In view of the development of al-carnitine deficiency, the metabolism ofl-carnitine and structure-related trimethylammonium compounds was studied inSalmonella typhimurium LT2 by means of thin-layer chromatography (TLC).l-Carnitine, crotonobetaine and acetyl-l-carnitine stimulated the anaerobic growth in a complex medium significantly. The stimulation depended on the formation of -butyrobetaine. The reduction ofl-carnitine proceeded in two steps: (1) Dehydration of thel-carnitine to crotonobetaine, (2) hydrogenation of crotonobetaine to -butyrobetaine. The reduction of crotonobetaine was responsible for the growth stimulation. Terminal electron acceptors of the anaerobic respiration such as nitrate and trimethylamine N-oxide, but not fumarate, suppressed the catabolism ofl-carnitine completely. Glucose fermentation, too, inhibited the reduction ofl-carnitine but optimal growth with a high carnitine catabolism was achieved byd-ribose. The esters of carnitine with medium- and long-chain fatty acids inhibited the growth considerably because of their detergent properties.Abbreviations TLC thin-layer chromatography  相似文献

The efflux of 20 amino acids, induced by either high K+ concentration or veratrine, was determined in pigeon tectal slices. Ca2+-dependent, K+-induced release of beta-alanine, gamma-aminobutyric acid (GABA), and glutamate was observed. Veratrine caused release of the same amino acids plus glycine in a tetrodotoxin-sensitive manner. beta-Alanine had a strong inhibitory effect on the activity of tectal neurons which was blocked by strychnine but not by bicuculline. The results indicated a transmitter function for beta-alanine in the optic tectum, and were consistent with the previously proposed transmitter role of GABA and glutamate in this structure.  相似文献

α-Satellite DNA from African green monkey cells was analysed with restriction nucleases in some detail confirming and complementing our earlier results. With EcoRI and HaeIII (or BsuRI isoschizomer), about 25 and 10%, respectively, of the satellite DNA were cleaved into a series of fragments of the 172 bp repeat length and multiples thereof. To allow studies with fragments of homogeneous sequence unit length, HindIII fragments were covalently joined with the plasmid pBR313. After transformation 19 clones were obtained, containing up to three monomer fragments. Nine of the clones were characterized by digestion with EcoRI. Three of these had cleavage sites for this nuclease in the satellite DNA portion. In the six clones tested with HaeIII no cleavage site was detected in the cloned DNA. The results are discussed in relation to the nucleotide sequence data recently published by Rosenberg et al. (1978) and in the context of random and nonrandom processes in satellite DNA evolution.  相似文献

Summary Serum AFP concentrations in normal adult BALB/c/J and in normal adult C3H/He mice were in the order of 0.6 g/ml and 0.1 g/ml, respectively. In BALB mice, AFP was localized in the cytoplasm of differentiated mono- and binucleated hepatocytes in centrolobular and intermediate zones of normal adult liver. No cellular AFP could be detected in liver sections of normal adult C3H mice.CCl4 intoxication was accompanied by increase of serum AFP levels. A maximum was reached on day 4. Afterwards, concentrations declined. In sera of BALB/c/J mice, AFP levels reached values 10-fold higher and more than in sera of C3H/He mice.From day one after CCl4 intoxication, cellular AFP was detected in hepatocytes of portal and periportal areas including intermediate zones adjacent to the necrosis. The intensity of AFP staining reached a maximum between the days 3 and 4. Hepatocytes in front of the necrotic areas usually contained the strongest AFP reactions. In both mouse strains, cellular AFP pattern was comparable, but strongest immunoreactivity was observed in liver sections of BALB/c/J mice.Liver injury and subsequent regeneration occurred to the same extent in both studied strains. The much higher serum AFP levels and the stronger AFP immunolocalizations in BALB mice were thought not due to increased numbers of AFP producing and releasing cells during liver regeneration. Additional mechanisms must play a role in increased AFP synthesis per single cell. The application of rotating frame spin-lattice relaxtion to the determination of the intramolecular motions in peptides is discussed, and results are presented on the application of 13C T1p to peptide microdynamics in solution. The effective molecular rotational reorientation times at the amide and amino nitrogens may be derived from appropriate data onT1p of the carbons adjacent to them. We also show by theoretical caculations that 1H and 13C T1p experiments of suitable 2H and 15N substituted peptides will allow intramolecular main- and sidechain motions, characterized by times in the range 10?1–10?6 sec, to be investigated.  相似文献
