Leaves on tomato plants infected with Fusarium oxysporum f. lycopersici frequently wilt unilaterally when the vascular bundles supplying the affected leaflets are diseased. However, when the vascular bundles on one side of healthy petioles are severed by notching the petiole base, the entire leaf remains turgid. Leaflets on the notched side receive water by diffusion between bundles at the petiole tip. Lateral translocation of water out of individual vessels and between bundles in diseased xylem is impaired by the impregnation of vessel walls, intercellular spaces, and cells adjacent to vessels with the products of vascular discoloration. Waterproofing of vascular bundles can be induced in excised healthy leaves by culture filtrates of the pathogen and catechol. Waterproofing of vessels may play an important role in vascular dysfunction by confining water to individual vessels and thereby increasing the importance of vessel occlusions. 相似文献

Reaction of radicals in the presence of O2, and singlet oxygen, with some amino acids, peptides, and proteins yields hydroperoxides. These species are key intermediates in chain reactions and protein damage. Previously we have shown that peptide and protein hydroperoxides react rapidly with thiols, and that this can result in inactivation of thiol-dependent enzymes. The major route for the cellular removal of damaged proteins is via catabolism mediated by proteosomal and lysosomal pathways; cysteine proteases (cathepsins) play a key role in the latter system. We hypothesized that inactivation of cysteine proteases by hydroperoxide-containing oxidised proteins may contribute to the accumulation of modified proteins within cells. We show here that thiol-dependent cathepsins, either isolated or in cell lysates, are rapidly and efficiently inactivated by amino acid, peptide, and protein hydroperoxides in a time- and concentration-dependent manner; this occurs with similar efficacy to equimolar H2O2. Inactivation involves reaction of the hydroperoxide with Cys residues as evidenced by thiol loss and formation of sulfenic acid intermediates. Structurally related, non-thiol-dependent cathepsins are less readily inactivated by these hydroperoxides. This inhibition, by oxidized proteins, of the system designed to remove modified proteins, may contribute to the accumulation of damaged proteins in cells subject to oxidative stress. 相似文献

A series of branched and unbranched anilinohexafluoroisopropanols related to the known sulfonamide T0901317 were prepared and evaluated as activators/modulators of both LXRalpha and LXRbeta. A structure-activity relationship was established and compounds with high potency on both the receptors were identified. Many compounds showed a tendency toward selectivity for LXRbeta versus LXRalpha. Several analogues were evaluated for effects on plasma lipoprotein levels in mice. A few of these significantly raised HDL-cholesterol levels in plasma but showed markedly different effects on liver triglyceride content, suggesting that this series may yield candidates with improved efficacy/safety profiles compared to existing molecules. 相似文献

Among the cells of the inner ear, the outer hair cells (OHCs) are the most important targets of noise-induced effects, being
the most sensitive cell types. The aim of this study was to examine the effects of noise (50 Hz-20 kHz, 80 dB sound pressure
level, 14 days) on intracellular calcium levels and on the expression pattern of purinoceptors in the membrane of the OHCs
of the guinea pig and to measure the stiffness changes of the lateral membrane of these cells. In noise-exposed animals, the
resting intracellular calcium concentration increased compared to nontreated animals and was slightly higher in the cells
of the basal (219 ± 29 nM) than in the apical (181 ± 24 nM) turns of the cochlea. After application of 180 μM adenosine triphosphate, the intracellular calcium level rose by 60 ± 22 nM in cells from the apical and by 44 ± 10 nM in cells from the basal turns, significantly less than in nontreated animals. Expression of the P2X1, P2X2, P2X4, P2X7, P2Y1 and P2Y4 receptor subtypes was suppressed, while expression of the P2Y2 subtype did not decrease in either of the two preparations. In parallel with the increase in intracellular calcium concentration,
the stiffness of the lateral wall of the OHCs was increased. Noise-induced changes in intracellular calcium homeostasis and
subsequently in the calcium-dependent regulatory mechanisms may modify OHC lateral wall stiffness and may lead to reduction

Predation management is an important component of managing species of conservation concern. The piping plover (Charadrius melodus; plover), a disturbance-dependent and conservation-reliant shorebird that nests on sandy beaches and barrier islands on the Atlantic Coast, was listed under the United States Endangered Species Act in 1986, with habitat loss and predation stated as key causes of its decline. We evaluated the relationship between a suite of predators and plover chick survival from 2015–2018. We used a camera grid to establish indices of the abundance of 3 known chick predators: red fox (Vulpes vulpes), raccoon (Procyon lotor), and domestic cat. We used camera detections in a survival model to assess potential relationships between predator species detection and plover chick survival. Plover chick survival was negatively related with red fox detection, but not with detection of the other 2 predators. In addition to the correlation with red fox detections, chick survival was negatively related to high plover nesting density. Our results suggest that red foxes were predators of piping plover chicks during our study, likely augmented by other density-dependent sources of mortality. Targeted predator management could aid in conservation of piping plovers in this system as a short-term solution, but long-term recovery plans must also address habitat limitation. 相似文献
