As key pollinators, honey bees are crucial to many natural and agricultural ecosystems. An important factor in the health of honey bees is the availability of diverse floral resources. However, in many parts of the world, high-intensity agriculture could result in a reduction in honey bee forage. Previous studies have investigated how the landscape surrounding honey bee hives affects some aspects of honey bee health, but to our knowledge there have been no investigations of the effects of intensively cultivated landscapes on indicators of individual bee health such as nutritional physiology and pathogen loads. Furthermore, agricultural landscapes in different regions vary greatly in forage and land management, indicating a need for additional information on the relationship between honey bee health and landscape cultivation. Here, we add to this growing body of information by investigating differences in nutritional physiology between honey bees kept in areas of comparatively low and high cultivation in an area generally high agricultural intensity in the Midwestern United States. We focused on bees collected directly before winter, because overwintering stress poses one of the most serious problems for honey bees in temperate climates. We found that honey bees kept in areas of lower cultivation exhibited higher lipid levels than those kept in areas of high cultivation, but this effect was observed only in colonies that were free of Varroa mites. Furthermore, we found that the presence of mites was associated with lower lipid levels and higher titers of deformed wing virus (DWV), as well as a non-significant trend towards higher overwinter losses. Overall, these results show that mite infestation interacts with landscape, obscuring the effects of landscape alone and suggesting that the benefits of improved foraging landscape could be lost without adequate control of mite infestations.  相似文献

The population structure of bottlenose dolphins, Tursiops truncatus, along the U.S. Atlantic coast has recently been redefined from one homogenous population into five coastal stocks. Local studies indicate even finer structure, primarily based on isolation of dolphins inhabiting estuaries. We identified population structuring of non‐estuarine coastal bottlenose dolphins during a study in New Jersey, the northern range along the Atlantic Coast. Using photo‐identification and distribution survey results, an analysis identified two major clusters of individuals significantly separated by five variables (distance from shoreline, group size, occurrence of the barnacle Xenobalanus globicipitis, avoidance behavior, and individual coloration). Sightings assigned to cluster 1 occurred in nearshore shallow waters (0–1.9 km, x?= 3.5 m), and those assigned to cluster 2 occurred further offshore in deeper waters (1.9–6 km, x?= 9.5 m). Only eight of 194 individuals (4%) were identified in both regions. Collectively, this suggests an occurrence of two stocks that are spatially, physically, and behaviorally distinguishable over a small distance. These results indicate that complexity in Tursiops population structure is not limited to latitudinal gradients or barriers created by estuarine habitats, but also by partitioning of habitat as a function of distance from shore and depth over small distances.  相似文献

This study investigated the potential of the conserved intron scanning approach to develop molecular markers for genes involved in lignin biosynthesis among members of the Saccharum complex and a distantly related Imperata species. Five intron-flanking primer sets targeting genes encoding five lignin biosynthetic enzymes??phenylalanine ammonia lyase (PAL), 4-coumarate coenzyme A ligase (4-CL), caffeoyl-CoA 3-O-methyltransferase (CCoAOMT), cinnamoyl-CoA reductase (CCR) and peroxidase (POX)??were designed based on the sequence analysis between sugarcane expressed sequence tags (ESTs) and Sorghum bicolor orthologs. Nucleotide sequence analyses of the amplicons of PAL, 4-CL and CCoAOMT orthologs revealed the presence of single nucleotide polymorphisms (SNPs) and insertions?Cdeletions (INDELs) in the target introns as well as (CT)-simple sequence repeats (SSRs) in CCoAOMT orthologs. The SSR marker screening against fifty-nine accessions of the Saccharum complex and an Imperata species confirmed that the identified SSR markers were highly polymorphic among Saccharum and Erianthus species. PCR-restriction fragment length polymorphism (PCR?CRFLP) and cleaved amplified polymorphic sequence (CAPS) marker screening of 4-CL and CCoAOMT orthologs developed genus-specific molecular markers that confirmed the intergeneric hybridization status of Saccharum Fiji hybrids. The current study showed that the conserved intron scanning strategy is applicable to multiple copy genes of polyploid monocots. The conserved intron scanning approach provides a novel way of investigating DNA polymorphisms among species within the Saccharum complex and has the potential to help in the development of marker-assisted selection in intergeneric hybrids.  相似文献

Focal laser ablation destroys cancerous cells via thermal destruction of tissue by a laser. Heat is absorbed, causing thermal necrosis of the target region. It combines the aggressive benefits of radiation treatment (destroying cancer cells) without the harmful side effects (due to its precise localization). MRI is typically used pre-treatment to determine the targeted area, and post-treatment to determine efficacy by detecting necrotic tissue, or tumor recurrence. However, no system exists to quantitatively evaluate the post-treatment effects on the morphology and structure via MRI. To quantify these changes, the pre- and post-treatment MR images must first be spatially aligned. The goal is to quantify (a) laser-induced shape-based changes, and (b) changes in MRI parameters post-treatment. The shape-based changes may be correlated with treatment efficacy, and the quantitative effects of laser treatment over time is currently poorly understood. This work attempts to model changes in gland morphology following laser treatment due to (1) patient alignment, (2) changes due to surrounding organs such as the bladder and rectum, and (3) changes due to the treatment itself. To isolate the treatment-induced shape-based changes, the changes from (1) and (2) are first modeled and removed using a finite element model (FEM). A FEM models the physical properties of tissue. The use of a physical biomechanical model is important since a stated goal of this work is to determine the physical shape-based changes to the prostate from the treatment, and therefore only physical real deformations are to be allowed. A second FEM is then used to isolate the physical, shape-based, treatment-induced changes. We applied and evaluated our model in capturing the laser induced changes to the prostate morphology on eight patients with 3.0 Tesla, T2-weighted MRI, acquired approximately six months following treatment. Our results suggest the laser treatment causes a decrease in prostate volume, which appears to manifest predominantly at the site of ablation. After spatially aligning the images, changes to MRI intensity values are clearly visible at the site of ablation. Our results suggest that our new methodology is able to capture and quantify the degree of laser-induced changes to the prostate. The quantitative measurements reflecting of the deformation changes can be used to track treatment response over time.  相似文献

A 11kD glycopeptide has been isolated by pepsin digestion of carcinoembryonic antigen (CEA) that is rapidly endocytosed by isolated rat Kupffer cells and lung alveolar macrophages. Uptake of this glycopeptide by the isolated cells can be inhibited by excess unmodified CEA. Removal of the N-linked oligosaccharide chains by N-glycanase did not alter cellular uptake but reduced the MW to approximately 5500. A seventeen amino acid N-terminal sequence locates this peptide at the junction of the N-terminal and first loop domain of CEA. It is suggested that the recognition of a peptide sequence in this area of CEA is responsible for its clearance from the circulation.  相似文献

The effect of temperature on the distributions of ectothermic vertebrates is well documented. Despite the increase of 6°C expected in the next 60 years in South America, numerous vertebrates are still considered as ‘Least Concern’ species by the IUCN due to their large distribution, insufficient widespread threats and insignificant population decline. One example is the lizard Tropidurus torquatus (Squamata: Tropiduridae), commonly found thermoregulating in anthropic environments throughout the Brazilian Cerrado, but restricted to gallery forests in the equator‐ward localities. The urban areas in this warmer region have been colonised by other closely related congeners (e.g. Tropidurus oreadicus). This study aimed to understand this divergence of habitat selection by these tropirudids that may explain some of the species responses to past and future climate warming. We collected body temperatures (Tb), micro‐environmental temperatures (Ta) and operative (Te) temperatures in four sites along a latitudinal gradient: a pole‐ward and two central sites where T. torquatus inhabit urban areas and one equator‐ward site where T. torquatus and T. oreadicus occur in the gallery forest and in urban microhabitats, respectively. All three populations of T. torquatus present similar Tb (35.5–36°C) and shared microhabitats with a similar Ta (34–37.3°C). The Te in the equator‐ward urban site was considerably higher than in the gallery forest. Tropidurus oreadicus Tb was 38.2 °C (30.1–41.3°C) and was active at a Ta of 30.5–42.3°C. The overlap between the genus Tb, Ta and Te highlights a decrease in the hours of activity that lizards would experience under climate warming. The reduction of hours of activity together with the devastation of natural habitats represents threats and an alarming scenario especially for the equator‐ward populations.  相似文献

For seven homologous compounds, the highest electro-antennography responses from male Diloba caeruleocephala (L.) were elicited by ( Z )-5-decenyl acetate ( Z )-7-dodecenyl acetate and ( Z )-8-tridecenyl acetate. The ( Z )-8-tridecenyl acetate caused significant attraction of males of this species in the field, in both Bulgaria and Hungary. ( Z )-10-pentadecenyl acetate, when added as a 10% admixture to the latter, acted as an attractant inhibitor. In Bulgaria, the flight period of this species, as established by means of pheromone traps, commenced at the beginning of September and lasted until the middle of October.  相似文献

Several solvent-producing clostridia, including Clostridium acetobutylicum and C. beijerinckii, were previously shown to be nitrogen-fixing organisms based on the incorporation of 15N2 into cellular material. The key nitrogen-fixation (nif) genes, including nifH, nifD, and nifK for nitrogenase component proteins as well as nifE, nifN, nifB and nifV for synthesis of the iron–molybdenum cofactor (FeMoco) of nitrogenase, have now been identified in C. acetobutylicum or C. beijerinckii or both. The organization of these genes is similar to the distinctive pattern that was first observed in Clostridium pasteurianum, with the nifN and nifB genes fused into the nifN-B gene and with the nifV gene split into the nifVω and nifVα genes. The corresponding nif genes of these three clostridial species are highly related to each other. However, in the two solvent-producing clostridia, the nifH and nifD genes are interspersed by two glnB-like genes, which are absent in the corresponding region in C. pasteurianum. However, the nifN-B and nifVω genes of C. pasteurianum are interspersed by the putative modA and modB genes (for molybdate transport), which are absent in the corresponding region in C. acetobutylicum. C. acetobutylicum and C. beijerinckii grew well under nitrogen-fixing conditions, and the acetylene-reducing activity of nitrogenase was measured in the two species. Acetone, butanol, and isopropanol production occurred in nitrogen-fixing cultures, but the peak of nitrogen-fixing activity preceded the active solventogenic phase.
