Adhesive interactions play important roles in coordinating T cell migration and activation, which are mediated by binding of integrins to RGD motif found on extracellular matrix proteins. Disintegrins, isolated from snake venoms, contain the RGD sequence that confers selectivity to integrin interaction. We have investigated the ability of three RGD-disintegrins, ligands of alpha(5)beta(1) and alpha(v)beta(3), Flavoridin (Fl), Kistrin (Kr) and Echistatin (Ech), in modulating the activation of human T lymphocyte. The disintegrins induced T cell proliferation and CD69 expression. This activation parallels with actin cytoskeleton reorganization and tyrosine phosphorylation. Furthermore, the peptides induced focal adhesion kinase (FAK) and phosphoinositide 3-kinase (PI3K) activation. Finally, RGD-disintegrins were capable of driving NF-kappaB nuclear translocation and c-Fos expression, in a PI3K and ERK1/2 activities dependent manner. This report is the first to show that RGD-disintegrins interact with integrins on human T lymphocyte surface, modulating cell proliferation and activation of specific pathways coupled to integrin receptor.  相似文献

Rhesus macaques are considered an important reservoir of different gastrointestinal (GI) zoonotic parasites affecting livestock and humans. Loads of GI parasites in the free-ranging rhesus macaques living in close proximity to communities in Bangladesh are still unknown. To estimate the prevalence and diversity of zoonotic GI parasites in rhesus macaques of Bangladesh, a total of 182 freshly voided fecal samples were collected from macaques living in rural (N = 67), peri-urban (N = 57), urban (N = 28), and Safari park (N = 30) between October 2015 and December 2016. All samples were tested by direct smear, sedimentation, flotation, and the McMaster techniques. A total of fourteen different taxa of GI parasites were detected, revealing an overall prevalence of 54.4% (n = 99; 95% confidence interval [CI]: 46.9–61.8). The prevalence of GI parasites was found to be significantly correlated with the mean parasitic taxa per individual in a group (r = 0.90; p = 0.002). The multivariable logistic regression analysis showed that the overall prevalence of GI parasites in macaques was significantly higher in those inhabiting rural areas (62.69%; odds ratio [OR]: 7.22; p = 0.001) and in macaques with interactions with other animals (60.98%; OR: 5.49; p = 0.005). Our results also indicated that the prevalence of Strongyloides spp. and Balantidium coli infections varied significantly between land gradients. Our results also indicate that macaques frequently visit human settlements for food and are found interacting with domestic animals. In conclusion, the high prevalence of zoonotic GI parasite infection in rhesus macaques found in our study may pose a significant public health risk to communities, particularly in rural areas of Bangladesh. Health promotion to at-risk communities focusing on limiting contact with rhesus macaques is necessary to mitigate potential zoonotic transmission.  相似文献

目的:探讨P13K特异性抑制剂LY294002逆转顺铂耐药口腔鳞癌细胞TCA8113/CDDP的可行性。方法:采用间歇性加药,逐步递增CDDP药量,体外连续诱导培养TCA8113/CDDP细胞;用不同浓度的LY294002和顺铂处理TCA8113和TCA8113/CDDP细胞;MTT法观察对细胞增殖的影响,Western印迹分析LY294002作用前后p-Akt、Akt、P13K蛋白的表达。结果:建立了舌鳞癌耐药细胞TCA8113/CDDP,耐药指数为7.7;MTT实验显示LY294002对TCA8113和TCA8113/CDDP细胞的抑制作用与浓度及作用时间呈正相关;LY294002联合顺铂对2种细胞的抑制作用比单用顺铂效果好;P13K、Akt、P—AKT蛋白表达明显降低,其中TCA8113/CDDP细胞中P13K、AKT、p-AKT蛋白的表达比TCA8113细胞明显增多(P〈0.05)。结论:LY294002能增加耐药口腔鳞癌顺铂化疗的敏感性。  相似文献

The surface myoelectric (ME) signal from the vastus lateralis muscle was studied during ergocycling at various mechanical powers (MPs) and pedalling rates (PRs). The envelope of the signal was described using a power spectral density function estimate at very low frequency. A highly smoothed burst pattern was found independently at 80 rpm on MP. The envelope presence in surface ME signals was demonstrated to affect the total band power spectrum estimate slightly. In addition, the total band power spectrum changes with MPs and PRs were described using the root mean square value and the mean power frequency. Total band power spectrum was enhanced and shifted toward the low frequencies as PR increased. It was shifted toward the high frequencies as MP increased. These changes were attributed to the progressive recruitment of fast fibers.  相似文献

Objective

The objective of this work was to isolate bacteria from Red Sea invertebrates, determine their antimicrobial activity, and screen for the biosynthetic gene clusters [polyketides (PKs) and nonribosomal peptides (NRPs)] which could be involved in the production of bioactive secondary metabolites.

Result

Eleven different samples of marine invertebrates’ were collected from Egypt’s Red Sea (El-Tor-Sharm El-Sheikh and Hurghada) by scuba diving, and a total 80 isolates of the associated microorganisms were obtained from the cultivation on six different cultural medium. Seven isolates of them showed an antimicrobial activity against five pathogenic reference strains, while the most active antimicrobial agent was isolate number HFF-8 which was 99% identical to Bacillus amyloliquefaciens. HFF-8’s extract showed positive results against Gram negative bacteria, Gram positive bacteria and yeast. Moreover, the isolates gave positive bands when screened for the presence of PK synthase (PKS) I and II and NRP synthetase (NRPS) I and II biosynthetic genes, those biosynthetic fragments when cloned and sequenced were primitively predicted as biosynthetic fragments for kirromycin and leinamycin production by NaPDoS program with 56 and 55%, respectively.

Conclusion

The Red Sea can provide a sustainable solution to combat bacterial resistance. The contribution of this work is that B. amyloliquefaciens was isolated from Heteroxenia fuscescens, Red Sea, Egypt. Moreover, the bacterial extract showed a broad spectrum with a potent antimicrobial activity.

A series of experiments was carried out using adult outbred Polish race rabbits of both sexes infected, during spring or autumn, with 10,000 larvae of Obeliscoides cuniculi, either fresh or stored at 4 degrees C. Extracts of mucosal proteins and bile were collected at postmortem 6 or 12 weeks after infection. Antibody levels were determined in antisera, bile and stomach mucosa by haemagglutination and precipitation tests. Local antibody responses were demonstrated in the stomach and bile, and reactions were obtained with the tissue fluids by haemagglutination and precipitation tests with worm antigens and ES products. Additionally, some specific immunological response was observed in the circulation during the primary infection. These results suggest a clear-cut relationship between increased levels of these antibodies and either larval inhibition or worm expulsion during O. cuniculi infections.  相似文献

The constant region of the gamma 1, gamma 2 and gamma 3 heavy chains of the human IgG1, IgG2 and IgG3 immunoglobulins carries antigenic determinants or G1m, G2m and G3m allotypes, which are genetic markers of these subclasses. The exceptional presence on gamma 1 and gamma 2 chains of Gm allotypes usually located on the CH3 domain of gamma 3 shows an unexpected clustering of base changes and subsequent identity of short DNA sequences in the CH3 exon of the non-allelic gamma 1, gamma 2 and gamma 3 genes. Such clusters of substitutions are not easily explained on the classical basis of point mutations. A gene conversion, which substituted a segment of the gamma 1 or gamma 2 gene with the homologous region of the non-allelic gamma 3 gene, is more likely. Other examples of possible conversion involving the gamma genes are described. The conservation or the restoration of short sequences produced by the conversion events might be related to the biological properties of the constant region of the heavy chains.  相似文献

The surface electromyogram (EMG) from the vastus lateralis muscle and the metabolic and respiratory parameters were studied simultaneously during an incremental exercise in order to identify EMG signal modifications during the aerobic-anaerobic transition. Subjects performed an incremental test on the bicycle ergometer from an initial work load of 175 W to exhaustion by steps of 25 W. Ventilatory flow (VE), oxygen uptake (VO2) and carbon dioxide flow (VCO2) were recorded continuously. For lactate concentration determination, venous blood samples were collected during the final 30 s of each step. EMG signals were stored on magnetic tape. They were then converted into successive spectra to allow the study of EMG total power (PEMG) and mean power frequency (MPF) evolutions. A non linear increase in blood lactate reflected by a breaking point at 250 W was observed. A change in VE/VO2 ratio occurred at 275 W. PEMG value showed a non linear increase reflected by a breaking point at 275 W. MPF value increased from the first to the seventh step with a tendency to decrease at the last step. A great interindividual variance of EMG data was observed indicating the difficulty of correlating mean values of EMG parameters with mean values of blood lactate in order to explain sudden lactate increase by fast twitch fibre recruitment. However, comparison of individual EMG data suggests a progressive recruitment of fast twitch fibres as work load increases.  相似文献
