Due to their large diversity of potential blood hosts, breeding habitats, and resting sites, zoological gardens represent highly interesting places to study mosquito ecology. In order to better assess the risk of mosquito‐borne disease‐agent transmission in zoos, potential vector species must be known, as well as the communities in which they occur. For this reason, species composition and dynamics were examined in 2016 in two zoological gardens in Germany. Using different methods for mosquito sampling, a total of 2,257 specimens belonging to 20 taxa were collected. Species spectra depended on the collection method but generally differed between the two zoos, while species compositions and relative abundances varied seasonally in both of them. As both sampled zoos were located in the same climatic region and potential breeding sites within the zoos were similar, the differences in mosquito compositions are attributed to immigration of specimens from surrounding landscapes, although the different sizes of the zoos and the different blood host populations available probably also have an impact. Based on the differences in species composition and the various biological characteristics of the species, the risk of certain pathogens to be transmitted must also be expected to differ between the zoos.  相似文献

Suggestions are made to define vegetation units by using formal logic. The characteristics that are used for the definition are logically combined using the AND, OR and NOT operators in such a way that relevés can be unequivocally allocated to vegetation units. Among all the characteristics of vegetation species groups are best suited to present allocation schemes for the Braun-Blanquet-system. It is shown that definition gaps within the system can be completely avoided by providing formal allocation rules.  相似文献

Sorbitol-fermenting (SF) enterohemorrhagic Escherichia coli (EHEC) O157:NM (nonmotile) is a unique clone that causes outbreaks of hemorrhagic colitis and hemolytic-uremic syndrome. In well-defined clusters of cases, we have observed significant variability in pulsed-field gel electrophoresis (PFGE) patterns which could indicate coinfection by different strains. An analysis of randomly selected progeny colonies of an outbreak strain after subcultivation demonstrated that they displayed either the cognate PFGE outbreak pattern or one of four additional patterns and were <89% similar. These profound alterations were associated with changes in the genomic position of one of two Shiga toxin 2-encoding genes (stx2) in the outbreak strain or with the loss of this gene. The two stx2 alleles in the outbreak strain were identical but were flanked with phage-related sequences with only 77% sequence identity. Neither of these phages produced plaques, but one lysogenized E. coli K-12 and integrated in yecE in the lysogens and the wild-type strain. The presence of two stx2 genes which correlated with increased production of Stx2 in vitro but not with the clinical outcome of infection was also found in 14 (21%) of 67 SF EHEC O157:NM isolates from sporadic cases of human disease. The variability of PFGE patterns for the progeny of a single colony must be considered when interpreting PFGE patterns in SF EHEC O157-associated outbreaks.  相似文献

Heterologous bacteriocin production in Propionibacterium freudenreichii is described. We developed an efficient system for DNA shuttling between Escherichia coli and P. freudenreichii using vector pAMT1. It is based on the P. freudenreichii rolling-circle replicating plasmid pLME108 and carries the cml(A)/cmx(A) chloramphenicol resistance marker. Introduction of the propionicin T1 structural gene (pctA) into pAMT1 under the control of the constitutive promoter (P4) yielded bacteriocin in amounts equal to those of the wild-type producer Propionibacterium thoenii 419. The P. freudenreichii clone showed propionicin T1 activity in coculture, killing 90% of sensitive bacteria within 48 h. The pamA gene from P. thoenii 419 encoding the protease-activated antimicrobial peptide (PAMP) was cloned and expressed in P. freudenreichii, resulting in secretion of the pro-PAMP protein. Like in the wild type, PAMP activation was dependent on externally added protease. Secretion of the antimicrobial peptide was obtained from a clone in which the pamA signal peptide and PAMP were fused in frame. The promoter region of pamA was identified by fusion of putative promoter fragments to the coding sequence of the pctA gene. The P4 and Ppamp promoters directed constitutive gene expression, and activity of both promoters was enhanced by elements upstream of the promoter core region.  相似文献

Summary The authors have studied the production of antigenic material, specific for the cervicovaginal epithelium, in organ cultures of the cervicovaginal region from neonatal mice. In control cultures the antigenic material appeared at a time corresponding to the normal appearance under in vivo conditions. When cyclic adenosine 3, 5-monophosphate (dibutyryl derivate) was added to the medium, a strong increase of the fluorescent material was seen.This investigations was supported by grants from the Norwegian Research Council and the Norwegian Cancer Society (Landsforeningen mot Kreft).  相似文献

Summary Four types of striated muscle fibers with distinctive ultrastructure were defined in the Atlantic hagfish (Myxine glutinosa, L.): white, intermediate, and red fibers of m. parietalis, and red fibers of m. craniovelaris.White fibers are thick, contain very few mitochondria and fat vacuoles, and possess distinct and separate myofibrils with thin Z-disks and distinct M-lines. Intermediate fibers are thinner, possess largely similar myofibrils that often are even better separated due to a higher content of fat vacuoles and especially mitochondria and glycogen granules. Red fibers of m. parietalis contain large amounts of mitochondria, fat vacuoles, and glycogen granules. Their myofibrils possess M-lines, and although branching more, the myofibrils of red fibers conform with a Fibrillenstruktur pattern like those of white and intermediate fibers. Red fibers of m. craniovelaris are very thin, possess many smaller fat vacuoles, and large amounts of mitochondria and glycogen granules. The myofibrils are significantly thinner than in m. parietalis fibers, run as quite independent well separated units, possess thicker Z-disks, and lack M-lines. Summary Motor nerve terminals on white and intermediate muscle fibers of the Atlantic hagfish (Myxine glutinosa, L.) contain translucent synpatic vesicles and about 1–2% dense-core vesicles. Terminals on red muscle fibers contain up to 40% dense-core vesicles with diameter 800–1100 Å. Examinations for formaldehyde-induced fluorescence indicate yellow fluorescence (5-HT ?) apparently corresponding with terminal axons on red muscle fibers in craniovelar muscles. Future phosphorus (P) scarcity and eutrophication risks demonstrate the need for systems‐wide P assessments. Despite the projected drastic increase in world‐wide fish production, P studies have yet to include the aquaculture and fisheries sectors, thus eliminating the possibility of assessing their relative importance and identifying opportunities for recycling. Using Norway as a case, this study presents the results of a current‐status integrated fisheries, aquaculture, and agriculture P flow analysis and identifies current sectoral linkages as well as potential cross‐sectoral synergies where P use can be optimized. A scenario was developed to shed light on how the projected 2050 fivefold Norwegian aquaculture growth will likely affect P demand and secondary P resources. The results indicate that, contrary to most other countries where agriculture dominates, in Norway, aquaculture and agriculture drive P consumption and losses at similar levels and secondary P recycling, both intra‐ and cross‐sectorally, is far from optimized. The scenario results suggest that the projected aquaculture growth will make the Norwegian aquaculture sector approximately 4 times as P intensive as compared to agriculture, in terms of both imported P and losses. This will create not only future environmental challenges, but also opportunities for cross‐sectoral P recycling that could help alleviate the mineral P demands of agriculture. Near‐term policy measures should focus on utilizing domestic fish scrap for animal husbandry and/or fish feed production. Long‐term efforts should focus on improving technology and environmental systems analysis methods to enable P recovery from aquaculture production and manure distribution in animal husbandry.  相似文献
