This paper describes the rational design, generation and testing of a molecularly imprinted polymer specific for Ibuprofen. Ibuprofen is a member of the class of drugs termed non-steroidal anti-inflammatory drugs (NSAIDS). In the present study, Ibuprofen was used as a template molecule for the preparation of molecularly imprinted polymers. A MIP has been produced which is capable of recognising Ibuprofen in aqueous media. Furthermore, Ibuprofen can be selectively extracted from aqueous conditions by molecularly imprinted solid phase extraction (MISPE). Recoveries were typically high (>80%) and good selectivity for Ibuprofen over structurally related analogues was seen. Moreover, the nature of the recognition between MIP and template has been investigated by NMR and molecular modelling to analyse whether or not it is possible to predict how well a given MIP will perform under set conditions. In addition, the physical characteristics of the MIP have been investigated including the particle size distribution on exposure of the MIP to different solvents. This has been related to the ability of the MIP to rebind Ibuprofen under the same conditions. The data from the characterisation of the MIP has been used to further enhance the understanding of the nature of MIP recognition. 相似文献

We compared the performances of two different methodologies for collecting orchid bees (Hymenoptera: Apidae): active sampling (at chemical baits with hand nets) versus passive sampling (with baited traps). Data were retrieved from previous field studies in which both methodologies were employed simultaneously. We evaluated the effectiveness of each method in terms of the number of species collected and the composition of the bee species through paired t test and Permutational Multivariate Analysis of Variance, respectively. Active sampling collected significantly more bee species than passive sampling in all but one study. The composition of bee species collected by each of the two methods was also significantly different. Larger bees belonging to Eulaema and Eufriesea species tended to be more frequent in traps. In contrast, all Euglossa species presented comparatively higher relative frequencies with active sampling. We challenge previous assertions that both methodologies are equally effective and argue that insisting on the sole use of baited traps in orchid-bee inventories may result in retrieving strongly biased raw data which will eventually lead to incorrect conclusions about the actual composition of orchid-bee assemblages. Since orchid bees have been more and more used as key organisms in studies on environmental impact in the Neotropics, particularly in Brazil, and the passive sampling methodology has been adopted as the “official” one by the Brazilian environmental agency (IBAMA), we suggest a re-evaluation of this practice and the adoption of the more expensive, but more reliable, methodology of using well-trained field researchers to actively collect data on these important Neotropical pollinators. 相似文献

HtrA1 is a secreted multidomain protein with serine protease activity. In light of increasing evidence implicating this protein in the regulation of skeletal development and pathology, we investigated the role of HtrA1 in osteoblast mineralization and identified domains essential for this activity. We demonstrate increased HtrA1 expression in differentiating 2T3 osteoblasts prior to the appearance of mineralization. HtrA1 is subsequently down-regulated in fully mineralized cultures. The functional role of HtrA1 in matrix calcification was investigated using three complementary approaches. First, we transfected a full-length HtrA1 expression plasmid into 2T3 cells and showed that overexpression of HtrA1 delayed mineralization, reduced expression of Cbfa1 and collagen type I mRNA, and prevented BMP-2-induced mineralization. Second, knocking down HtrA1 expression using short interfering RNA induced mineral deposition by 2T3 cells. Third, by expressing a series of recombinant HtrA1 proteins, we demonstrated that the protease domain and the PDZ domain are essential for the inhibitory effect of HtrA1 on osteoblast mineralization. Finally, we tested whether HtrA1 cleaves specific matrix proteins that are known to regulate osteoblast differentiation, mineralization, and/or BMP-2 activity. Full-length recombinant HtrA1 cleaved recombinant decorin, fibronectin, and matrix Gla protein. Both the protease domain and the PDZ domain were necessary for the cleavage of matrix Gla protein, whereas the PDZ domain was not required for the cleavage of decorin or fibronectin. Type I collagen was not cleaved by recombinant HtrA1. These results suggest that HtrA1 may regulate matrix calcification via the inhibition of BMP-2 signaling, modulating osteoblast gene expression, and/or via the degradation of specific matrix proteins. 相似文献

The critically endangered mangrove finch is now limited to one small population on the west coast of Isabela Island in the Galápagos, but 100 years ago multiple populations were found on the islands of Isabela and Fernandina. By accessing genetic datasets through museum sampling, we are able to put current levels of genetic diversity and hybridization with congenerics into a historical context for enhanced conservation. In this study, we compared neutral genetic diversity of the now extinct Fernandina population to historical and current diversity of the Isabela population using 14 microsatellite markers. We found that current genetic diversity of the last remnant population (~80–100 individuals) is far below levels 100 years ago, with only about half of the allelic diversity retained. Current genetic diversity is close to levels in the Fernandina population that went extinct by the 1970s. Bottleneck analysis did not show a strong signature of recent decline, and instead implies that this species may have consistently had low population sizes with wide fluctuations. Hybridization with congeneric woodpecker finches was found in the modern Isabela population, implying that some individuals within the few remaining breeding pairs are finding mates with woodpecker finches. Within the context of historical low population sizes and wide fluctuations, current conservation efforts may help the mangrove finch face current extinction threats and avoid the fate of the Fernandina population. However, this historically small lineage will likely continue to face challenges associated with small specialist species surrounded by a widely-distributed sister lineage producing viable hybrids. 相似文献

Anthropogenic disturbances often affect the abundance and diversity of ants (Hymenoptera: Formicidae) but relatively few studies have explored the implications of such changes on the ecosystem services mediated by these insects. Here, we evaluated how the transformation of Cerrado savanna habitats into crop plantations affects the abundance, diversity, and the predatory activity of ants. A survey of the ant faunas foraging above‐ and belowground was performed in six crop and six non‐crop (i.e., native vegetation) sites. Above‐ and belowground rates of ant predation were estimated at these same sites using mealworms, Tenebrio molitor L. (Coleoptera: Tenebrionidae), as baits, simulating crop herbivores. Belowground predation rates were significantly greater in the non‐crop sites, despite the lack of difference in overall abundance and species richness of ants foraging belowground between the crop vs. non‐crop sites. In contrast, we did not detect any significant difference in aboveground predation rates between crop vs. non‐crop sites even though there were significantly more species of ants foraging aboveground in the non‐crop sites. Army ants (subfamily Dorylinae) were the main predatory species belowground, and their abundance was significantly greater in non‐crop sites. In contrast, the main predators aboveground were omnivore ants of the genera Pheidole and Solenopsis, which had similar abundances in the crop and non‐crop sites. Overall, our results indicate that transformation of native Cerrado habitats into crop plantations reduces the abundance of some important predatory species, notably those that forage belowground, and this may negatively affect the potential for ants to provide pest control services in agroecosystems. 相似文献

The seeds of most tropical plants are dispersed by animals, many of which also act as seed predators. Shifts in animal community composition, such as those driven by the clearing of native vegetation, are therefore likely to drive changes in plant recruitment. We used manipulative experiments excluding ants, small rodents, and birds to quantify the relative impacts of these granivores on animal-dispersed pioneer trees (ADPT) in fragments of savanna vegetation and adjacent soy plantations in Brazil’s Cerrado. We found that ants were the main consumers of ADPT seeds, that the rates of seed removal varied with seed size, and that removal rates were higher in savanna fragments than in soy plantations. However, we also found significant interactions between habitat type, seed species, and the type of seed predator being excluded. Our results underscore how challenging it can be to predict the influence of human disturbances on the interactions between plant and animal communities. Because ants, rodents, and birds are Cerrado’s the main seed dispersers and granivores, seedling recruitment in Cerrado landscape mosaics will depend on how these distinct but related processes are each influenced by species-specific patterns of seed size and seed abundance. 相似文献

Leafcutter ants propagate co‐evolving fungi for food. The nearly 50 species of leafcutter ants (Atta, Acromyrmex) range from Argentina to the United States, with the greatest species diversity in southern South America. We elucidate the biogeography of fungi cultivated by leafcutter ants using DNA sequence and microsatellite‐marker analyses of 474 cultivars collected across the leafcutter range. Fungal cultivars belong to two clades (Clade‐A and Clade‐B). The dominant and widespread Clade‐A cultivars form three genotype clusters, with their relative prevalence corresponding to southern South America, northern South America, Central and North America. Admixture between Clade‐A populations supports genetic exchange within a single species, Leucocoprinus gongylophorus. Some leafcutter species that cut grass as fungicultural substrate are specialized to cultivate Clade‐B fungi, whereas leafcutters preferring dicot plants appear specialized on Clade‐A fungi. Cultivar sharing between sympatric leafcutter species occurs frequently such that cultivars of Atta are not distinct from those of Acromyrmex. Leafcutters specialized on Clade‐B fungi occur only in South America. Diversity of Clade‐A fungi is greatest in South America, but minimal in Central and North America. Maximum cultivar diversity in South America is predicted by the Kusnezov–Fowler hypothesis that leafcutter ants originated in subtropical South America and only dicot‐specialized leafcutter ants migrated out of South America, but the cultivar diversity becomes also compatible with a recently proposed hypothesis of a Central American origin by postulating that leafcutter ants acquired novel cultivars many times from other nonleafcutter fungus‐growing ants during their migrations from Central America across South America. A sensitive and specific competitive enzyme immunoassay (EIA) for rat growth hormone was developed using reagents from the National Institutes of Arthritis, Diabetes, Digestive Diseases and Kidney, Bethesda, Md. In this assay soluble growth hormone and growth hormone adsorbed to a solid-phase support compete for monkey anti-growth hormone antibody binding sites. The immobilized goat anti-monkey immunoglobin G covalently conjugated to horse radish peroxides. Therefore a high concentration of soluble growth hormone in the sample will result in low absorbance detection from the colored products of the enzyme reaction. Assay parameters were optimized by investigating the concentration of reagents and the reaction kinetics in each of the assay steps. The assay can be performed in 27 hours. A sensitivity range of 0.19 ng to 25 ng in the region of 10 to 90% binding was obtained. Near 50% binding (3 ng) the intraassay coefficient of variation (CV) was 5.54% and the interassay CV was 5.33%. The correlation coefficient (r2) between radioimmunoassay and EIA was 0.956 and followed the curve Y=0.78X + 1.9. Selected applications were described as follows. Alkaline extracts of pituitary tissue increase 2 fold in GH content after mercaptoethanol treatment. Alkaline extracts of pituitary tissue chromatographed on HPLC molecular sieving columns showed s molecular weight. Fractions representing a molecular weight > 200kD were enhanced 6 fold. Fractions whose molecular weight range was 22kD to 50 kD were enhanced 2 fold. This assay provides a reliable alternative to RIA and offers the major advantage of eliminating radioactive reagents and counting equipment. 相似文献
