change from year to year. They first occur in May, develop a maximum in June/July, sometimes a second one in August/September and disappear in October. But the species succession is different in the various years, occasionally only one species (H. mira or H. polyodonta) being present. There is a fairly consistent relation between the chemical conditions and the prevalent species; an increase in salinity favours the development of H. polyodonta. Low temperature and wind generated suspended particles have a negative influence on the development of the Hexarthra populations. Smaller populations of Hexarthra are in a relation to the occurrence of Leptodora indicating predation pressure of the latter species. In Neusiedler See the Hexarthra populations seem to be controlled to a great extent by abiotic factors, but predation by Leptodora and most probably by young fish seems to play an important role too.  相似文献

The best performing low bandgap copolymers PTB series to date which is based on thieno[3,4‐b]thiophene‐alt‐benzodithiophene units blended with [6,6]‐phenyl‐C71‐butyric acid methyl ester (PC71BM), have been the focus of polymer‐based solar cells. Here, novel fluorinated polymers PTB7‐Fx (fluorine units coupled with submonomer thieno[3,4‐b]thiophene) with varied degree of fluorination are used as electron donor materials. The PTB7‐Fx:PC71BM bulk heterojunction (BHJ) films spin‐coated from the host solvent chlorobenzene without and with solvent additive 1,8‐diiodooctane (DIO) and the corresponding solar cell devices are systematically investigated to address the morphology‐efficiency relationship. Self‐assembled BHJ morphology is already observed for as‐spun blend films. After adding the solvent additive DIO, the pronounced ordered structures are suppressed and better intermixed films with much smaller domain sizes result. Full fluorination of the third C‐atom of thienothiophene gives rise to the highest power conversion efficiency. As the absorption properties, film morphology and crystallinity remain similar for different degrees of fluorination, the main influence of the photovoltaic performance is ascribed to the different lowest unoccupied molecular orbital (LUMO) of each polymer instead of the film morphology. Thus the device performance can be efficiently improved by tuning the energy level of the polymer without necessarily changing either the film nanomorphology or crystallinity dramatically.  相似文献

WC1 molecules are implicated in augmenting cellular activation as well as inducing cell cycle arrest of gammadelta T cells. Since WC1 is a large multigene family differences in outcome could result from modulation of different WC1 molecules. To further investigate this family of molecules, peripheral blood WC1(+) gammadelta T cell subpopulations were evaluated by 2-D Western blotting and RT-PCR. We found 13 cDNA intracytoplasmic tail sequences with differences in signaling motifs among them and at least 20 biochemically distinguishable WC1 spots associated with cell membranes, with some in lipid rafts. An understanding of the diversity of 2-D spots could not be resolved by evaluating T cell clones, removing sialyated carbohydrates or blotting with anti-WC1.1 or anti-WC1.2-specific antibodies. Nevertheless, while the major gammadelta T cell subpopulations in blood (WC1.1(+)/WC1.2(-) and WC1.2(+)/WC1.1(-)) both had complex 2-D patterns, virtually all spots associated with WC1.2(+)/WC1.1(-) cells bore the WC1.2 epitope, distinguishing them from the WC1.1(+) cells.  相似文献
