From specimens of the bryozoan Membranipora membranacea collected in the Baltic Sea, bacteria were isolated on four different media, which significantly increased the diversity of the isolated groups. All isolates were classified according to 16S rRNA gene sequence analysis and tested for antimicrobial properties using a panel of five indicator strains and six different media. Each medium featured a unique set of isolated phylotypes, and a phylogenetically diverse collection of isolates was obtained. A total of 96 isolates were assigned to 49 phylotypes and 29 genera. Only one-third of the members of these genera had been isolated previously from comparable sources. The isolates were affiliated with Alpha- and Gammaproteobacteria, Bacilli, and Actinobacteria. A comparable large portion of up to 22 isolates, i.e., 15 phylotypes, probably represent new species. Likewise, 47 isolates (approximately 50%) displayed antibiotic activities, mostly against grampositive indicator strains. Of the active strains, 63.8 % had antibiotic traits only on one or two of the growth media, whereas only 12.7 % inhibited growth on five or all six media. The application of six different media for antimicrobial testing resulted in twice the number of positive hits as obtained with only a single medium. The use of different media for the isolation of bacteria as well as the variation of media considered suitable for the production of antibiotic substances significantly enhanced both the number of isolates obtained and the proportion of antibiotic active cultures. Constraints on lake communities are complex and are usually studied by using limited combinations of variables derived from measurements within or adjacent to study waters. While informative, results often provide limited insight about magnitude of simultaneous influences operating at multiple scales, such as lake- vs. watershed-scale. To formulate comparisons of such contrasting influences, we explored factors controlling the abundance of predominant aquatic invertebrates in 75 shallow lakes in western Minnesota, USA. Using robust regression techniques, we modeled relative abundance of Amphipoda, small and large cladocera, Corixidae, aquatic Diptera, and an aggregate taxon that combined Ephemeroptera-Trichoptera-Odonata (ETO) in response to lake- and watershed-scale characteristics. Predictor variables included fish and submerged plant abundance, linear distance to the nearest wetland or lake, watershed size, and proportion of the watershed in agricultural production. Among-lake variability in invertebrate abundance was more often explained by lake-scale predictors than by variables based on watershed characteristics. For example, we identified significant associations between fish presence and community type and abundance of small and large cladocera, Amphipoda, Diptera, and ETO. Abundance of Amphipoda, Diptera, and Corixidae were also positively correlated with submerged plant abundance. We observed no associations between lake-watershed variables and abundance of our invertebrate taxa. Broadly, our results seem to indicate preeminence of lake-level influences on aquatic invertebrates in shallow lakes, but historical land-use legacies may mask important relationships. 相似文献

The formation of intersegmental blood vessels (ISVs) in the zebrafish embryo serves as a paradigm to study angiogenesis in vivo. ISV formation is thought to occur in discrete steps. First, endothelial cells of the dorsal aorta migrate out and align along the dorsoventral axis. The dorsal-most cell, also called tip cell, then joins with its anterior and posterior neighbours, thus establishing a simple vascular network. The vascular lumen is then established via formation of vacuoles, which eventually fuse with those of adjacent endothelial cells to generate a seamless tube with an intracellular lumen. To investigate the cellular architecture and the development of ISVs in detail, we have analysed the arrangement of endothelial cell junctions and have performed single cell live imaging. In contrast to previous reports, we find that endothelial cells are not arranged in a linear head-to-tail configuration but overlap extensively and form a multicellular tube, which contains an extracellular lumen. Our studies demonstrate that a number of cellular behaviours, such as cell divisions, cell rearrangements and dynamic alterations in cell-cell contacts, have to be considered when studying the morphological and molecular processes involved in ISV and endothelial lumen formation in vivo. 相似文献

The mysterious death of Mr. Alexander Litvinenko who was most possibly poisoned by Polonium-210 (210Po) in November 2006 in London attracted the attention of the public to the kinetics, dosimetry and the risk of this high
radiotoxic isotope in the human body. In the present paper, the urinary excretion of seven persons who were possibly exposed
to traces of 210Po was monitored. The values measured in the GSF Radioanalytical Laboratory are in the range of natural background concentration.
To assess the effective dose received by those persons, the time-dependence of the organ equivalent dose and the effective
dose after acute ingestion and inhalation of 210Po were calculated using the biokinetic model for polonium (Po) recommended by the International Commission on Radiological
Protection (ICRP) and the one recently published by Leggett and Eckerman (L&E). The daily urinary excretion to effective dose
conversion factors for ingestion and inhalation were evaluated based on the ICRP and L&E models for members of the public.
The ingestion (inhalation) effective dose per unit intake integrated over one day is 1.7 × 10−8 (1.4 × 10−7) Sv Bq−1, 2.0 × 10−7 (9.6 × 10−7) Sv Bq−1 over 10 days, 5.2 × 10−7 (2.0 × 10−6) Sv Bq−1 over 30 days and 1.0 × 10−6 (3.0 × 10−6) Sv Bq−1 over 100 days. The daily urinary excretions after acute ingestion (inhalation) of 1 Bq of 210Po are 1.1 × 10−3 (1.0 × 10−4) on day 1, 2.0 × 10−3 (1.9 × 10−4) on day 10, 1.3 × 10−3 (1.7 × 10−4) on day 30 and 3.6 × 10−4 (8.3 × 10−5) Bq d−1 on day 100, respectively. The resulting committed effective doses range from 2.1 × 10−3 to 1.7 × 10−2 mSv by an assumption of ingestion and from 5.5 × 10−2 to 4.5 × 10−1 mSv by inhalation. For the case of Mr. Litvinenko, the mean organ absorbed dose as a function of time was calculated using
both the above stated models. The red bone marrow, the kidneys and the liver were considered as the critical organs. Assuming
a value of lethal absorbed dose of 5 Gy to the bone marrow, 6 Gy to the kidneys and 8 Gy to the liver, the amount of 210Po which Mr. Litvinenko might have ingested is therefore estimated to range from 27 to 1,408 MBq, i.e 0.2–8.5 μg, depending

A recent meta‐analysis indicates that trophic cascades (indirect effects of predators on plants via herbivores) are weak in marine plankton in striking contrast to freshwater plankton ( Shurin et al. 2002 , Ecol. Lett., 5, 785–791). Here we show that in a marine plankton community consisting of jellyfish, calanoid copepods and algae, jellyfish predation consistently reduced copepods but produced two distinct, opposite responses of algal biomass. Calanoid copepods act as a switch between alternative trophic cascades along food chains of different length and with counteracting effects on algal biomass. Copepods reduced large algae but simultaneously promoted small algae by feeding on ciliates. The net effect of jellyfish on total algal biomass was positive when large algae were initially abundant in the phytoplankton, negative when small algae were dominant, but zero when experiments were analysed in combination. In contrast to marine systems, major pathways of energy flow in Daphnia‐dominated freshwater systems are of similar chain length. Thus, differences in the length of alternative, parallel food chains may explain the apparent discrepancy in trophic cascade strength between freshwater and marine planktonic systems. 相似文献

Summary Organ cultures of rodent and human prostate glands have shown marked differences in their morphological response to testosterone.
In this study, explants from 19 canine prostate glands were cultivated for a minimum of 9 days in Trowell’s T-8 medium. Groups
of explants were exposed to media containing from 0.05 to 100 μm testosterone. While the higher testosterone levels (50 and
100 μm) markedly decreased explant viability, explants cultivated at lower levels (0.05 to 5 μm) appeared similar to control
explants in testosterone-free Trowell’s T-8 medium. Atmospheric mixtures containing either 95% or 50% oxygen were equally
effective.
Shortly after the cultures were initiated, large amounts of secretory product were liberated into the lumen. After 9 or more
days in vitro, glandular epithelium appeared cuboidal and never revealed the acid phosphatase-rich secretory granules seen
in the preculture control. However, the epithelium exhibited an increase in alkaline phosphatase and lipid content following
cultivation.

A series of diaryl ethers were designed and synthesized to discern the structure activity relationships against the two closely related mono-(ADP-ribosyl)transferases PARP10 and PARP14. Structure activity studies identified 8b as a sub-micromolar inhibitor of PARP10 with?~15-fold selectivity over PARP14. In addition, 8k and 8m were discovered to have sub-micromolar potency against PARP14 and demonstrated moderate selectivity over PARP10. A crystal structure of the complex of PARP14 and 8b shows binding of the compound in a novel hydrophobic pocket and explains both potency and selectivity over other PARP family members. In addition, 8b, 8k and 8m also demonstrate selectivity over PARP1. Together, this study identified novel, potent and metabolically stable derivatives to use as chemical probes for these biologically interesting therapeutic targets. 相似文献
