BACKGROUND: Rat trachea display a differential topographical distribution of connective tissue mast cells (CTMC) and mucosal mast cells (MMC) that may imply regional differences in the release of allergic mediators such as tumor necrosis factor-alpha (TNF-alpha) and eicosanoids. AIM: To evaluate the role of CTMC and MMC for release of TNF-alpha and eicosanoids after allergenic challenge in distinct segments of rat trachea. MATERIALS AND METHODS: Proximal trachea (PT) and distal trachea (DT) from ovalbumin (OVA)-sensitized rats, treated or not with compound 48/80 (48/80) or dexamethasone, were incubated in culture medium. After OVA challenge, aliquots were collected to study release of TNF-alpha and eicosanoids. RESULTS: Release of TNF-alpha by PT upon OVA challenge peaked at 90 min and decayed at 6 and 24 h. Release from DT peaked at 30-90 min and decayed 6 and 24 h later. When CTMC were depleted with 48/80, OVA challenge exacerbated the TNF-alpha release by PT at all time intervals, while DT exacerbated TNF-alpha levels 6 and 24 h later only. Dexamethasone reduced TNF-alpha production after 90 min of OVA challenge in PT and at 3 and 6h in DT. OVA challenge increased prostaglandin D2) in DT and leukotriene B4 in both segments but did not modify prostaglandin E2 and leukotriene C4 release. CONCLUSION: OVA challenge induces TNF-alpha release from MMC, which is negatively regulated by CTMC. The profile of TNF-alpha and eicosanoids depends on the time after OVA challenge and of the tracheal segment considered.  相似文献

The goal of this study was to determine the baroreflex influence on systolic arterial pressure (SAP) and pulse interval (PI) variability in conscious mice. SAP and PI were measured in C57Bl/6J mice subjected to sinoaortic deafferentation (SAD, n = 21) or sham surgery (n = 20). Average SAP and PI did not differ in SAD or control mice. In contrast, SAP variance was enhanced (21 +/- 4 vs. 9.5 +/- 1 mmHg2) and PI variance reduced (8.8 +/- 2 vs. 26 +/- 6 ms2) in SAD vs. control mice. High-frequency (HF: 1-5 Hz) SAP variability quantified by spectral analysis was greater in SAD (8.5 +/- 2.0 mmHg2) compared with control (2.5 +/- 0.2 mmHg2) mice, whereas low-frequency (LF: 0.1-1 Hz) SAP variability did not differ between the groups. Conversely, LF PI variability was markedly reduced in SAD mice (0.5 +/- 0.1 vs. 10.8 +/- 3.4 ms2). LF oscillations in SAP and PI were coherent in control mice (coherence = 0.68 +/- 0.05), with changes in SAP leading changes in PI (phase = -1.41 +/- 0.06 radians), but were not coherent in SAD mice (coherence = 0.08 +/- 0.03). Blockade of parasympathetic drive with atropine decreased average PI, PI variance, and LF and HF PI variability in control (n = 10) but had no effect in SAD (n = 6) mice. In control mice, blockade of sympathetic cardiac receptors with propranolol increased average PI and decreased PI variance and LF PI variability (n = 6). In SAD mice, propranolol increased average PI (n = 6). In conclusion, baroreflex modulation of PI contributes to LF, but not HF PI variability, and is mediated by both sympathetic and parasympathetic drives in conscious mice.  相似文献

Roadkill studies are typically conducted without regard to the carcass permanency time on the highway, which may lead to underestimation of roadkill data, especially small animals that are quickly removed by scavengers. To evaluate the carcass duration time on the lane and its relation to the roadkill time event, we conducted an experiment comparing the removal rate of small carcasses between different stretches of a highway and stretches of dirt roads. The rates found in the study were considered high, with 89 % of carcasses being removed in the first 24 h and 66 % within 12 h. The removals were high for both road categories but reached their peaks at different periods of the day, being higher during the day on the highway and at night on dirt roads. We believe that removal on the highway is dictated by higher vehicular traffic and mainly by the action of scavenger birds during the day, while on dirt roads, it is due to the action of different opportunistic scavengers during the night.  相似文献

Young plants of two Brazilian Green Dwarf coconut ecotypes from contrasting regions in relation to climate (UGD, from a hot and humid climate and JGD, from a hot and dry climate) were submitted to three consecutive drying/recovery cycles, under greenhouse conditions to determine the photosynthetic limitations encountered during the recovery phase of water deficiency. Three hypotheses were tested: (1) non-stomatal factors contribute to the incomplete recovery of net photosynthesis rate (A), (2) photochemical impairment is an important component of the non-stomatal limitations to A and (3) the two coconut ecotypes respond differently to the drought recovery. In each drying/recovery cycle, irrigation was suppressed until A reached zero. Then the plants were rewatered and the recovery was accompanied for 8 days. After 4 days of rewatering, CO2 and light response curve parameters were used to discriminate the stomatal and non-stomatal factors contributing to the incomplete recovery of A. Upon rewatering for 4 days, the pre-dawn leaf water potential of the stressed plants increased from ?1.20 to ?0.15 MPa on average. Only small differences were detected in the intercellular to atmospheric CO2 concentration ratio but A did not recover completely to the control values. In both ecotypes, light and CO2-saturated A, carboxylation efficiency, maximum quantum yield of PSII and maximum electron transport rate were significantly lower in stressed plants in comparison with the well-watered ones. Relative stomatal limitation (LS) ranged from 16 to 20% without significant differences between treatments and between ecotypes. The relative mesophyll limitation (Lm) was significantly higher in JGD in cycle 1 but this changed after repeated drought stress, decreasing to values significantly lower than those in UGD in the other two cycles. The results showed that after 4 days of rewatering stressed plant stomatal limitations were comparable to that of well-watered control plants (~20%). However A did not recover completely after drought stress due to non-stomatal factors. Important differences between the ecotypes were identified. The ecotype JGD presented smaller Lm, faster recovery of the apparent quantum efficiency of CO2 assimilation, adjustment of some carbon balance components and signs of photosynthetic capacity acclimation after repeated drying/recovery cycles.  相似文献

Parasitism by isopods is a common phenomenon in marine fishes and may induce negative effects, especially in juvenile stages. The surf-zones of sandy beaches are an important nursery habitat for many fish species, including commercially important ones, and are structured by beach characteristics and human activities, which are suggested to influence the fishes’ susceptibility to parasitism. Here, we analyze the early infestation and prevalence of parasitism in surf zones fishes of 27 beaches in southeastern Brazil with different characteristics. Parasites were recorded in only seven of the beaches. A high infestation was found in a single site, the highly urbanized Itaguá beach, where the prevalence of parasitism on juveniles and early adults of the Atlantic anchoveta Cetengraulis edentulus was high (68.11%). Four species of parasitic isopods were found, all belonging to the Cymothoidae family, but Livoneca desterroensis was the most abundant (78.7%). No effects of parasitism were detected on the size, weight and physiological condition of fishes, but presence of parasites and prevalence of infestation were positively affected by urbanization. Overall, our results indicate that the surf-zones have low parasite infestation, yet, they suggest that anthropic effects such as urbanization may enhance the prevalence of isopod parasitism.

Because the regulation of vascular function involves complex mutual interactions between nitric oxide (NO) synthase (NOS) and cyclooxygenase (COX) products, we examined the contribution of NO and prostanoids derived from the COX pathway in modulating aortic baroreceptor resetting during an acute (30 min) increase in arterial pressure in anesthetized rats. Increase in pressure was induced either by administration of the nonselective NOS inhibitor N(G)-nitro-L-arginine methyl ester (L-NAME) or aortic coarctation (COA) with or without treatment with the COX inhibitor indomethacin (INDO) or the selective neuronal NOS inhibitor 1-(2-trifluoromethylphenyl)imidazole (TRIM). The activity of the aortic depressor nerve and arterial pressure were simultaneously recorded, and the degree of resetting was determined by the shift of the pressure-nerve activity curve using the ratio [delta systolic pressure at 50% of maximum baroreceptor activity/delta systolic pressure] x 100. The magnitude of pressure rise was similar in the different groups (59 +/- 6, 53 +/- 5, 53 +/- 5, 45 +/- 5, 49 +/- 3, and 41 +/- 3 mmHg for COA, L-NAME, INDO+COA, INDO+L-NAME, TRIM+COA, and TRIM+INDO+COA, respectively, P = 0.27). The degree of resetting that occurred with L-NAME or COA combined with treatment with TRIM was attenuated compared with COA alone (7 +/- 4, 5 +/- 2, and 31 +/- 6%, respectively, P = 0.04). INDO failed to influence baroreceptor resetting to higher pressure but prevented L-NAME- and TRIM-induced effects (20 +/- 7, 21 +/- 8, and 32 +/- 6% for INDO+COA, INDO+L-NAME, and INDO+TRIM+COA, respectively; P = 0.38). Baroreceptor gain was affected only by l-NAME. These findings indicate that NO, probably from neuronal origin, may exert stimulatory influence on the degree of rapid baroreceptor resetting to hypertension that involves COX-derived prostanoids.  相似文献

Human Papillomavirus infection is considered as the main etiological factor of cervical cancer (ICC), although, the role of host genetic factors in ICC susceptibility has been increasing. Immunological response is crucial for the prevention of viral associated diseases. Interleukin 1 receptor antagonist (IL-1RN) is considered to be an important regulator of host immunity and several studies have shown a potential role of a 86?bp VNTR polymorphism within intron 2 of the IL-1RN gene in host immune response variability. We investigated the role of this polymorphism in cervical cancer development in Portugal with a case–control study developed with peripheral blood samples from 196 healthy women and 340 women with cervical lesions from the Northern Region of Portugal. We observed that IL-1RN Allele 2 homozygosis was significantly higher in cases than in controls. In fact, IL-1RN A2*A2 homozygous revealed to be associated with an increased risk of HSIL?+?ICC (OR?=?1.90; 95?% IC 1.13–3.21; p?=?0.015). Furthermore, we also observed that median age of onset of HSIL?+?ICC was significantly different (46.0 vs 52.0) in IL-1RN A2*A2 homozygous comparing to non-A2*A2 (p?=?0.028). Our results indicated that IL-1RN A2 allele is associated with an increased susceptibility to cervical cancer development, probably by increasing predisposition to shorter immune responses.  相似文献
