Bacillus stearothermophilus strains PV 72 and ATCC 12980 carry a crystalline surface layer (S-layer) with hexagonal (p6) and oblique (p2) symmetry, respectively. Sites of insertions of new subunits into the regular lattice during cell growth have been determined by the indirect fluorescent antibody technique and the protein A/colloidal gold technique.During S-layer growth on both bacillus strains the following common features were noted: 1. shedding of intact S-layer or turnover of individual subunits was not seen; 2. new S-layer was deposited in helically-arranged bands over the cylindrical surface of the cell at a pitch angle related to the orientation of the lattice vectors of the crystalline array; 3. little or no S-layer was inserted into pre-existing S-layer at the poles, and 4. septal regions and, subsequently, newly formed cell poles were covered with new S-layer protein.  相似文献

The influence of human T lymphocytes expressing the enzyme dipeptidyl peptidase IV (DPP IV) was investigated with respect to human peripheral B-lymphocyte differentiation. B cells stimulated with pokeweed mitogen in the presence of DPP IV-positive T cells produced high amounts of immunoglobulin. Moderate amounts of immunoglobulin could be measured when B cells were cultured in the presence of DPP IV-negative T cells. DPP IV defines a T-cell subset partially overlapping the subsets characterized by the differentiation antigens Leu 3a (helper/inducer) and Leu 2a (suppressor/cytotoxic). DPP IV-positive T cells exert, in contrast to DPP IV-negative T cells, high interleukin-2 activity after stimulation with phytohemagglutinin and pokeweed mitogen. To further functionally characterize DPP IV-positive and DPP IV-negative T cells, the helper effects of Leu 3a-positive T-cell subsets, differing in DPP IV expression, were investigated in pokeweed mitogen- and Staphylococcus aureus-driven B-cell differentiation systems. After pokeweed mitogen stimulation, immunoglobulin production was markedly reduced when B cells were cultured in the presence of Leu 3a-positive T cells expressing DPP IV (DPP IV+/Leu 3a+). In contrast, high amounts of immunoglobulin were produced in cultures with Leu 3a-positive but DPP IV-negative T cells (DPP IV-/Leu 3a+). This difference in immunoglobulin production of B cells cultured with DPP IV+/Leu 3a+ and DPP IV-/Leu 3a+ T cells could not be observed in Staphylococcus aureus-stimulated cultures. Here, both T-cell subsets supported terminal differentiation of B cells. We conclude that in the pokeweed mitogen-driven culture systems, DPP IV+/Leu 3a+ and DPP IV-/Leu 3a+ T cells may differ in the production of growth and/or differentiation factors distinct from interleukin-2.  相似文献

The effect of phorbol myristate acetate (PMA) on T4 (CD4) expression by monocytoid cells was studied. Greater than 99% of untreated U937 and HL-60 cells expressed surface T4 as measured with a fluorescence-activated cell sorter. The percentage of T4 positive cells decreased to less than 20% after incubation with PMA (10(-8) M). A decrease was observed within 15 min of PMA exposure, was maximal within 1 hr, and persisted for at least 3 days in the continuous presence of PMA. The susceptibility of untreated and PMA-treated U937 cells to human immunodeficiency virus (HIV) was also studied. Pretreatment of cells with PMA for 18 hr decreased the production of viral RNA and p24 antigen 24 hr after infection. The dose of PMA resulted in a parallel reduction of both T4 expression and infection by HIV. When PMA was washed from cultures and replaced with fresh medium for 48 hr, then T4 expression and the production viral RNA and p24 antigen following infection were restored. These data suggest that pharmacologic manipulation of surface T4 expression may have a potential role in the prevention or treatment of HIV infection.  相似文献

Summary Using cultured trophoblast cells obtained by chorionic villus biopsy, we diagnosed Fanconi anemia (FA) in two pregnancies and excluded it in eight pregnancies at risk for the syndrome. Baseline chromosomal breakage and breakage induced by diepoxybutane (DEB) were analyzed. Increased breakage was used as a marker for the syndrome. Our results were unambiguous and provide a reliable method for prenatal detection of FA in the first trimester of pregnancy.  相似文献

Fluorescein-labeled muscle actin was microinjected into Amoeba proteus and followed during intracellular redistribution by means of the image-intensification technique. The fully polymerization-competent protein becomes part of the endogenous actomyosin system undergoing dynamic changes over time periods of several hours. Single-frame analysis of long-term sequences enabled the direct demonstration of both the contractile activities and morphological transformations of microfilaments in normally locomoting, immobilized and phagocytozing specimens. In normally locomoting cells the filament layer undergoes continuous changes in spatial distribution depending on the actual pattern of cytoplasmic streaming and cell shape. The highest degree of differentiation is always maintained in the intermediate region between the front and the uroid, thus indicating this segment of the cortex to be the most important site in generating motive force for pseudopodium formation and ameboid movement. In immobilized cells contracted by the application of ruthenium red or relaxed by different anesthetics, the filament layer forms a continuous thick sheath beneath the cell surface or becomes completely disintegrated. In phagocytozing cells the local polymerization of actin at the tip of pseudopodia forming the food-cup and around the nascent phagosome points to a significant participation of the actomyosin system in the process of capturing and constricting prey organisms. Although our results provide clear evidence for the overall importance of motive force generation according to the hydraulic pressure theory, some motile phenomena exist in Amoeba proteus that cannot exclusively be explained by this mechanism.  相似文献

Special attention is paid to the mechanism of bacterial reinfection and how it is influenced by deviations in can construction or can handling procedures. The changes in activities of glyoxysomal and peroxisomal enzymes have been correlated with the fine structure of microbodies in cotyledons of the cucumber (Cucumis sativus L.) during the transition from fat degradation to photosynthesis in light-grown plants, and in plants grown in the dark and then exposed to light. During early periods of development in the light (days 2 through 4), the microbodies (glyoxysomes) are interspersed among lipid bodies and contain relatively high activities of glyoxylate cycle enzymes involved in lipid degradation. Thereafter, these activities decrease rapidly as the cotyledons expand and become photosynthetic, and the activity of glycolate oxidase rises to a peak (day 7); concomitantly the microbodies (peroxisomes) become preferentially associated with chloroplasts.  相似文献

Zusammenfassung Die Muskelwand des Rattenoesophagus besteht ausschließlich aus quergestreiften Muskelfasern. Diese weisen keine Strukturunterschiede gegenüber gewöhnlichen Skeletmuskeln der Ratte auf. Sie sind in zwei spiralig gegenläufigen Muskelschichten angeordnet. Zwischen den beiden Muskelschichten befindet sich ein mit ChE-Darstellungsmethoden anfärbbarer Plexus myentericus, der an seinen Kreuzungspunkten regelmäßig Ganglienzellen aufweist. 10% der Ganglienzellen sind stark ChE-positiv, es handelt sich um tiberwiegend spezifische AChE.Die Rr. oesophagei des N. vagus zeigen sowohl marklose als auch myelinisierte Axone. Die Endäste, die den Plexus verlassen, um an motorischen Endplatten zu endigen, sind marklos.Die motorischen Endplatten weisen muldenförmige Einsenkungen des Sarkolemms auf. Die subsynaptische Membran ist reichlich in Palten gelegt, auch dort, wo ihr kein terminales Axon aufliegt. Es wurden 2 Arten von terminalen Axonen an den Endplatten festgestellt: 1.Dicke Axone, die mit vielen synaptischen Bläschen angefüllt sind, daneben aber auch einige granulierte Bläschen enthalten. 2. Dünne, fast ausschließlich mit granulierten Bläschen erfüllte Axone. Oft berühren die terminalen Axone einander unmittelbar, bisweilen unter Bildung von Membranverdickungen.
Structure and innervation of the striated muscle fibres of the esophagus of the rat
Summary The muscular wall of the esophagus of the rat consists exclusively of striated muscle fibers which do not differ from the skeletal muscles of the same animal. These fibres are arranged in two layers running spirally in contrary directions. Between these two layers a ChE-positive myenteric plexus occurs, containing several ganglionic cells at its crossing points. 10 per cent of these ganglionic cells are strongly AChE-positive. The tiny branches of the vagus nerve, passing through the outer muscular layer into the myenteric plexus consist of bundles of unmyelinated as well as myelinated fibres. The terminal branches of the plexus leading to motor endplates are unmyelinated.The motor endplates are situated in trough-shaped impressions of the surface of muscle fibres. The subsynaptic sarcolemma is richly folded, even in places where there is no overlying synaptic terminal.There are two kinds of nerve terminals in the motor endplates: (1) Thick terminals abundantly filled with clear synaptic vesicles but also containing a few dense core vesicles. (2) Thin axons containing predominantly granulated vesicles.The synaptic nerve terminals often come into contact with each other, occasionally forming membrane thickenings at such places.
