Gastrointestinal (GI) parasitic infection is the main health constraint for small ruminant production, causing loss of weight and/or death. Red Maasai sheep have adapted to a tropical environment where extreme parasite exposure is a constant, especially with highly pathogenic Haemonchus contortus. This breed has been reported to be resistant to gastrointestinal parasite infection, hence it is considered an invaluable resource to study associations between host genetics and resistance. The aim of this study was to identify polymorphisms strongly associated with host resistance in a double backcross population derived from Red Maasai and Dorper sheep using a SNP-based GWAS analysis. The animals that were genotyped represented the most resistant and susceptible individuals based on the tails of phenotypic distribution (10% each) for average faecal egg counts (AVFEC). AVFEC, packed cell volume (AVPCV), and live weight (AVLWT) were adjusted for fixed effects and co-variables, and an association analysis was run using EMMAX. Revised significance levels were calculated using 100,000 permutation tests. The top five significant SNP markers with - log10 p-values >3.794 were observed on five different chromosomes for AVFEC, and BLUPPf90/PostGSf90 results confirmed EMMAX significant regions for this trait. One of these regions included a cluster of significant SNP on chromosome (Chr) 6 not in linkage disequilibrium to each other. This genomic location contains annotated genes involved in cytokine signalling, haemostasis and mucus biosynthesis. Only one association detected on Chr 7 was significant for both AVPCV and AVLWT. The results generated here reveal candidate immune variants for genes involved in differential response to infection and provide additional SNP marker information that has potential to aid selection of resistance to gastrointestinal parasites in sheep of a similar genetic background to the double backcross population.  相似文献

Lactobacillus salivarius cells contain an inducible transport system for folate. Influx via this system is time- and temperature-dependent, requires glucose and glutamine for optimum activity, and is half-maximal at folate concentrations in the nanomolar range. The folate internalized after 30 min at 30 degrees C is not released from the cells by excess extracellular folate and is recovered in cell extracts primarily in metabolized forms. A membrane-associated folate-binding protein is also present in cells that have been induced to transport folate. This binding protein constitutes 1% of total cellular protein, exhibits a high affinity for folate (KD = 0.40 nM), and requires divalent cations for optimum binding activity. Folate binds rapidly to this protein, while the exchange of bound substrate with folate added subsequently is relatively slow and dependent on the metabolic state of the cell. The transport rate per binding site is 0.05/min at 30 degrees C. A comparison of substrate specificity showed that folate binding and transport are both inhibited to the same extent by several different folate compounds, and a parallel irreversible inhibition of both processes is observed by prior treatment of the cells with a carbodiimide-activated derivative of folic acid. Binding protein labeled covalently with [3H]folate and solubilized with Triton X-100 was purified by a fractionation procedure involving absorption and elution from microgranular silica and molecular sieve chromatography. The isolated protein appeared homogeneous by gel electrophoresis and had an apparent molecular weight of 21,000. Monoclonal antibodies to the folate transport protein of Lactobacillus casei showed a high degree of cross-reactivity to the isolated binding protein from L. salivarius, indicating that these proteins share common epitopes. These results suggest that folate uptake by L. salivarius proceeds via an abundant membrane-associated binding protein which facilitates the movement of folate across the membrane as an electroneutral complex with cations. The substrate then slowly dissociates from internalized binding sites and is metabolized sequentially to coenzyme forms and then to membrane-impermeable folylpolyglutamates.  相似文献

Summary Physical, nutritional and defensive qualities of seeds differ in the extent to which they influence granivore preference. In a study aimed to quantifying those differences, Ord's kangaroo rats (Dipodomys ordii) were found to prefer the seeds of just three of twenty-nine species: Cryptantha crassisepala, Oryzopsis hymenoides and Salsola kali. Oryzopsis hymenoides was most preferred during the early plant growth season (April–July); preference for S. kali peaked during late (August–November) and dormant (December–March) seasons; and greatest preference for C. crassisepala occurred during dormant and early seasons. Regression of forage ratios, averaged across seasons, against seed length, mass, abundance, patchiness, percent nitrogen, energy content, and chemical defenses showed seed length to be the most important predictor of seed preference. Seed length combined with nitrogen (protein) content and levels of two defensive compounds, saponins and non-protein amino acids, to account for 68% of the variation in seed preference. The importance of seed length rather than biomass indicated that there are limits to the ability of D. ordii to detect small seeds and that small size facilitated escape of dispersed seeds. Seasonality in preference suggested, however, that seed escape was encountered by predispersal harvesting of newly maturing seeds still on plants. Maximization of protein intake contradicted previously published observations, but presumably reflected low nitrogen availability. In addition to small size, the presence of saponins or non-protein amino acids in seeds was sufficient to negate the positive influence of higher protein content.  相似文献

1. Mitochondrial alpha-glycerophosphate dehydrogenase (GPD) activity was assayed in liver homogenates from active and torpid ground squirrels. 2. Arrhenius plots of GPD activity were linear in non-hibernating animals and discontinuous in hibernators. Compared with non-hibernators, the energy of activation in hibernators was reduced between 37 and 25 degrees C, but increased between 25 and 6 degrees C. 3. A dose-response relation between GPD activity and injected thyroxine was determined in active animals. No correlation was found between enzyme activity at 37 or 6 degrees C and circulating titres of thyroid hormones, in ground squirrels sampled during the preparative and hibernating phases.  相似文献
