Resistance to citrus tristeza virus (CTV) was evaluated in 554 progeny of 10 populations derived from Poncirus trifoliata. A dominant gene (Ctv) controlled CTV resistance in P. trifoliata. Twenty-one dominant PCR-based DNA markers were identified as linked to Ctv by bulked segregant analysis. Of the 11 closest markers to Ctv, only 2 segregated in all populations. Ten of these markers were cloned and sequenced, and codominant RFLP markers were developed. Seven RFLP markers were then evaluated in 10 populations. Marker orders were consistent in all linkage maps based on data of single populations or on combined data of populations with similar segregation patterns. In a consensus map, the six closest marker loci spanned 5.3 cM of the Ctv region. Z16 cosegregated with Ctv. C19 and AD08 flanked Ctv at distances of 0.5 and 0.8 cM, respectively. These 3 markers were present as single copies in the Poncirus genome, and could be used directly for bacterial artificial chromosome library screening to initiate a walk toward Ctv. BLAST searches of the GenBank database revealed high sequence similarities between 2 markers and known plant disease resistance genes, indicating that a resistance gene cluster exists in the Ctv region in P. trifoliata.  相似文献

Detailed evaluation of coronary function early in diabetes mellitus (DM)-associated coronary artery disease (CAD) development is difficult in patients. Therefore, we investigated coronary conduit and small artery function in a preatherosclerotic DM porcine model with type 2 characteristics. Streptozotocin-induced DM pigs on a saturated fat/cholesterol (SFC) diet (SFC + DM) were compared with control pigs on SFC and standard (control) diets. SFC + DM pigs showed DM-associated metabolic alterations and early atherosclerosis development in the aorta. Endothelium-dependent vasodilation to bradykinin (BK), with or without blockade of nitric oxide (NO) synthase, endothelium-independent vasodilation to an exogenous NO-donor (S-nitroso-N-acetylpenicillamine), and vasoconstriction to endothelin (ET)-1 with blockade of receptor subtypes, were assessed in vitro. Small coronary arteries, but not conduit vessels, showed functional alterations including impaired BK-induced vasodilatation due to loss of NO (P < 0.01 vs. SFC and control) and reduced vasoconstriction to ET-1 (P < 0.01 vs. SFC and control), due to a decreased ET(A) receptor dominance. Other vasomotor responses were unaltered. In conclusion, this model demonstrates specific coronary microvascular alterations with regard to NO and ET-1 systems in the process of early atherosclerosis in DM. In particular, the altered ET-1 system correlated with hyperglycemia in atherogenic conditions, emphasizing the importance of this system in DM-associated CAD development.  相似文献

Photosystem II (PSII) is a large membrane bound molecular machine that catalyzes light-driven oxygen evolution from water. PSII constantly undergoes assembly and disassembly because of the unavoidable damage that results from its normal photochemistry. Thus, under physiological conditions, in addition to the active PSII complexes, there are always PSII subpopulations incompetent of oxygen evolution, but are in the process of undergoing elaborate biogenesis and repair. These transient complexes are difficult to characterize because of their low abundance, structural heterogeneity, and thermodynamic instability. In this study, we show that a genetically tagged Psb27 protein allows for the biochemical purification of two monomeric PSII assembly intermediates, one with an unprocessed form of D1 (His27ΔctpAPSII) and a second one with a mature form of D1 (His27PSII). Both forms were capable of light-induced charge separation, but unable to photooxidize water, largely because of the absence of a functional tetramanganese cluster. Unexpectedly, there was a significant amount of the extrinsic lumenal PsbO protein in the His27PSII, but not in the His27ΔctpAPSII complex. In contrast, two other lumenal proteins, PsbU and PsbV, were absent in both of these PSII intermediate complexes. Additionally, the only cytoplasmic extrinsic protein, Psb28 was detected in His27PSII complex. Based on these data, we have presented a refined model of PSII biogenesis, illustrating an important role of Psb27 as a gate-keeper during the complex assembly process of the oxygen-evolving centers in PSII.  相似文献

Spatially averaged models of root–soil interactions are often used to calculate plant water uptake. Using a combination of X‐ray computed tomography (CT) and image‐based modelling, we tested the accuracy of this spatial averaging by directly calculating plant water uptake for young wheat plants in two soil types. The root system was imaged using X‐ray CT at 2, 4, 6, 8 and 12 d after transplanting. The roots were segmented using semi‐automated root tracking for speed and reproducibility. The segmented geometries were converted to a mesh suitable for the numerical solution of Richards' equation. Richards' equation was parameterized using existing pore scale studies of soil hydraulic properties in the rhizosphere of wheat plants. Image‐based modelling allows the spatial distribution of water around the root to be visualized and the fluxes into the root to be calculated. By comparing the results obtained through image‐based modelling to spatially averaged models, the impact of root architecture and geometry in water uptake was quantified. We observed that the spatially averaged models performed well in comparison to the image‐based models with <2% difference in uptake. However, the spatial averaging loses important information regarding the spatial distribution of water near the root system.  相似文献

To manage and intelligently mine the avalanche of genomic sequences intuitive and user-friendly graphical interfaces are required. Here we present BlastXtract2 which exclusively facilitates early exploration of un-annotated genomic and metagenomic sequences. Various formats of translated searches, including the commonly used BlastX, of multiple sequences against multiple protein databases can be uploaded to a relational database server, which can be accessed via a locally installed web-server. There, an intuitive GUI allows straightforward data-mining and enables quick detection of potential frameshifts and poorly sequenced or assembled regions, thereby contributing in making BlastXtract2 a unique and valuable tool for early exploration of (meta)genomic sequences.

Arbuscular mycorrhizas, associations between plant roots and soil fungi, are ubiquitous among land plants. Arbuscular mycorrhizas can be beneficial for plants by overcoming limitations in nutrient supply. Hyphae, which are long and thin fungal filaments extending from the root surface into the soil, increase the volume of soil accessible for plant nutrient uptake. However, no models so far specifically consider individual hyphae. We developed a mathematical model for nutrient uptake by individual fungal hyphae in order to assess suitable temporal and spatial scales for a new experimental design where fungal uptake parameters are measured on the single hyphal scale. The model was developed based on the conservation of nutrients in an artificial cylindrical soil pore (capillary tube) with adsorbing wall, and analysed based on parameter estimation and non-dimensionalisation. An approximate analytical solution was derived using matched asymptotic expansion. Results show that nutrient influx into a hypha from a small capillary tube is characterized by three phases: Firstly, uptake rapidly decreases as the hypha takes up nutrients, secondly, the depletion zone reaches the capillary wall and thus uptake is sustained by desorption of nutrients from the capillary wall, and finally, uptake goes to zero after nutrients held on the capillary wall have been completely depleted. Simulating different parameter regimes resulted in recommending the use of capillaries filled with hydrogel instead of water in order to design an experiment operating over measurable time scales.  相似文献

CFSE based tracking of the lymphocyte proliferation using flow cytometry is a powerful experimental technique in immunology allowing for the tracing of labelled cell populations over time in terms of the number of divisions cells undergone. Interpretation and understanding of such population data can be greatly improved through the use of mathematical modelling. We apply a heterogenous linear compartmental model, described by a system of ordinary differential equations similar to those proposed by Kendall. This model allows division number-dependent rates of cell proliferation and death and describes the rate of changes in the numbers of cells having undergone j divisions. The experimental data set that we specifically analyze specifies the following characteristics of the kinetics of PHA-induced human T lymphocyte proliferation assay in vitroL (1) the total number of live cells, (2) the total number of dead but not disintegrated cells and (3) the number of cells divided j times. Following the maximum likelihood approach for data fitting, we estimate the model parameters which, in particular, present the CTL birth- and death rate “functions”. It is the first study of CFSE labelling data which convincingly shows that the lymphocyte proliferation and death both in vitro and in vivo are division number dependent. For the first time, the confidence in the estimated parameter values is analyzed by comparing three major methods: the technique based on the variance–covariance matrix, the profile-likelihood-based approach and the bootstrap technique. We compare results and performance of these methods with respect to their robustness and computational cost. We show that for evaluating mathematical models of differing complexity the information-theoretic approach, based upon indicators measuring the information loss for a particular model (Kullback–Leibler information), provides a consistent basis. We specifically discuss methodological and computational difficulties in parameter identification with CFSE data, e.g. the loss of confidence in the parameter estimates starting around the sixth division. Overall, our study suggests that the heterogeneity inherent in cell kinetics should be explicitly incorporated into the structure of mathematical models.   相似文献

Clinical efficacy of the antiplatelet drug clopidogrel is hampered by its variable biotransformation into the active metabolite. The variability in the clinical response to clopidogrel treatment has been attributed to genetic factors, but the specific genes and mechanisms underlying clopidogrel bioactivation remain unclear. Using in vitro metabolomic profiling techniques, we identified paraoxonase-1 (PON1) as the crucial enzyme for clopidogrel bioactivation, with its common Q192R polymorphism determining the rate of active metabolite formation. We tested the clinical relevance of the PON1 Q192R genotype in a population of individuals with coronary artery disease who underwent stent implantation and received clopidogrel therapy. PON1 QQ192 homozygous individuals showed a considerably higher risk than RR192 homozygous individuals of stent thrombosis, lower PON1 plasma activity, lower plasma concentrations of active metabolite and lower platelet inhibition. Thus, we identified PON1 as a key factor for the bioactivation and clinical activity of clopidogrel. These findings have therapeutic implications and may be exploited to prospectively assess the clinical efficacy of clopidogrel.  相似文献

Immunocontraception has been widely used as a management tool to reduce population growth in captive as well as wild populations of various fauna. We model the use of an individual-based rotational immunocontraception plan on a wild elephant, Loxodonta africana, population and quantify the social and reproductive advantages of this method of implementation using adaptive management. The use of immunocontraception on an individual, rotational basis stretches the inter-calving interval for each individual female elephant to a management-determined interval, preventing exposing females to unlimited long-term immunocontraception use (which may have as yet undocumented negative effects). Such rotational immunocontraception can effectively lower population growth rates, age the population, and alter the age structure. Furthermore, such structured intervention can simulate natural process such as predation or episodic catastrophic events (e.g., drought), which regulates calf recruitment within an abnormally structured population. A rotational immunocontraception plan is a feasible and useful elephant population management tool, especially in a small, enclosed conservation area. Such approaches should be considered for other long-lived, social species in enclosed areas where the long-term consequences of consistent contraception may be unknown.  相似文献
