Little is known on the role played by Neotropical wild carnivores in the Trypanosoma cruzi transmission cycles. We investigated T. cruzi infection in wild carnivores from three sites in Brazil through parasitological and serological tests. The seven carnivore species examined were infected by T. cruzi, but high parasitemias detectable by hemoculture were found only in two Procyonidae species. Genotyping by Mini-exon gene, PCR-RFLP (1f8/Akw21I) and kDNA genomic targets revealed that the raccoon (Procyon cancrivorus) harbored TcI and the coatis (Nasua nasua) harbored TcI, TcII, TcIII-IV and Trypanosoma rangeli, in single and mixed infections, besides four T. cruzi isolates that displayed odd band patterns in the Mini-exon assay. These findings corroborate the coati can be a bioaccumulator of T. cruzi Discrete Typing Units (DTU) and may act as a transmission hub, a connection point joining sylvatic transmission cycles within terrestrial and arboreal mammals and vectors. Also, the odd band patterns observed in coatis’ isolates reinforce that T. cruzi diversity might be much higher than currently acknowledged. Additionally, we assembled our data with T. cruzi infection on Neotropical carnivores’ literature records to provide a comprehensive analysis of the infection patterns among distinct carnivore species, especially considering their ecological traits and phylogeny. Altogether, fifteen Neotropical carnivore species were found naturally infected by T. cruzi. Species diet was associated with T. cruzi infection rates, supporting the hypothesis that predator-prey links are important mechanisms for T. cruzi maintenance and dispersion in the wild. Distinct T. cruzi infection patterns across carnivore species and study sites were notable. Musteloidea species consistently exhibit high parasitemias in different studies which indicate their high infectivity potential. Mesocarnivores that feed on both invertebrates and mammals, including the coati, a host that can be bioaccumulator of T. cruzi DTU’s, seem to take place at the top of the T. cruzi transmission chain. 相似文献

Background
The Brazilian Cerrado is one of the most important biodiversity reservoirs in the world. The sugarcane cultivation is expanding in this biome and necessitates the study of how it may impact the soil properties of the Cerrado. There is a lack of information especially about the impacts of different sugarcane management on the native bacterial communities of Cerrado soil. Therefore, our objective was to evaluate and compare the soil bacterial community structure of the Cerrado vegetation with two sugarcane systems.Methods
We evaluated samples under native vegetation and the impact of the two most commonly used management strategies for sugarcane cultivation (burnt cane and green cane) on this diversity using pyrosequencing and quantitative PCR of the rrs gene (16S rRNA).Results and Conclusions
Nineteen different phyla were identified, with Acidobacteria (≈35%), Proteobacteria (≈24%) and Actinobacteria (≈21%) being the most abundant. Many of the sequences were represented by few operational taxonomic units (OTUs, 3% of dissimilarity), which were found in all treatments. In contrast, there were very strong patterns of local selection, with many OTUs occurring only in one sample. Our results reveal a complex bacterial diversity, with a large fraction of microorganisms not yet described, reinforcing the importance of this biome. As possible sign of threat, the qPCR detected a reduction of the bacterial population in agricultural soils compared with native Cerrado soil communities. We conclude that sugarcane cultivation promoted significant structural changes in the soil bacterial community, with Firmicutes phylum and Acidobacteria classes being the groups most affected. 相似文献63.

In about 10–15% of patients with inflammatory bowel diseases (IBD) there is no clear definitive differential diagnosis between Crohn''s disease (CD) and ulcerative colitis (UC) and the disease is classified as indeterminate colitis. Since pharmacological and surgical treatments differ in CD and UC, establishing a correct diagnosis is critical. The aim of this work was to access the expression profile of proteins involved in colonic inflammation and cancer in samples from CD and UC. For that, colon samples from 24 CD, 21 UC and 10 control patients were processed for immunohistochemistry using anti-phosphorylated RB at Ser807/811 and anti-β-catenin. Crypts were blinded, analyzed and counted for phosphorylated RB-positive (phospho-RB) cells or scored for positive β-catenin staining. Western blot was used for confirming immuhistochemical results: RB phosphorylation was significantly greater in colon samples from patients with CD compared with UC (p<0.005). In contrast, the expression of β-catenin was significantly increased in UC compared with CD (p<0.005) samples. Phospho-RB and β-catenin are negatively correlated (CC: −0.573; p = 0.001). A positive phospho-RB test yielded high levels of sensitivity, specificity, negative and positive predictive values, and accuracy for the diagnosis of CD against UC. This work indicates that RB phosphorylation and β-catenin nuclear translocation are differently expressed in CD and UC, and provide novel insights into the pathogenic mechanisms of IBD. In particular, rates of phospho-RB-positive cells in mucosal samples emerge as a promising tool for the differential diagnosis of patients with IBD. 相似文献

Bioethanol from sugarcane is becoming an increasingly important alternative energy source worldwide as it is considered to be both economically and environmentally sustainable. Besides being produced from a tropical perennial grass with high photosynthetic efficiency, sugarcane ethanol is commonly associated with low N fertilizer use because sugarcane from Brazil, the world's largest sugarcane producer, has a low N demand. In recent years, several models have predicted that the use of sugarcane ethanol in replacement to fossil fuel could lead to high greenhouse gas (GHG) emission savings. However, empirical data that can be used to validate model predictions and estimates from indirect methodologies are scarce, especially with regard to emissions associated with different fertilization methods and agricultural management practices commonly used in sugarcane agriculture in Brazil. In this study, we provide in situ data on emissions of three GHG (CO2, N2O, and CH4) from sugarcane soils in Brazil and assess how they vary with fertilization methods and management practices. We measured emissions during the two main phases of the sugarcane crop cycle (plant and ratoon cane), which include different fertilization methods and field conditions. Our results show that N2O and CO2 emissions in plant cane varied significantly depending on the fertilization method and that waste products from ethanol production used as organic fertilizers with mineral fertilizer, as it is the common practice in Brazil, increase emission rates significantly. Cumulatively, the highest emissions were observed for ratoon cane treated with vinasse (liquid waste from ethanol production) especially as the amount of crop trash on the soil surface increased. Emissions of CO2 and N2O were 6.9 kg ha?1 yr?1 and 7.5 kg ha?1 yr?1, respectively, totaling about 3000 kg in CO2 equivalent ha?1 yr?1. 相似文献

In this study we explore the ability of a previously developed model of pulsatile flow for explaining the observed reduction of arterial distensibility with heart rate. The parameters relevant for the analysis are arterial wall distensibility together with permeability and reflection coefficients of the end capillaries. A non-specific artery and the ensemble of tissues supplied by that artery were considered in the model. The blood current within that artery was equalized to the sum of all micro currents in the tissues supplied by that artery. A formula emerged that relates changes in arterial distensibility with heart rate, and also with some particular aspects of microcirculation. Then, that formula was tested with data of distensibilities of the radial and carotid arteries observed at the heart rates of 63, 90, and 110 b.p.m. The formula correctly predicted the trend of decreased distensibility with heart rate for both arteries. Moreover, due to the fact that the carotid artery supplies the brain, and because the Blood–Brain barrier is highly restrictive to colloids in the blood, for the carotid artery the formula predicted a less marked decrease in distensibility than in the case of the radial artery feeding muscle tissue, which has a greater permeability to colloids, a trend that was confirmed by data. It was found that reduction of arterial distensibility with heart rate was greater in arteries that supply end capillaries with high permeability and low reflection coefficients. 相似文献

Background and aims
Gaseous losses of ammonia (NH3) have been observed in citrus orchards when urea is surface-applied to the soils, and this loss might significantly limit the effectiveness of the nitrogen (N) fertilizer. However, a portion of the volatilized NH3 might be absorbed by the plants through the leaves. To quantify the contribution of the leaf absorption of 15NH3, a study with sweet oranges was conducted in two field areas where trees were grown at standard (480 trees ha?1) and high densities (617 trees ha?1).Methods
Plastic trays were filled with soil, covered with mown grass to simulate field management conditions, fertilized with 15N labeled urea (12 atom % excess) and placed under each of three trees in the orchards. This experimental procedure prevented the uptake of N from the labeled urea by the roots. Two weeks after 15N fertilization, the trays were removed from the field, and the soil was homogenized and sampled for chemical analyses. The citrus trees under which the trays were placed were destructively harvested, and the total N concentrations and 15N/14N ratios were determined.Results
After urea application, the NH3 losses peaked within three days and subsequently decreased to negligible amounts after 10 days. The total NH3 losses accounted for 55–82 % of the applied N. Although the NH3 absorption by the citrus leaves was proportional to the tree density in the field, only 3–7 % of the 15NH3 volatilized from the soil was recovered by the citrus trees, and the NH3 absorption was also influenced by the proximity of citrus trees to the site of urea application and the leaf areas of the trees.Conclusions
The citrus trees can absorb the NH3 volatilized from urea, even though, the amount recovered by the trees is small and does not represent a significant proportion of total gaseous N losses, what demonstrates the importance of enhanced N use efficiency practices in field to reduce losses of NH3 when urea is applied to soil surfaces. 相似文献67.

The Eocene Nanjemoy Formation crops out on the Maryland and Virginia Coastal Plain, along the eastern coast of the United States. This formation is composed of sands, silts and clays and is divided into the Potapaco and Woodstock members. Remains of fishes, reptiles, birds, mammals, molluscs, fruits and seeds are common in the Potapaco Member, in addition to vertebrate coprolites. Here, we present an analysis of more than 2000 coprolites from the Fisher/Sullivan Site in Virginia. The chemical composition (phosphatic) and the type of inclusions (fish bones) indicate that only scats of carnivorous animals were preserved. The analysed specimens were grouped into six morphotypes: (1) the cylindrical morphotype is a cylinder with rounded ends; (2) the segmented morphotype is a cylinder segmented with rounded ends, and occasionally one end is concave; (3) the oval morphotype represents a bean‐shaped coprolite; (4) the scroll morphotype is cylindrical to conical in lateral view and has coils seen only at the ends; (5) the folded morphotype is a spiral that is concentrically folded; and (6) the sinuous morphotype is serpentine, with rounded ends. Coprophagy‐related scrape traces occur in different morphotypes and represent both invertebrate burrows and bite traces made by fishes. The mineralogical and chemical analyses indicate an early precipitation of phosphate and pyrite minerals, probably induced by the microbial community. All coprolites at the Fisher/Sullivan Site were produced by fishes: carcharhiniform sharks for the scroll morphotype and lamniform sharks, probably the genus Carcharias, for the folded morphotype; the oval, cylindrical and segmented morphotypes were likely produced by actinopterygian fishes. 相似文献

A case-control study was conducted to determine the presence of Mycobacterium
leprae DNA in nasal secretions of leprosy cases and nonleprosy
individuals in Fortaleza, Brazil. It included 185 cases identified by physicians at
the Dona Libânia National Reference Centre for Sanitary Dermatology (CDERM). A
control group (Co) (n = 136) was identified among individuals from CDERM not
diagnosed as leprosy cases. To augment the spatial analysis of M. leprae specific
repetitive element (RLEP) positive prevalence, an external group (EG) (n = 121), a
convenience sample of healthy students, were included. Polymerase chain reaction for
the RLEP sequence was conducted for all participants. Prevalence of RLEP positivity
for cases and Co were 69.2% and 66.9%, respectively, significantly higher than for EG
(28.1%), and reported elsewhere. Male sex, belonging to a lower socioeconomic status
(D/E), history of a previous contact with a case and being older, were associated
with being a leprosy case. Our geographical analysis demonstrated that the bacillus
is widespread among the healthy population, with clusters of RLEP positive
multibacillary cases concentrated in distinct areas of the city. Our results suggest
that in endemic areas, as in Fortaleza, surveillance for both nonhousehold leprosy
contacts and members of the general population living in cluster areas should be

Resistant hypertension, a complex multifactorial hypertensive disease, is triggered by genetic and environmental factors and involves multiple physiological pathways. Single genetic variants may not reveal significant associations with resistant hypertension because their effects may be dependent on gene-gene or gene-environment interactions. We examined the interaction of angiotensin I-converting enzyme (ACE), angiotensinogen (AGT), and endothelial nitric oxide synthase (NOS3) polymorphisms with environmental factors (gender, age, body mass index, glycemia, total cholesterol, low-density lipoprotein cholesterol, high-density lipoprotein cholesterol, triglycerides, estimated glomerular filtration rate, and urinary sodium excretion) in 70 resistant, 80 well-controlled hypertensive patients, and 70 normotensive controls. All subjects were genotyped for ACE insertion/deletion (rs1799752); AGT M235T (rs699), and NOS3 Glu298Asp (rs 1799983). Multifactorial associations were tested using two statistical methods: the traditional parametric method (adjusted logistic regression analysis) and gene-gene and gene-environment interactions evaluated by multifactor dimensionality reduction analyses. While adjusted logistic regression found no significant association between the studied polymorphisms and controlled or resistant hypertension, the multifactor dimensionality reduction analyses showed that carriers of the AGT 235T allele were at increased risk for resistant hypertension, especially if they were older than 50 years. The AGT 235T allele constituted an independent risk factor for resistant hypertension. 相似文献
