Brazil is one of the most important endemic areas for leishmaniasis worldwide. Protected areas that are tourist attractions likely present an important risk of transmission of cutaneous leishmaniasis (CL). Furthermore, with the geographical expansion of visceral leishmaniasis (VL), several studies have recorded the occurrence of its vector, Lutzomyia longipalpis, and cases of human and canine VL in such tourist areas. The Parque Estadual do Sumidouro is an environmentally protected area located in the Brazilian Cerrado biome and in an important area endemic for leishmaniasis in the state of Minas Gerais. The purpose of this study was to monitor the sand fly fauna in areas of tourist activity in the park. Sampling was performed every month, from September 2011 to August 2013, using CDC light traps at six sites of differing environmental characteristics. Sampled specimens were identified following Galati (2003), and females were submitted to molecular techniques for the detection and identification of Leishmania DNA. A total of 4,675 sand fly specimens of 25 species belonging to nine genera were collected. The most abundant species were Micropygomyia quinquefer, Lutzomyia renei and Pintomyia pessoai, although only Pi. pessoai is implicated in the transmission of Leishmania braziliensis. The species accumulation curve reached saturation on the 16th sampling event. Species richness, diversity and evenness differed among the sampled areas. The seasonal curve was not determined by a single unique species, and no single species was the most abundant in all environments sampled. The main vector of Leishmania (Leishmania) infantum, Lutzomyia longipalpis, accounted for only 5.35% of the specimens collected. Proven or suspected vectors of Leishmania (Viannia) braziliensis were recorded, and one female of the cortellezzii complex tested positive for Le. braziliensis DNA. Even with a low infection rate (0.62%), these data indicate the circulation of the parasite and reinforce the need for entomological and epidemiological surveillance in the park and its surroundings. 相似文献

Rapid, phasic dopamine (DA) release in the mammalian brain plays a critical role in reward processing, reinforcement learning, and motivational control. Fast scan cyclic voltammetry (FSCV) is an electrochemical technique with high spatial and temporal (sub-second) resolution that has been utilized to examine phasic DA release in several types of preparations. In vitro experiments in single-cells and brain slices and in vivo experiments in anesthetized rodents have been used to identify mechanisms that mediate dopamine release and uptake under normal conditions and in disease models. Over the last 20 years, in vivo FSCV experiments in awake, freely moving rodents have also provided insight of dopaminergic mechanisms in reward processing and reward learning. One major advantage of the awake, freely moving preparation is the ability to examine rapid DA fluctuations that are time-locked to specific behavioral events or to reward or cue presentation. However, one limitation of combined behavior and voltammetry experiments is the difficulty of dissociating DA effects that are specific to primary rewarding or aversive stimuli from co-occurring DA fluctuations that mediate reward-directed or other motor behaviors. Here, we describe a combined method using in vivo FSCV and intra-oral infusion in an awake rat to directly investigate DA responses to oral tastants. In these experiments, oral tastants are infused directly to the palate of the rat – bypassing reward-directed behavior and voluntary drinking behavior – allowing for direct examination of DA responses to tastant stimuli. 相似文献

Consequences of reef phase shifts on fish communities remain poorly understood. Studies on the causes, effects and consequences of phase shifts on reef fish communities have only been considered for coral-to-macroalgae shifts. Therefore, there is a large information gap regarding the consequences of novel phase shifts and how these kinds of phase shifts impact on fish assemblages. This study aimed to compare the fish assemblages on reefs under normal conditions (relatively high cover of corals) to those which have shifted to a dominance of the zoantharian Palythoa cf. variabilis on coral reefs in Todos os Santos Bay (TSB), Brazilian eastern coast. We examined eight reefs, where we estimated cover of corals and P. cf. variabilis and coral reef fish richness, abundance and body size. Fish richness differed significantly between normal reefs (48 species) and phase-shift reefs (38 species), a 20% reduction in species. However there was no difference in fish abundance between normal and phase shift reefs. One fish species, Chaetodon striatus, was significantly less abundant on normal reefs. The differences in fish assemblages between different reef phases was due to differences in trophic groups of fish; on normal reefs carnivorous fishes were more abundant, while on phase shift reefs mobile invertivores dominated. 相似文献

Kappaphycus alvarezii is a seaweed of great economic importance for the extraction of kappa carrageenan from its cell walls. The most common strains are dark red, brown, yellow, and different gradations of green.
It is known that ultraviolet radiation (UVR) affects macroalgae in many important ways, including reduced growth rate, reduction
of primary productivity, and changes in cell biology and ultrastructure. Therefore, we examined the brown strain of K. alvarezii exposed to ultraviolet-B radiaton (UVBR) for 3 h per day during 28 days of cultivation. The control plants showed growth
rates of 7.27% d−1, while plants exposed to UVBR grew only 4.0% d−1. Significant differences in growth rates and in phycobiliproteins between control and exposed plants were also found. Compared
with control plants, phycobiliprotein contents were observed to decrease after UV-B exposure. Furthermore, the chlorophyll
a (Chl a) contents decreased and showed significant differences. UVBR also caused changes in the ultrastructure of cortical and subcortical
cells, which included increased thickness of the cell wall and number of plastoglobuli, reduced intracellular spaces, changes
in the cell contour, and destruction of chloroplast internal organization. Reaction with Toluidine Blue showed an increase
in the thickness of the cell wall, and Periodic Acid-Schiff stain showed a decrease in the number of starch grains. By the
significant changes in growth rates, photosynthetic contents and ultrastructual changes observed, it is clear that UVBR negatively
affects intertidal macroalgae and, by extension, their economic viability. 相似文献

Iron accumulation has been suggested to contribute to an increase of the susceptibility to mycobacterial infections. In this study we tested the effect of an array of iron chelating ligands of the 3-hydroxy-4-pyridinone family, in the intramacrophagic growth of Mycobacterium avium. We found that bidentate chelators, namely N-alkyl-3-hydroxy-4-pyridinones and N-aryl-3-hydroxy-4-pyridinones, did not affect the growth of M. avium inside mouse macrophages. In the case of the hexadentate chelators, those synthesized using an alkylamine (CP262) or a benzene ring (CP252) to link the three bidentate units, did not have an inhibitory effect on intramacrophagic growth of M. avium while those synthesized from a tripodal structure to anchor the bidentate units were capable of inhibiting the intramacrophagic growth of M. avium. The molecule we designated CP777 had the strongest inhibitory activity. The growth-reducing activity of CP777 was abrogated when this molecule was saturated with iron. These results confirm that iron deprivation, by the use of iron chelating compounds, restricts M. avium growth and that new iron chelators offer an approach to controlling mycobacterial infections. 相似文献

The Ni(II) and Zn(II) derivatives of Desulfovibrio vulgaris rubredoxin (DvRd) have been studied by NMR spectroscopy to probe the structure at the metal centre. The βCH2 proton pairs from the cysteines that bind the Ni(II) atom have been identified using 1D nuclear Overhauser enhancement (NOE)
difference spectra and sequence specifically assigned via NOE correlations to neighbouring protons and by comparison with
the published X-ray crystal structure of a Ni(II) derivative of Clostridium pasteurianum rubredoxin. The solution structures of DvRd(Zn) and DvRd(Ni) have been determined and the paramagnetic form refined using
pseudocontact shifts. The determination of the magnetic susceptibility anisotropy tensor allowed the contact and pseudocontact
contributions to the observed chemical shifts to be obtained. Analysis of the pseudocontact and contact chemical shifts of
the cysteine Hβ protons and backbone protons close to the metal centre allowed conclusions to be drawn as to the geometry
and hydrogen-bonding pattern at the metal binding site. The importance of NH–S hydrogen bonds at the metal centre for the
delocalization of electron spin density is confirmed for rubredoxins and can be extrapolated to metal centres in Cu proteins:
amicyanin, plastocyanin, stellacyanin, azurin and pseudoazurin. 相似文献

Food sharing with immatures is an important and relatively well studied aspect of infant care in many cooperative species.
A key point that has not yet been fully addressed, however, is how increasing the difficulty of obtaining food influences
the willingness of breeders and helpers to provision immature offspring. We used captive golden headed lion tamarins (Leontopithecus chrysomelas) to examine how breeders and helpers differ in provisioning juvenile individuals according to the level of difficulty of
obtaining food. The level of difficulty in obtaining food was varied by placing the food inside tubes that allowed access
only by adults. When food acquisition became more difficult, food sharing with juveniles and breeding females increased significantly.
Begging calls by breeding females and juveniles increased during the experimental condition, which probably led to increases
in food sharing. Breeders and helpers did not differ in their contribution to provisioning when food was easily available,
nor did they differ in their contribution when food was difficult to obtain. Breeding males in callitrichids have a prominent
role in transferring food to offspring, but contrary to our expectations, they did not increase food transfer in the experimental
condition. An unexpected result was the increased investment of the breeding female into her current offspring when the level
of difficulty of obtaining food was higher. We suggest that breeding lion tamarin females are not as constrained by reproductive
costs as breeding females of other callitrichids. Degree of reproductive skew is hypothesized as a factor affecting the contribution
of breeders and helpers to offspring care in cooperative breeding mammals, though we suggest that more studies are needed

How best to summarize large and complex datasets is a problem that arises in many areas of science. We approach it from the point of view of seeking data summaries that minimize the average squared error of the posterior distribution for a parameter of interest under approximate Bayesian computation (ABC). In ABC, simulation under the model replaces computation of the likelihood, which is convenient for many complex models. Simulated and observed datasets are usually compared using summary statistics, typically in practice chosen on the basis of the investigator's intuition and established practice in the field. We propose two algorithms for automated choice of efficient data summaries. Firstly, we motivate minimisation of the estimated entropy of the posterior approximation as a heuristic for the selection of summary statistics. Secondly, we propose a two-stage procedure: the minimum-entropy algorithm is used to identify simulated datasets close to that observed, and these are each successively regarded as observed datasets for which the mean root integrated squared error of the ABC posterior approximation is minimized over sets of summary statistics. In a simulation study, we both singly and jointly inferred the scaled mutation and recombination parameters from a population sample of DNA sequences. The computationally-fast minimum entropy algorithm showed a modest improvement over existing methods while our two-stage procedure showed substantial and highly-significant further improvement for both univariate and bivariate inferences. We found that the optimal set of summary statistics was highly dataset specific, suggesting that more generally there may be no globally-optimal choice, which argues for a new selection for each dataset even if the model and target of inference are unchanged. 相似文献
