A novel method for immobilization of yeast cells in alginate gels of various shapes by internal liberation of Ca-ions

Summary In situ gelation of alginate, in which Ca-ions are liberated internally in the gel, was used for immobilization of yeast cells. Compared to the traditional alginate gelation method, internal gelling gave immobilized yeast particles of higher strength, without reduction in fermentation rate.     

Properties of action potentials carried by divalent cations in identified leech neurons

Properties of divalent cation potentials carried by either Sr2+ or Ca2+ ions in Na+-free, TEA-Ringer solution were characterized in identified neurons of two species of leeches (Macrobdella and Haementeria). In Macrobdella, the overshoot of the potentials varied logarithmically with [Sr2+]0 (28.5 mV per 10-fold change). The overshoot, Vmax, and duration of the potentials increased with increasing divalent cation concentration and saturated at about 20 to 30 mM [Sr2+]0. The Vmax, amplitude, and duration of the potentials were reversibly blocked by Co2+ and Mn2+. The block by Mn2+ could be well-fitted by a reverse Langmuir-curve with an apparent KI of 100 micromolar. The local anesthetic procaine also reversibly inhibited the Vmax and duration of the potentials. The inhibition was greater at alkaline pH suggesting that procaine blocks the calcium channel from inside the membrane. The identified leech neurons examined in Macrobdella varied considerably in their ability to sustain somatic divalent cation potentials. Stimulation of T cells and most motoneurons produced no or only weak potentials, whereas stimulation of Retzius, N, Nut, and AP cells evoked overshooting potentials of several seconds' duration. Stimulation of the ALG cell of Haementeria in normal Ringer solution evoked a slowly-rising, purely Ca2+-dependent potential of approximately 100 ms duration. This response was TTX-resistant, unaffected by complete removal of Na+ from the Ringer solution, and abolished by 1 mM Mn2+. The overshoot varied logarithmically with a slope of 28 mV/decade change in [Ca2+]0.     

Monoclonal antibodies to Rhizobium meliloti and surface mutants insensitive to them.

Monoclonal antibodies were produced to the surface of the symbiotic nitrogen-fixing bacterium Rhizobium meliloti. Bacterial lysis in the presence of complement or cycles of agglutination and growth were used to select mutants no longer recognized by the antibodies. The mutants were used to produce new antibodies with different specificities. Several mutants had altered sensitivity to one or more bacteriophages. R. meliloti strains from different sources had distinct patterns of sensitivity to monoclonal antibodies and phages, which together can be used for discriminative typing.     

Responses of aerial ventilation to hypoxia and hypercapnia inChanna argus,an air-breathing fish

Summary The snake-head fish (Channa argus) is an obligate air-breather inhabiting fresh waters in the temperate zone of East Asia.Ventilation of the air-breathing organ and aerial gas exchange were measured in 1 to 2 kg specimens at 15 and 25°C. Additionally, the ventilatory responses to hypoxia and hypercapnia were studied. Aerial ventilation increased from 1.1 to 2.9 mlbtps·kg^–1·min^–1 when temperature rose from 15 to 25°C. Concomitantly, O₂-uptake through airbreathing increased from 0.1 mlstpd·kg^–1·min^–1 (15°C) to 0.28 mlstpd·kg^–1·min^–1 (25°C), whereas aerial gas exchange was less important for CO₂-climination as evident from low gas exchange ratios (0.16 at 15°C, 0.29 at 25°C).Ventilation increases only slightly in response to inspiration of hypercapnic gas mixtures or to hypoxic conditions in water. By contrast, inspiration of hypoxic gas mixtures caused marked increases of ventilation in particular at the higher temperature.Aerial ventilation inChanna is low compared to values for ectothermic pulmonary breathers. However, its ventilatory responses to hypoxia strikingly resemble those of reptiles: The most marked ventilatory response to hypoxia occurs at the higher temperature where the demands for O₂ are greatest.     

Differential activation of the mouse beta-globin promoter by enhancers.

A series of plasmids was constructed to study the effect of two enhancers, the simian virus 40 72-base-pair repeat and the Harvey sarcoma virus 73-base-pair repeat, on the mouse beta maj-globin promoter. These plasmids contain the mouse beta maj-globin promoter linked to the Escherichia coli galK gene, thus allowing galactokinase enzyme activity to be used as a measure of promoter function. In CV-1 (primate) cells, it was found that an enhancer is required for optimal promoter activity and that the simian virus 40 (primate) enhancer increases galactokinase fourfold more than the Harvey sarcoma virus (mouse) enhancer. In L (mouse) cells, however, the Harvey sarcoma virus enhancer is 1.3-fold stronger than the simian virus 40 enhancer. These data support the hypothesis that enhancer activity can be species specific. Furthermore, when both enhancers are present on the same plasmid, their effect is additive on the beta-globin promoter whether the plasmid is in CV-1 cells or L cells.     

Structural organization of the normal and anoxic heart of Scyllium stellare

Summary The general and ultrastructural organization of the heart of the elasmobranch, Scyllium stellare, was studied in normal and in anoxic animals. The rich coronary supply was revealed three-dimensionally by the use of corrosion casts, showing a thebesian system of coronary arterioles and capillaries in the thin, outer compact layer as well as in the predominant, inner spongy layer of trabeculae.Only the sinus venosus received a neuronal input of large bundles of granule-containing axons terminating at fenestrated regions of the endocardium and suggesting a neurohormonal function.A simple, tubular sarcoplasmic reticulum with flattened junctional cisternae was present in myocardial cells of 1–5 m diameter, which contained one or two bundles of myofibrils. The latter were closely apposed to the inner aspect of the plasmalemma. Mitochondria were located centrally in the cells, which were joined by unfolded desmosomes involving Z-band material.Long periods of anoxia were tolerated without loss of heart function, but at the expense of cytoplasmic glycogen. Lipid granules were abundant in all layers and chambers, notably in animals prepared in the summer. The lipid granules displayed a marked increase in electron density when the heart was incubated in a buffered oxalate solution prior to fixation. A glycogen-sparing effect of the lipids during anoxia was observed.

1. Chromogranin A has been isolated from the washed membrane fraction of highly purified chromaffin granules after solubilization in Triton X-100. A hydrated molecular weight of 2.9·10⁵ has been obtained for chromogranin A_M which in its electrophoretic mobility and molecular volume appears very similar to chromogranin A_I of the water-soluble fraction.
