Background

Serum type II secretory phospholipase A2 (sPLA2-IIa) has been found to be predictive of adverse outcomes in patients with stable coronary heart disease. Compounds targeting sPLA2-IIa are already under development. This study investigated if an association of sPLA2-IIa with secondary cardiovascular disease (CVD) events may be of causal nature or mainly a matter of confounding by correlated cardiovascular risk markers.

Methodology/Principal Findings

Eight-year follow-up data of a prospective cohort study (KAROLA) of patients who underwent in-patient rehabilitation after an acute cardiovascular event were analysed. Associations of polymorphisms (SNP) in the sPLA2-IIa-coding gene PLA2G2A with serum sPLA2-IIa and secondary fatal or non-fatal CVD events were examined by multiple regression. Hazard ratios (HR) were compared with those expected if the association between sPLA2-IIa and CVD were causal. The strongest determinants of sPLA2-IIa (rs4744 and rs10732279) were associated with an increase of serum concentrations by 81% and 73% per variant allele. HRs (95% confidence intervals) estimating the associations of the SNPs with secondary CVD events were increased, but not statistically significant (1.16 [0.89–1.51] and 1.18 [0.91–1.52] per variant allele, respectively). However, these estimates were very similar to those expected when assuming causality (1.18 and 1.17), based on an association of natural log-transformed sPLA2-IIa concentration with secondary events with HR = 1.33 per unit.

Conclusion

The present findings regarding genetic polymorphisms, determination of serum sPLA2-IIa, and prognosis in CVD patients are consistent with a genuine causal relationship and thus might point to a valid drug target for prevention of secondary CVD events.  相似文献

Glycine decarboxylase, or P-protein, is a pyridoxal 5′-phosphate (PLP)-dependent enzyme in one-carbon metabolism of all organisms, in the glycine and serine catabolism of vertebrates, and in the photorespiratory pathway of oxygenic phototrophs. P-protein from the cyanobacterium Synechocystis sp. PCC 6803 is an α2 homodimer with high homology to eukaryotic P-proteins. The crystal structure of the apoenzyme shows the C terminus locked in a closed conformation by a disulfide bond between Cys972 in the C terminus and Cys353 located in the active site. The presence of the disulfide bridge isolates the active site from solvent and hinders the binding of PLP and glycine in the active site. Variants produced by substitution of Cys972 and Cys353 by Ser using site-directed mutagenesis have distinctly lower specific activities, supporting the crucial role of these highly conserved redox-sensitive amino acid residues for P-protein activity. Reduction of the 353–972 disulfide releases the C terminus and allows access to the active site. PLP and the substrate glycine bind in the active site of this reduced enzyme and appear to cause further conformational changes involving a flexible surface loop. The observation of the disulfide bond that acts to stabilize the closed form suggests a molecular mechanism for the redox-dependent activation of glycine decarboxylase observed earlier.  相似文献

Empirical data on the signals and processes that direct animal movement during dispersal in heterogeneous landscapes are scarce. Our understanding could benefit from utilising simulation approaches and searching for simple rules across species and landscapes. This study sought to identify general movement behaviours that optimise butterfly movements during hilltopping. This widespread dispersal‐like phenomenon in butterflies, where males and virgin females ascend to mountain summits for the purpose of mating, benefits from the uniqueness of knowing the purpose (mating) and the orientation signal (topography). We used an individual‐based simulation model to search for movement rules that can optimise mating success, mating time, and the success of mated females in subsequently finding habitat patches across differently structured landscapes. We found that a strong response to topography was optimal for males and virgin females to reach summits, but slight deviation from ‘perfect’, namely a certain level of randomness in response to topography, was inherently essential to avoid the risk of being trapped on local summits. The optimal response of mated females deviated only slightly from a random movement, indicating a potential weak response to topography which could be easily overlooked by field studies. Notably, the parameter values identified by the model as optimal corresponded with the observed behaviour of butterflies in the field. Finally, the optimal movement behaviours were affected by the lifespan of butterflies and the spatial distribution of host plant patches, but less so by landscape structure. We therefore suggest that modelling approaches that build on simple biological rules can facilitate the development and parameterisation of models for understanding and potentially predicting dispersal and connectivity in complex landscapes, also in circumstances where observed patterns seem complex and empirical data are scarce.  相似文献

The understanding of airborne pollen transportation is crucial for the reconstruction of the paleoenvironment. Under favorable conditions, a considerable amount of long-distance-transported pollen can be deposited far from its place of origin. In extreme arid regions, in most cases, such situations occur and increase the difficulty to interpret fossil pollen records. In this study, three sets of Cour airborne pollen trap were installed on the northern slope of Tianshan Mountains to collect airborne Picea schrenkiana (spruce) pollen grains from July 2001 to July 2006. The results indicate that Picea pollen disperses extensively and transports widely in the lower atmosphere far away from spruce forest. The airborne Picea pollen dispersal period is mainly concentrated between mid-May and July. In desert area, weekly Picea pollen began to increase and peaked suddenly in concentration. Also, annual pollen indices do not decline even when the distance increased was probably related to the strong wind may pick up the deposited pollen grains from the topsoil into the air stream, leading to an increase of pollen concentration in the air that is irrelevant to the normal and natural course of pollen transport and deposition. This, in turn, may lead to erroneous interpretations of the pollen data in the arid region. This study provided insight into the shift in the Picea pollen season regarding climate change in arid areas. It is recorded that the pollen pollination period starts earlier and the duration became longer. The results also showed that the temperature of May and June was positively correlated with the Picea pollen production. Furthermore, the transport of airborne Picea pollen data is useful for interpreting fossil pollen records from extreme arid regions.  相似文献

Single-molecule force spectroscopy (SMFS) using the atomic force microscope (AFM) has emerged as an important tool for probing biomolecular interaction and exploring the forces, dynamics, and energy landscapes that underlie function and specificity of molecular interaction. These studies require attaching biomolecules on solid supports and AFM tips to measure unbinding forces between individual binding partners. Herein we describe efficient and robust protocols for probing RNA interaction by AFM and show their value on two well-known RNA regulators, the Rev-responsive element (RRE) from the HIV-1 genome and an adenine-sensing riboswitch. The results show the great potential of AFM–SMFS in the investigation of RNA molecular interactions, which will contribute to the development of bionanodevices sensing single RNA molecules.  相似文献

Background

Experimental autoimmune neuritis (EAN) is a well-known animal model of human demyelinating polyneuropathies and is characterized by inflammation and demyelination in the peripheral nervous system. Fascin is an evolutionarily highly conserved cytoskeletal protein of 55 kDa containing two actin binding domains that cross-link filamentous actin to hexagonal bundles.

Methods

Here we have studied by immunohistochemistry the spatiotemporal accumulation of Fascin?+?cells in sciatic nerves of EAN rats.

Results

A robust accumulation of Fascin?+?cell was observed in the peripheral nervous system of EAN which was correlated with the severity of neurological signs in EAN.

Conclusion

Our results suggest a pathological role of Fascin in EAN.

Background

Tungiasis, a parasitic skin disease caused by the female sand flea Tunga penetrans, is a prevalent condition in impoverished communities in the tropics. In this setting, the ectoparasitosis is associated with important morbidity. It causes disfigurement and mutilation of the feet. Feasible and effective treatment is not available. So far prevention is the only means to control tungiasis-associated morbidity.

Methodology

In two villages in Central Madagascar, we assessed the efficacy of the availability of closed shoes and the twice-daily application of a plant-based repellent active against sand fleas (Zanzarin) in comparison to a control group without intervention. The study population was randomized into three groups: shoe group, repellent group and control group and monitored for ten weeks. The intensity of infestation, the attack rate and the severity of tungiasis-associated morbidity were assessed every two weeks.

Findings

In the repellent group, the median attack rate became zero already after two weeks. The intensity of the infestation decreased constantly during the observation period and tungiasis-associated morbidity was lowered to an insignificant level. In the shoe group, only a marginal decrease in the intensity of infestation and in the attack rate was observed. At week 10, the intensity of infestation, the attack rate and the severity score for acute tungiasis remained significantly higher in the shoe group than in the repellent group. Per protocol analysis showed that the protective effect of shoes was closely related to the regularity with which shoes were worn.

Conclusions

Although shoes were requested by the villagers and wearing shoes was encouraged by the investigators at the beginning of the study, the availability of shoes only marginally influenced the attack rate of female sand fleas. The twice-daily application of a plant-based repellent active against sand fleas reduced the attack to zero and lowered tungiasis-associated morbidity to an insignificant level.  相似文献

Increasing capacity utilization and lowering manufacturing costs are critical for pharmaceutical companies to improve their competitiveness in a challenging environment. Development of next generation cell lines, improved media formulations, application of mature technologies and innovative operational strategies have been deployed to improve yields and capacity utilization. This article describes a large‐scale perfusion strategy for the N‐1 seed train bioreactor that was successfully applied to achieve higher inoculation cell densities in the production culture. The N‐1 perfusion at 3,000‐L scale, utilizing a inclined settler, achieved cell densities of up to 158 × 105 cell mL?1 at perfusion rates of 2950 L day?1 and a retention efficiency of >85%. This approach increased inoculation cell densities and decreased cultivation times by ~20% in a CHO‐based, fed‐batch antibody manufacturing process while providing comparable culture performance, productivity, and product quality. The strategy therefore yielded significant increase in capacity utilization and concomitant cost improvement in a large scale cGMP facility.
