A novel approach is developed for mathematically modeling the variability observed in experimentally determined elastic moduli of longitudinally oriented fibrous tissues such as ligaments and tendons. The elastic modulus of these tissues is modeled with a rule of mixtures (ROM) where each parameter (fibril and matrix moduli and fibril volume fraction) is assumed to be an independent random variable. A joint density function formed from the independent densities results in a probabilistic ROM (pROM). This pROM is used to generate a distribution of moduli which agrees well with moduli determined from tests of rabbit medial collateral ligaments (Woo and Ohland, 1994, Unpublished experimental data as gift). Minimizing the error between the pROM and experimental distributions resulted in an integrated error of 9% for a constrained set of independent distribution parameters derived from the literature. This pROM thus incorporates microstructural observations (fibril and matrix moduli and fibril volume fraction) to partially explain the experimentally observed variability in a macroscopic property (tissue modulus).  相似文献

Aqueous extracts of Ailanthus altissima bark and foliage were previously shown to be toxic to other plants. Using bioassay-directed fractionation, I isolated the phytotoxic compound from A. altissima root bark and identified it to be ailanthone, a quassinoid compound having molecular mass of 376. Ailanthone was highly phytotoxic, with concentrations of 0.7 ml/L causing 50% inhibition of radicle elongation in a standardized bioassay with garden cress (Lepidium sativum) seeds. Ailanthone exhibited potent pre- and postemergence herbicidal activity in greenhouse trials. Postcmergence activity was especially striking; even the lowest application rate (0.5 kg/ha) caused complete mortality of five of the seven plant species tested within 5 d of treatment. In contrast, the highest application rate (8 kg/ha) did not cause any detectable injury to A. altissima seedlings, indicating the presence of a protective mechanism in the producer species to prevent autotoxicity. Ailanthone was rapidly detoxified in field soil as a result of microbial activity. Applications of ailanthone equivalent to 0.5 and 4.0 kg/ha completely lost their phytotoxicity within ≤5 d when incubated in the presence of nonsterile soil. When incubated with sterile soil under identical conditions, however, ailanthone remained highly phytotoxic throughout the 21-d duration of the investigation. The high level of postemergence herbicidal activity in conjunction with its rapid biodegradation in soil suggest ailanthone may have potential for development as a natural-product herbicide.  相似文献

Bacteria able to grow on purified natural rubber in the absence of other organic carbon sources were isolated from soil. Ten isolates reduced the weight of vulcanized rubber from latex gloves by >10% in 6 weeks. Scanning electron microscopy clearly revealed the ability of the microorganisms to colonize, penetrate, and dramatically alter the physical structure of the rubber. The rubber-metabolizing bacteria were identified on the basis of fatty acid profiles and cell wall characteristics. Seven isolates were strains of Streptomyces, two were strains of Amycolatopsis, and one was a strain of Nocardia.  相似文献

Extracts of Ailanthus altissima stem bark were evaluated for herbicidal effects under field conditions in two outdoor trials. Previous investigations had shown A. altissima bark, extracted with methanol, yielded a strongly phytotoxic extract that contained ailanthone as one of the major herbicidal compounds. The first field trial investigated the level of activity and selectivity of the extract. A. altissima bark extract was sprayed post-emergence onto 17 species of weeds and crops at rates of 366, 177, 93, 47, 23, and 0 kg ha–1. These application rates provided herbicidal activity equivalent to 4.5, 2.2, 1.1, 0.6, 0.3, and 0.0 kg of pure ailanthone per hectare, based on the results of a laboratory bioassay of extract and pure ailanthone. Strong herbicidal effects were observed within several days. Even the lowest rate caused mortality and injury in excess of 50% for nine of the 17 species, and a significant reduction in shoot biomass for 13 species. The second field trial tested the ability of bark extract to control weeds under field conditions with horticultural crops (bush bean, cauliflower, sweet corn, tomato). A. altissima bark extract was sprayed post-emergence at rates of 99, 50, 26, 13, and 0 kg ha–1, providing herbicidal activity equivalent to 1.1, 0.6, 0.3, 0.14, and 0.0 kg of pure ailanthone per hectare. Extract treatment provided partial weed control (greatest reduction in weed biomass was 40%), but also caused serious crop injury. Bush bean was the only crop that showed a significant increase in shoot biomass and fruit yield, compared to the non-weeded control. None of the crops, regardless of application rate, showed a level of shoot biomass or fruit yield comparable to the hand-weeded control. The herbicidal effects of A. altissima bark extract declined within the first few weeks after application, supporting previous evidence that ailanthone is rapidly degraded under field conditions.  相似文献

Effects of vasoconstriction on the distribution of perfusion among alveoli are not well understood. To address this, we used a new method we developed to determine how microvascular perfusion distribution was affected by a potent vasoconstrictor, the thromboxane receptor analog U-46619. Our method was to infuse 4-microm-diameter fluorescent latex microspheres into the circulation of isolated rat lungs vasoconstricted with U-46619. We used a confocal microscope to image trapping patterns of the particles in dried sections of the lungs and then used dispersion index analysis to quantify the particle patterns in the images, which encompassed approximately 2,000 alveoli. Dispersion indexes revealed significantly more particle clustering (inhomogeneous distribution) in vasoconstricted lungs than in normal flow controls or in controls in which flow was reduced by either lowering pulmonary arterial pressure or raising left atrial pressure. These results suggest that vasoconstriction occurred in the microvessels themselves, which are much smaller vessels than those previously thought to be capable of vasoconstriction.  相似文献

Chronic wasting disease (CWD) is a transmissible spongiform encephalopathy (TSE) of cervids now detected in 19 states of the United States, three Canadian provinces, and South Korea. Whether noncervid species can be infected by CWD and thereby serve as reservoirs for the infection is not known. To investigate this issue, we previously used serial protein misfolding cyclic amplification (sPMCA) to demonstrate that CWD prions can amplify in brain homogenates from several species sympatric with cervids, including prairie voles (Microtus ochrogaster) and field mice (Peromyscus spp.). Here, we show that prairie voles are susceptible to mule deer CWD prions in vivo and that sPMCA amplification of CWD prions in vole brain enhances the infectivity of CWD for this species. Prairie voles inoculated with sPMCA products developed clinical signs of TSE disease approximately 300 days prior to, and more consistently than, those inoculated with CWD prions from deer brain. Moreover, the deposition patterns and biochemical properties of protease-resistant form of PrP (PrP(RES)) in the brains of affected voles differed from those in cervidized transgenic (CerPrP) mice infected with CWD. In addition, voles inoculated orally with sPMCA products developed clinical signs of TSE and were positive for PrP(RES) deposition, whereas those inoculated orally with deer-origin CWD prions did not. These results demonstrate that transspecies sPMCA of CWD prions can enhance the infectivity and adapt the host range of CWD prions and thereby may be useful to assess determinants of prion species barriers.  相似文献

Chronic wasting disease (CWD) is a fatal disease of deer, elk, and moose transmitted through direct, animal-to-animal contact, and indirectly, via environmental contamination. Considerable attention has been paid to modeling direct transmission, but despite the fact that CWD prions can remain infectious in the environment for years, relatively little information exists about the potential effects of indirect transmission on CWD dynamics. In the present study, we use simulation models to demonstrate how indirect transmission and the duration of environmental prion persistence may affect epidemics of CWD and populations of North American deer. Existing data from Colorado, Wyoming, and Wisconsin's CWD epidemics were used to define plausible short-term outcomes and associated parameter spaces. Resulting long-term outcomes range from relatively low disease prevalence and limited host-population decline to host-population collapse and extinction. Our models suggest that disease prevalence and the severity of population decline is driven by the duration that prions remain infectious in the environment. Despite relatively low epidemic growth rates, the basic reproductive number, R(0), may be much larger than expected under the direct-transmission paradigm because the infectious period can vastly exceed the host's life span. High prion persistence is expected to lead to an increasing environmental pool of prions during the early phases (i.e. approximately during the first 50 years) of the epidemic. As a consequence, over this period of time, disease dynamics will become more heavily influenced by indirect transmission, which may explain some of the observed regional differences in age and sex-specific disease patterns. This suggests management interventions, such as culling or vaccination, will become increasingly less effective as CWD epidemics progress.  相似文献
