To better understand the production of microcystins (MCs) in Microcystis colonies, fluorescence in situ hybridization (FISH) methods were developed to detect DNA involved in the synthesis of these cyanobacterial hepatotoxins. Using colonies of Microcystis aeruginosa (Kütz.) Kütz. isolated from environmental blooms of cyanobacteria and from a colony‐forming, MC‐producing laboratory strain of Microcystis, amplified PCR products were observed, coincident with positive controls. The total MC content of individual colonies of Microcystis, determined by ELISA, showed a positive correlation with colony cross‐sectional area. FISH analysis of Microcystis colonies gave high fluorescence in comparison to negative controls, indicating the presence of MC synthetase DNA (mcyA) in situ. FISH analysis for MC synthetase genes has the potential to be developed into an effective early warning tool for drinking and recreational water management.  相似文献

The issues we attempt to tackle here are what the first peptides did look like when they emerged on the primitive earth, and what simple catalytic activities they fulfilled. We conjecture that the early functional peptides were short (3-8 amino acids long), were made of those amino acids, Gly, Ala, Val and Asp, that are abundantly produced in many prebiotic synthesis experiments and observed in meteorites, and that the neutralization of Asp's negative charge is achieved by metal ions. We further assume that some traces of these prebiotic peptides still exist, in the form of active sites in present-day proteins. Searching these proteins for prebiotic peptide candidates led us to identify three main classes of motifs, bound mainly to Mg2+ ions: D(F/Y)DGD corresponding to the active site in RNA polymerases, DGD(G/A)D present in some kinds of mutases, and DAKVGDGD in dihydroxyacetone kinase. All three motifs contain a DGD submotif, which is suggested to be the common ancestor of all active peptides. Moreover, all three manipulate phosphate groups, which was probably a very important biological function in the very first stages of life. The statistical significance of our results is supported by the frequency of these motifs in today's proteins, which is three times higher than expected by chance, with a P-value of 3×10−2. The implications of our findings in the context of the appearance of life and the possibility of an experimental validation are discussed.  相似文献

Purpose  New prognostic markers are needed for malignant melanoma. Inducible nitric oxide synthase (iNOS) and cyclooxygenase type 2 (COX-2) have been described to correlate with progression of melanoma. Moreover, activating mutations in BRAF/NRAS oncogenes are often detected in melanoma. The BRAF/NRAS mutation status and expression of COX-2 and iNOS were examined to compare their prognostic value for overall survival (OS) in stage III malignant cutaneous melanoma. Experimental design  The expression of iNOS and COX-2 in metastatic lymph nodes from 21 rapidly progressing (OS from date of diagnosis of stage III disease ≤14 months) and 17 slowly progressing (OS ≥60 months) stage III cutaneous melanoma patients was examined by immunohistochemistry. The presence of BRAF/NRAS mutations was analyzed using direct DNA sequencing. χ2 exact trend test and logistic regression analysis were used for statistical analysis. Results  Both iNOS (P = 0.002) and COX-2 (P = 0.048) alone significantly predicted OS. The BRAF/NRAS mutation status did not significantly differ between patient groups, although iNOS significantly (P = 0.013) correlated with BRAF mutation frequency. Furthermore, the odds ratio (OR) with respect to OS of iNOS (OR = 10.4) was higher than that of COX-2 (OR = 5.6) and was stable in the multivariate analysis of OS together with disease stage IIIB/C, ulceration, number of metastatic lymph nodes, and Breslow tumor thickness. Conclusion  Our data show that iNOS is an independent and stronger prognostic factor for OS in stage III malignant cutaneous melanoma than COX-2.  相似文献

Migratory animals such as seabirds, salmon and whales can transport large quantities of nutrients across ecosystem boundaries, greatly enriching recipient food webs. As many of these animals biomagnify contaminants, they can also focus pollutants at toxic levels. Seabirds arguably represent the most significant biovectors of nutrients and contaminants from the ocean to the land, given their sheer numbers and global distribution. However, long-term census data on seabirds are rare. Using palaeolimnological proxies, we show that a colony of Arctic seabirds has experienced climate-induced population increases in recent decades. We then document increasing concentrations of contaminants, including polychlorinated biphenyls and cadmium, in pond sediments that are linked to biotransport by seabirds. Our findings suggest that climate-related shifts in global seabird populations will have the unexpected consequence of restructuring coastal ecosystems.  相似文献

The effects of organic enrichment on sediment biogeochemistry was studied in diffusion controlled sediment mesocosms, where labile organic matter (OM) (fish feed) pulses were added once a week to the sediment surface. Two types of sediments, differing mainly in content of reactive Fe, were used. The aim of this experiment was two-fold, (1) to evaluate the importance of Fe-driven sulfide buffering for sulfide accumulation in surface enriched sediments, and (2) to estimate the diagenetic capacity for degradation of labile OM near the sediment surface. The simulated OM loading rate of 375 mmol C m?2 day?1 led to a 5–6 times increase in CO2-production and a 4–5 times increase in O2-uptake. Sulfate reduction estimated by radiotracer experiments and CO2-release was 105–131 mmol m?2 day?1, but accumulation of porewater sulfide was low in both sediment types. Instead 99% of sulfide was oxidized with O2 at the sediment water interface in the low Fe treatment, whereas 46% of produced sulfide precipitated as Fe-S compound in the high Fe treatment resulting in significantly lower O2-uptake. Furthermore, the accumulation of up to 30% of added OM by the end of the experiment indicated a saturation of the heterotrophic microbial communities in the upper enriched surface layer. These results suggest a maximum diagenetic capacity for OM degradation in the range of ~25 μmol C cm?3 day?1 or 260 mmol m?2 day?1 for the present sediment types.  相似文献

Relationships between key phytoplankton attributes including Chl a-specific light absorption, pigment composition and concentration, photosynthesis, primary production and community structure were studied in two open shallow nutrient-poor coastal systems receiving similar amounts of sewage water. Both systems were significantly nitrogen limited. However, differences in wastewater treatment (primary vs secondary) and sewage dilution (50%) between the two systems caused a greater difference between systems than locally around the outflows. For both systems, water at the outlet had significantly lower water transparency caused by a 20% higher absorption by coloured dissolved organic matter. Nutrient concentrations were also elevated, gradually decreasing with distance north (governing current) of the outflows, causing higher abundance of nano-sized phytoplankton, higher content of carotenoid pigments, 20–50% higher Chl a-specific absorption coefficients and higher photosynthetic capacity. Although maximum rates of Chl a-normalised photosynthesis were strongly related to nitrate availability, no effects were found on the derived areal primary production or algal biomass suggesting that photosynthetic and optical parameters are more sensitive indicators of nutrient enrichment than biomass or productivity. The narwhal (Monodon monoceros) is a deep diving cetacean with a strictly Arctic distribution. The challenges associated with the remoteness of narwhals have resulted in a lack of knowledge of its social behaviour requiring direct, systematic observations. Bruce Head, a peninsula at the mouth of Koluktoo Bay (Nunavut), provides an exceptional site in Canada for nearshore observation of narwhals during the summer. In this study, we document the movement, timing and grouping patterns of narwhals observed from Bruce Head and how they relate to environmental factors such as the tide and the circadian cycle. Narwhals travelled in clusters of 1–25 individuals of mixed sex and age class. Narwhals entered the bay in bigger clusters than when they exited it. The clusters were part of herds that comprised up to 642 clusters. Narwhal movement patterns were not randomly distributed in time but did not consistently follow the tidal or circadian cycles across years. Bruce Head could host long-term behavioural studies of narwhals to unravel several unanswered aspects of narwhal biology.  相似文献

8-iso-PGF isoprostane (IP) is one of the most-used markers of lipid peroxidation in experimental models and humans. After its formation, it is promptly metabolized to 2,3 dinor (DIN) in peroxisomes.Conjugated linoleic acid (CLA) is preferentially β-oxidized in peroxisomes which may compete with IP, and thereby may affect its metabolism.In order to verify whether CLA is able to influence IP formation and/or metabolism and to explain the mechanism, we challenged rats supplemented with CLA or with triolein (as a control fatty acid), with a single dose of carbon tetrachloride (CCl4) or of bacterial lipopolysaccharide (LPS). The results showed that IP and its precursor arachidonic acid hydroperoxide, as well as malondialdheyde (MDA), increase significantly in the liver of rats challenged with CCl4, irrespective of the diet, while in LPS-treated rats only nitrites in liver and isoprostane in plasma increase. On the other hand, the peroxisomal β-oxidation products of IP, the DIN, is significantly lower in the CLA group with respect to control and triolein groups.To further investigate whether this is due to competition between CLA and IP at the cellular level, we incubated human fibroblasts from healthy subjects or patients with adrenoleukodystrophy (ALD), with CLA and/or commercially available IP. The rationale of this approach is based on the deficient peroxisomal β-oxidation of fibroblasts from ALD patients, leading to a reduced formation of DIN. In both normal and ALD cells, the presence of CLA significantly inhibits the formation of DIN from IP.We may conclude that both in vitro and in vivo studies strongly suggest that CLA may impair IP catabolism in peroxisomes. Consequently an increase of IP, as a sole result of CLA intake, cannot be considered as a marker of lipid peroxidation.  相似文献
