A 42-year-old man presented with a polypoid endobronchial mass of the right apical segmental bronchus. Bronchial brushing smears contained clusters of cells exhibiting abundant diffusely granular cytoplasm with indistinct borders. A cytologic diagnosis of granular-cell tumor was rendered. Histologic examination of the upper right lobectomy specimen provided confirmation. Immunohistochemically, the granular cells strongly reacted with the S-100 protein antibody. This case demonstrates that the cytologic diagnosis of bronchial granular-cell tumor is possible if this lesion is considered in the differential diagnosis of lung tumors.  相似文献

The variations in the maximal specific growth rate of the psychrotrophic bacterium Pseudomonas fluorescens MF0 with respect to temperature were studied between 0 and 30 degrees C (optimal for growth). The Arrhenius plot showed a drastic change in slope at the intermediate temperature of 17 degrees C. Over the cold domain from 0 to 17 degrees C, the temperature characteristic was twofold higher than over the suboptimal domain from 17 to 30 degrees C. The macromolecular composition of exponentially growing cells was invariant over the entire range from 0 to 30 degrees C. Variations of temperature and growth rate were independently investigated through chemostat experiments in order to characterize their respective effects on cell macromolecular composition and size. The effect of growth rate in this psychrotrophic strain is identical to that of all other bacteria assayed so far. In contrast, an original biphasic variation of total protein concentration was demonstrated in strain MF0 with respect to temperature, with a maximum at 17 to 20 degrees C. Indeed, increasing the temperature in the chemostat resulted in a biphasic decrease in the net protein production rate: a very slight decrease below 17 degrees C and a much larger decrease from 17 to 28 degrees C. These results could signify an increase in the cellular protein degradation rate with increasing temperature, especially above 17 degrees C.  相似文献

Dimers of aspartokinase I/homoserine dehydrogenase I from Escherichia coli K 12 have been isolated under very mild conditions. The dimers which cannot be distinguished from the tetramers by their kinetic properties, reassociate in the presence of potassium ions or L-aspartate. The selective sensitivity of aspartokinase I/homoserine dehydrogenase I to mild proteolytic digestion of dimers has been used to probe the reassociation reaction under the conditions of aspartokinase assay. We demonstrate that rapid reassociation occurs and that the protein species present in the assay when dimers are used to test the activity is tetrameric. These results confirm the previously proposed model for the subunit association of aspartokinase I/homoserine dehydrogenase I.  相似文献

The cellulolytic rumen bacteriumRuminococcus flavefaciens 17 was found to produce multiple xylanases ranging in apparent molecular weight from 55 to 200 kDa. A 55 kDa xylanase showed constitutive synthesis, but formation of the larger enzymes was increased in cultures grown with avicel, straw, or xylan, compared with cellobiose, as the energy source. At least six xylanases were detected in cultures grown with oat straw or oat xylan. Polyclonal antibodies were raised against the amino (A) or carboxy terminal (C) domains of the bifunctional XYNA product of the clonedR. flavefaciens xynA gene. Both antibody preparations recognized several xylanases larger than 80 kDa fromR. flavefaciens cells grown with avicel, straw, or xylan, indicating the production of multiple, antigenically related enzymes during growth on these substrates. Neither antibody preparation recognized the constitutive 55-kDa xylanase.  相似文献

1. One force-fed meal containing labelled 14C-astaxanthin (14C-Ax) and 3H-canthaxanthin (3H-Cx) or 3H-zeaxanthin (3H-Zx) was given to eight mature female rainbow trout. Ninety-six hours after the test meal ingestion, trout were killed and liver, skin, muscle and ovaries were dissected out. 2. Ax accumulated slightly more in muscle than Cx but in all tissues Ax and Cx were very significantly more concentrated than Zx. 3. 3H-Zx metabolites were found only in the liver, whereas 14C-phoenicoxanthin was the only metabolic pigment from 14C-Ax detected and was found in all investigated tissues. 4. 3H-Ax was found in the liver of all trout indicating that 3H-Cx and 3H-Zx were Ax precursors, and that salmonids probably possess carotenoid oxidative pathways unknown until now. 5. Labelled retinol1 and retinol2 were detected only in the liver and 3H-Zx was largely the predominant precursor of these two vitamin A forms.  相似文献

The reference strains of Enterococcus seriolicida (ATCC 49156T) (T = type strain) and of Lactococcus garvieae (ATCC 43921T) and 30 field strains of Gram-positive cocci isolated from diseased rainbow trout in Italy were found to be phenotypically (API 20 STREPT and API 50 CH) and genetically (DNA-DNA hybridization) similar. The high DNA-DNA homologies (70–100%) and the low ΔTm(e) (less than 1.1°C) among these strains showed that Enterococcus seriolicida and Lactococcus garvieae are synonyms, describing a single bacterial species. E. seriolicida strains should be classified as L. garvieae, which must be considered as a major pathogen of freshwater and salt water fish with a world-wide distribution.  相似文献

  1. Dispersal, defined as the movement of individuals among local communities in a landscape, is a central regional determinant of metacommunity dynamics in ecosystems. Whereas both natural and anthropogenic ecosystem fragmentations can limit dispersal, previous attempts to measure such limitations have faced considerable context dependency, due to a combination of spatial extent and associated environmental variability, the wide range of dispersal modes, and abilities of organisms or variation in network topologies. Therefore, the role dispersal plays compared to local environmental filtering in explaining metacommunity dynamics remains unclear in fragmented dendritic ecosystems.
  2. We quantified α- and β-diversity components of invertebrate metacommunities across 10 fragmented headwater stream networks and tested the hypothesis that dispersal is the primary determinant of biodiversity organisation in these dynamic and spatially constrained ecosystems.
  3. Alpha-diversity was much lower in intermittent than perennial reaches, even long after rewetting, indicating an overwhelming effect of drying including a legacy effect on local communities.
  4. Beta-diversity was never correlated with environmental distances but predominantly explained by spatial distances accounting for river network fragmentation. The nestedness proportion of β-diversity was considerable and reflected compositional differences where communities from intermittent reaches were subsets of perennial reaches.
  5. Altogether, these results indicate dispersal as the primary process shaping metacommunity dynamics in these 10 headwater stream networks, where local communities recurrently undergo extinction and recolonisation events. This challenges previous conceptual views that local environment filtering is the main driver of headwater stream metacommunities.
  6. As river networks become increasingly fragmented due to global change, our results suggest that some freshwater ecosystems currently driven by local environment filtering could gradually become dispersal-limited. In this perspective, shifts from perennial to intermittent flow regimes represent ecological thresholds that should not be crossed to avoid jeopardising river biodiversity, functional integrity, and the ecosystem services they provide to society.
