Tuberculosis (TB) is one of the most common infectious diseases worldwide. IL‐37, a novel member of the IL‐1 family, has anti‐inflammatory activity. Various cytokine genes polymorphisms are reportedly associated with susceptibility to TB infection. However, an association between genetic variations in the IL‐37 gene and susceptibility to TB infection has not been investigated. The aim of this case‐control study was therefore to identify such an association in Saudi subjects, in which five single‐nucleotide polymorphisms (SNPs) in the IL‐37 gene were assessed. Serum concentrations of IL‐37 were evaluated using ELISA, and genetic variants genotyped by multiplex PCR and ligase detection reaction. It was found that the C/C genotype of rs2723176 (–6962 A/C) occurs significantly more frequently in patients with active TB and that the C allele of this SNP is associated with TB. In addition, the C allele of rs2723176 SNP was associated with high circulating concentrations of IL‐37. However, the genotype and allele frequency of the other four SNPs (rs3811046, rs3811047, rs2723186 and rs2723187) were not significantly associated with TB infection. In conclusion, the present data suggest that rs2723176 SNP of IL‐37 is involved in the development of TB infection. Furthermore, high circulating concentrations of IL‐37 may have a negative effect on protective immunity against TB infection.  相似文献

We investigated the interaction between GM-CSF and its receptor on human granulocytes and on several human tumor cell lines. Specific high-affinity binding for GM-CSF was characterized by Scatchard plot analysis. The specific radioactivity of the 125I-labeled derivative of rH. GM-CSF was determined by self-displacement analysis and calculated to be 30 microCi/micrograms. The maximum concentration of binding sites (B max) in granulocytes was 40 fmol/mg protein (2,200 molecules GM-CSF bound/cell) and the dissociation constant (KD) was 0.42 nM. No binding sites for GM-CSF were found in two lung cancer cell lines, SCLC-16HV and NCI-N417 or in the urinary bladder carcinoma cell line 5637, whereas the promyelocytic leukemia cell line HL60 was positive for GM-CSF binding. Time course experiments showed maximum binding of GM-CSF in granulocytes after an incubation period of 60 min and a decrease in binding after an incubation period of 2 h. In parallel, we found a maximum biological signal when granulocytes were preincubated for 90 min with GM-CSF, and a decrease after an incubation time of 120 min. Preincubation of the cells with rH. GM-CSF induced an enhancement of the production of activated oxygen species by the cells in response to PMA.  相似文献

This work deals with the effect of temperature variations on growth, reproduction, amino acid synthesis, fat and sugar content in Ulva fasciata in habiting the littoral region of the Mediterranean shore bordering Alexandria.Higher temperatures above 30°C have lethal effects on some parts of the algal fronds while the other parts are rather resistant. The latter are scattered groups of cells, which usually regenerate new plants when carried even to distant parts due to detrimental action of waves and currents. They serve a good means for vegetative propagation and spread better than zooids, which probably perish under such severe and violent conditions.The rate of growth of Ulva indicated by the increase in dry weight and total nitrogen, increases at rising temperature till the latter becomes limiting.Higher temperatures favour the growth and development of sporophytic phases, known by their deep green colouration, while temperatures below 20°C favour the production of gametophytes, identified by their yellowish green colour. This might explain why most of the algal crops of Ulva fasciata produced from parental stock (3 crops per year i.e. annually), are sporophytes.Moderate temperatures were favourable for growth, reproduction and the various metabolic processes. Amino acids and sugar contents increase with the rise of temperature, reaching their maximum around 25°C, while fat content increases at still lower temperatures.  相似文献

Summary Experiments were conducted to study the survival of Alternaria cheiranthi in wet and dry soil. As free spores, survival was demonstrated only in air-dry soil. In wet soil, lysis of the germ tubes commenced after the second day. On wallflower seeds, the pathogen was viable until the seeds were decomposed. Microbial resistance to the available drugs poses a serious threat in modern medicine. We report the design, synthesis and in vitro antimicrobial evaluation of new functionalized 2,3-dihydrothiazoles and 4-thiazolidinones tagged with sulfisoxazole moiety. Compound 8d was most active against Bacillis subtilis (MIC, 0.007?µg/mL). Moreover, compounds 7cd and 8c displayed significant activities against B. subtilis and Streptococcus pneumoniae (MIC, 0.03–0.06?µg/mL and 0.06–0.12?µg/mL versus ampicillin 0.24?µg/mL and 0.12?µg/mL; respectively). Compounds 7a and 7cd were highly potent against Escherichia coli (MIC, 0.49–0.98?µg/mL versus gentamycin 1.95?µg/mL). On the other hand, compounds 7e and 9c were fourfolds more active than amphotericin B against Syncephalastrum racemosum. Molecular docking studies showed that the synthesized compounds could act as inhibitors for the dihydropteroate synthase enzyme (DHPS). This study is a platform for the future design of more potent antimicrobial agents.  相似文献

Cyclodextrin glycosyltransferase (CGTase, EC 2.4.1.19) was isolated from the cell-free culture medium of Bacillus macerans strain BKMB-506. The purified enzyme was used to immunize BALB/c mice. Polyclonal antibodies (PAbs) and 15 hybridoma cells producing monoclonal antibodies (MAbs) against CGTase were obtained. The properties of MAbs were studied using ELISA and immunoblotting. A method for detecting small amounts of CGTase was developed. The interactions of PAbs and MAbs with CGTase of Bac. circulans var. alkalophilis were investigated.  相似文献

Abstract

Four organic solvents along with water were applied for the conventional extraction of Eucalyptus camaldulensis (Myrtaceae), phenolic contents and antioxidant activities were investigated through variable protocols and correlation coefficients were considered, the phenolic composition was also characterized by Gas chromatography-mass spectrometry (GC-MS). Using solvents with dissimilar polarities affected the phenolic yields extracted from E. camaldulensis and their related antioxidant activities varied significantly among the four investigated plant organs. The leaf extract of acetone 70% contained the highest amount of phenolic compounds (46.56?mg/g dry weight); while the bud-water boiled extract maintained the maximum value of tannins (45.68?mg/g dry weight). Correlation coefficients indicated that phenolic compounds were mostly accountable for the phosphomolybednum antioxidant potentials (0.520), followed by tannins (0.460). Also, both the reducing power activities and hydrogen peroxide scavenging of E. camaldulensis extracts positively correlated with tannins, but at different significance degrees. However, the GC-MS analysis revealed that most of the detected phenolic constituents were more abundant in the plant seed. So, the existence of some other compounds such as organic acids, along with phenolics, may have increased the antioxidant potentials of leaf and bud. Undeniably, the optimization of extraction conditions could stimulate the antioxidant capabilities of the plant extracts of E. camaldulensis.  相似文献
