Gelsolin and CapG are actin regulatory proteins that remodel the cytoskeleton in response to phosphatidylinositol 4,5-bisphosphate (PIP2) and Ca2+ during agonist stimulation. A physiologically relevant rise in Ca2+ increases their affinity for PIP2 and can promote significant interactions with PIP2 in activated cells. This may impact divergent PIP2- dependent signaling processes at the level of substrate availability. We found that CapG overexpression enhances PDGF-stimulated phospholipase Cγ (PLCγ) activity (Sun, H.-q., K. Kwiatkowska, D.C. Wooten, and H.L. Yin. 1995. J. Cell Biol. 129:147–156). In this paper, we examined the ability of gelsolin and CapG to compete with another PLC for PIP2 in live cells, in semiintact cells, and in vitro. We found that CapG and gelsolin overexpression profoundly inhibited bradykinin-stimulated PLCβ. Inhibition occurred at or after the G protein activation step because overexpression also reduced the response to direct G protein activation with NaF. Bradykinin responsiveness was restored after cytosolic proteins, including gelsolin, leaked out of the overexpressing cells. Conversely, exogenous gelsolin added to permeabilized cells inhibited response in a dose-dependent manner. The washout and addback experiments clearly establish that excess gelsolin is the primary cause of PLC inhibition in cells. In vitro experiments showed that gelsolin and CapG stimulated as well as inhibited PLCβ, and only gelsolin domains containing PIP2-binding sites were effective. Inhibition was mitigated by increasing PIP2 concentration in a manner consistent with competition between gelsolin and PLCβ for PIP2. Gelsolin and CapG also had biphasic effects on tyrosine kinase– phosphorylated PLCγ, although they inhibited PLCγ less than PLCβ. Our findings indicate that as PIP2 level and availability change during signaling, cross talk between PIP2-regulated proteins provides a selective mechanism for positive as well as negative regulation of the signal transduction cascade.  相似文献

The molecular properties of a novel membrane quinol oxidase from the marine bacterium Pseudomonas nautica 617 are presented. The protein contains 2b hemes/mole which may be distinguished by EPR spectroscopy but not by optical spectroscopy and electrochemistry. Respiration, though being cyanide insensitive, is not inhibited by carbon monoxide and oxygen reduction is carried out only half-way with production of hydrogen peroxide. The terminal oxidase represents, therefore, a unique example in the large family of terminal oxidases known up to date.  相似文献

1. In the absence of historical water chemistry data, predictive biological indicator groups preserved in lake sediments can be employed to reconstruct the history of lake eutrophication. Diatoms are well established in this role, but to augment diatom‐based inferences of nutrient status we investigate the potential use of chironomid midges (Insecta: Chironomidae). 2. Canonical correspondence analysis (CCA) of modern chironomid assemblages in surface sediments from 44 lakes in the English Midlands and Wales, U.K., shows that five environmental variables (total phosphorus (TP), bottom dissolved oxygen, maximum lake depth, Secchi depth and surface water temperature) make a statistically significant (P < 0.05) contribution to explaining the variance in the chironomid data, of which TP makes the largest contribution (29%). 3. The relationship is used to develop a series of weighted averaging (WA) and partial least squares (PLS), (WA‐PLS) models to infer log10TP. The models are evaluated by leave‐one‐out (jack‐knifing) cross‐validation. The simplest minimal adequate model is provided by WA with unweighted inverse deshrinking of root mean square error of prediction (RMSEPjack=0.34 and r2jack=0.60). 4. Using this model, the trophic history of Betton Pool, Shropshire, U.K., is reconstructed from the mid‐19th century to the present day and the results from the chironomid‐TP model are compared with inferences from a diatom‐TP model ( 13 ). Both reconstructions suggest that there was a gradual rise in TP since 1850 AD until about 1974, followed by a more pronounced and rapid increase that has continued until the present. Inferred TP values from the WA chironomid inference model agree with diatom‐inferred values. 5. The study demonstrates that fossil chironomid assemblages can be used to investigate quantitatively the trophic history of lakes.  相似文献

Novel data on the biometry, size distribution and parasitesof Aequorea aequorea and Chrysaora hysoscella are provided frominvestigations conducted during summer and winter in the northernBenguela ecosystem. The relationship between mass and diameterof C. hysoscella did not change on a seasonal basis, and thispossibly reflects the aseasonal nature of the food environment.The changes in the size structure of C. hysoscella across theshelf and with depth agree with postulated population maintenancestrategies in the region. Aequorea aequorea was not stronglyparasitized, butC. hysoscella was subject to occasional parasitismby Hyperia medusarum, especially in winter when C. hysoscellais thought to reproduce. Parasites were distributed in a typical,negative-binomial manner on their hosts, but load was independentof host size. As medusae increased in diameter so H. medusarumtended to move from other tissues to the gonads.  相似文献

Litter quality parameters of Danthonia richardsonii grown under CO2 concentrations of ≈ 359 & ≈ 719 μL L? 1 at three mineral N supply rates (2.2, 6.7 & 19.8 g m? 2 y? 1) were determined. C:N ratio was increased in senesced leaf (enhancement ratios, Re/c, of 1.25–1.67), surface litter (1.34–1.64) and root (1.13–1.30) by CO2 enrichment. After 3 years of growth, nonstructural carbohydrate concentrations were reduced in senesced leaf lamina (avg. Re/c= 0.84) but not in root in response to CO2 enrichment. Cellulose concentrations increased slightly in senesced leaf (avg. Re/c= 1.07) but not in root in response to CO2 enrichment. Lignin and polyphenolic concentrations in senesced leaf and root were not changed by CO2 enrichment. Decomposition, measured as cumulative respiration in standard conditions in vitro, was reduced in leaf litter grown under CO2 enrichment. Root decomposition in vitro was lower in the material produced under CO2 enrichment at the two higher rates of mineral N supply. Significant correlations between decomposition of leaf litter and initial %N, C:N ratio and lignin:N ratio were found. Decomposition in vivo, measured as carbon disappearance from the surface litter was not affected by CO2 concentration. Arbuscular mycorrhizal infection was not changed by CO2 enrichment. Microbial carbon was higher under CO2 enrichment at the two higher rates of mineral N supply. Possible reasons for the lack of effect of changes in litter quality on in‐sward decomposition rates are discussed.  相似文献

Abstract The first instances of egg parasitism of Chrysophtharta agricola , a pest of eucalypt plantations, are recorded. Enoggera nassaui was found parasitising C. agricola egg batches in Tasmania, the Australian Capital Territory (ACT), New South Wales and Victoria: this is the first record of this parasitoid species from Victoria. One instance of Neopolycystus sp. parasitising C. agricola eggs in Victoria was also recorded. Parasitism of egg batches by E. nassaui ranged from 0 to 55% between five geographical populations collected in mainland Australia ( n  = 45), and from 0 to 2% between two populations collected in Tasmania ( n  = 300). For mainland sites at which parasitism was recorded, parasitism rates within sites differed significantly from either population in Tasmania. Reciprocal exposure experiments using one Tasmanian (Florentine Valley) and one parasitised mainland (Picadilly Circus, ACT) population were conducted in the laboratory to examine whether these different parasitism rates were attributable to egg or parasitoid origin. Parasitoids from the ACT parasitised C. agricola eggs of both origins more successfully than parasitoids from Tasmania, with up to 65% wasp emergence compared with 33% from Tasmania. Parasitoid origin significantly affected the number of wasps that emerged from exposed batches, but not the total loss from parasitism.  相似文献

Proanthocyanidin contents of needles ranged from a mean of 150 to 300 μg per mg dry wt in five species from five genera of the Taxodiaceae, Sequoiadendron giganteum (Lindl.) Bucch., Metasequoiaglyptostroboides H. Hu and Cheng, Sequoia sempervirens (D. Don) Endl., Taxodium distichum L. Rich., and Sciadopitys verticillata Siebold and Zucc. However, significantly lower amounts (70 μg per mg dry wt) were found in Cryptomeria japonica (L.f.) D. Don. This latter species as well as Sciadopitys verticillata, contained little or no prodelphinidin, while the other four species contained a ratio of procyanidin to prodelphinidin up to about 1:5. These data were based on analyses from three trees from each species. In addition, one tree from each species was examined in more detail. The major flavan-3-ol in all cases was (+)-catechin, with only non-detectable or trace amounts of (–)-epicatechin. The triphenolic flavan-3-ol, (+)-gallocatechin, was a minor constituent in all species, except Sciadopitys and Cryptomeria. The (–)-epigallocatechin was detected in Metasequoia, Sequoiadendron and Sequoia. All contained either (–)-epicatechin-(+)-catechin or (+)-catechin-(+)-catechin as the major procyanidin dimer. Prodelphinidin dimers were only tentatively identified.  相似文献
