Oligodeoxyribonucleotide mutagenesis has been used to produce a G----A mutation at nucleotide 557 of the phi X174 genome. This changes the ribosome-binding sequence GAGG of gene E to GAAG without affecting the amino acid, glutamine, encoded by the overlapping gene D. The phi X174rb(E)557 mutant does not lyse infected Escherichia coli C and therefore results in the accumulation of a large number intracellular mature phage particles. Thus, the mutation inactivates production of the gene E lytic product, presumably by blocking translation of gene E, without affecting other phage functions.  相似文献

Two methods are described for detecting less than 1 microgram of highly glycosylated proteins, such as mucins, on sodium dodecyl sulfate-polyacrylamide gels. They combine commonly employed periodic acid-Schiff (PAS) and Alcian blue dyes with silver stain. Carbohydrate prestaining renders mucins more cationic and favors greater complexation with ionic silver. Comparisons of different mucin samples stained either with PAS-silver or alcian blue-silver indicate differential staining between the two techniques. Such differences may, in part, be due to an affinity of Alcian blue for sulfated glycoproteins. These two staining protocols when used in conjunction with silver staining alone are particularly valuable for assessing sample purity and for detecting contaminating proteins during mucin purification protocols.  相似文献

Reliable signal transmission constitutes a key requirement for neural circuit function. The propagation of synchronous pulse packets through recurrent circuits is hypothesized to be one robust form of signal transmission and has been extensively studied in computational and theoretical works. Yet, although external or internally generated oscillations are ubiquitous across neural systems, their influence on such signal propagation is unclear. Here we systematically investigate the impact of oscillations on propagating synchrony. We find that for standard, additive couplings and a net excitatory effect of oscillations, robust propagation of synchrony is enabled in less prominent feed-forward structures than in systems without oscillations. In the presence of non-additive coupling (as mediated by fast dendritic spikes), even balanced oscillatory inputs may enable robust propagation. Here, emerging resonances create complex locking patterns between oscillations and spike synchrony. Interestingly, these resonances make the circuits capable of selecting specific pathways for signal transmission. Oscillations may thus promote reliable transmission and, in co-action with dendritic nonlinearities, provide a mechanism for information processing by selectively gating and routing of signals. Our results are of particular interest for the interpretation of sharp wave/ripple complexes in the hippocampus, where previously learned spike patterns are replayed in conjunction with global high-frequency oscillations. We suggest that the oscillations may serve to stabilize the replay.  相似文献

Developmental toxicity evaluation of berberine in rats and mice   总被引:1,自引:0,他引:1  
BACKGROUND: Berberine, a plant alkaloid, is found in some herbal teas and health-related products. It is a component of goldenseal, an herbal supplement. Berberine chloride dihydrate (BCD) was evaluated for developmental toxicity in rats and mice. METHODS: Berberine chloride dihydrate was administered in the feed to timed-mated Sprague-Dawley (CD) rats (0, 3,625, 7,250, or 14,500 ppm; on gestational days [GD] 6-20), and Swiss Albino (CD-1) mice (0, 3,500, 5,250, or 7,000 ppm; on GD 6-17). Ingested doses were 0, 282, 531, and 1,313 mg/kg/day (rats) and 0, 569, 841, and 1,155 mg/kg/day (mice). RESULTS: There were no maternal deaths. The rat maternal lowest observed adverse effect level (LOAEL), based on reduced maternal weight gain, was 7,250 ppm. The rat developmental toxicity LOAEL, based on reduced fetal body weight per litter, was 14,500 ppm. In the mouse study, equivocal maternal and developmental toxicity LOAELs were 5,250 ppm. Due to scattering of feed in the high dose groups, a gavage study at 1,000 mg/kg/day was conducted in both species. CONCLUSIONS: In rats, maternal, but not fetal adverse effects were noted. The maternal toxicity LOAEL remained at 7,250 ppm (531 mg/kg/day) based on the feed study and the developmental toxicity NOAEL was raised to 1,000 mg/kg/day BCD based on the gavage study. In the mouse, 33% of the treated females died. Surviving animals had increased relative water intake, and average fetal body weight per litter decreased 5-6% with no change in live litter size. The maternal toxicity LOAEL remained at 5,250 ppm (841 mg/kg/day) BCD, based on increased water consumption. The developmental toxicity LOAEL was raised to 1,000 mg/kg/day BCD based on decreased fetal body weight.  相似文献

Gas exchange is generally regarded to occur between the leaf interior and ambient air, i.e. in vertical (anticlinal) directions of leaf blades. However, inside homobaric leaves, gas movement occurs also in lateral directions. The aim of the present study was to ascertain whether lateral CO2 diffusion affects leaf photosynthesis when illuminated leaves are partially shaded. Measurements using gas exchange and chlorophyll fluorescence imaging techniques were performed on homobaric leaves of Vicia faba and Nicotiana tabacum or on heterobaric leaves of Glycine max and Phaseolus vulgaris. For homobaric leaves, gas exchange inside a clamp-on leaf chamber was affected by shading the leaf outside the chamber. The quantum yield of photosystem II (Phi(PSII)) was highest directly adjacent to a light/shade border (LSB). Phi(PSII) decreased in the illuminated leaf parts with distance from the LSB, while the opposite was observed for nonphotochemical quenching. These effects became most pronounced at low stomatal conductance. They were not observed in heterobaric leaves. The results suggest that plants with homobaric leaves can benefit from lateral CO2 flux, in particular when stomata are closed (e.g. under drought stress). This may enhance photosynthetic, instead of nonphotochemical, processes near LSBs in such leaves and reduce the photoinhibitory effects of excess light.  相似文献

Despite its potential importance for the biological control of European rabbits, relatively little is known about the evolution and molecular epidemiology of rabbit calicivirus Australia 1 (RCV-A1). To address this issue we undertook an extensive evolutionary analysis of 36 RCV-A1 samples collected from wild rabbit populations in southeast Australia between 2007 and 2009. Based on phylogenetic analysis of the entire capsid sequence, six clades of RCV-A1 were defined, each exhibiting strong population subdivision. Strikingly, our estimates of the time to the most recent common ancestor of RCV-A1 coincide with the introduction of rabbits to Australia in the mid-19th century. Subsequent divergence events visible in the RCV-A1 phylogenies likely reflect key moments in the history of the European rabbit in Australia, most notably the bottlenecks in rabbit populations induced by the two viral biocontrol agents used on the Australian continent, myxoma virus and rabbit hemorrhagic disease virus (RHDV). RCV-A1 strains therefore exhibit strong phylogeographic separation and may constitute a useful tool to study recent host population dynamics and migration patterns, which in turn could be used to monitor rabbit control in Australia.Lagoviruses form a genus within the Caliciviridae family of RNA viruses (16). All representatives of this genus are highly species specific and infect only their respective hosts, i.e., rabbits and hares. The prototype species of the genus Lagovirus is rabbit hemorrhagic disease virus (RHDV), which was first described in China in 1984 when a severe infectious necrotizing hepatitis with mortality rates up to 90% was observed in angora rabbits (22). However, phylogenetic analysis indicates that the pathogenic strains of RHDV likely evolved from nonpathogenic lagoviruses several decades before they were initially described (20). Today, RHDV is found on most continents, causing ongoing damage to the rabbit meat industry (24) and threatening wild native rabbit populations in Europe. On the Iberian peninsula the rabbit is considered an endangered species and is itself a staple food of endangered predators, such as the imperial eagle and the Iberian lynx (10).In marked contrast, since 1995 Australia has been using RHDV as a successful viral biocontrol agent for rabbits (7), which cause severe environmental and economic damage in this country. The use of a viral biocontrol agent for a vertebrate species has been and remains controversial, but there is little doubt that since its release RHDV has generated close to $6 billion of savings to the Australian agricultural industry (41), as well as some much needed relief for the regeneration of many native plant species (35).Notably, RHDV-induced mortality is lower in certain areas of Australia, namely, the cooler and more humid southeast region of the continent, which is believed to be in part due to the presence of related but nonpathogenic lagoviruses circulating in the population (8). Recently, such a virus was identified in Australian wild rabbits (38). This new member of the genus Lagovirus, termed rabbit calicivirus Australia 1 (RCV-A1), is a nonpathogenic virus causing a predominantly enteric infection in rabbits. Other benign or moderately pathogenic RCV strains have also been described in the United States and Europe (1, 5, 14, 15). While the Italian nonpathogenic RCV provides complete cross-immunity to RHDV, only partial protection is conveyed by the Australian virus RCV-A1 (37) although this may be sufficient to reduce overall RHDV-induced mortality.Like all lagoviruses, RCV-A1 has a single-stranded positive-sense RNA genome of approximately 7.5 kb that is polyadenylated and has a viral protein (VpG) covalently bound to its 5′ end (25). The genome is organized into two open reading frames (ORFs) (6, 39). ORF1, which represents the majority (7 kb) of the genome, encodes a polyprotein that is auto-proteolytically cleaved during posttranslational processing into several smaller proteins, including the helicase, protease, and polymerase (26). In contrast, ORF2 is only 351 nucleotides (nt) long, and the function of its VP10 gene product is unknown although it is present in small amounts in the virion (39).Given the potential interactions between RCVs and pathogenic RHDV, which could impact rabbit survival in both Australia and Europe, it is clearly important to understand the evolutionary history, genetic diversity, and geographic distribution of RCV-A1 and other benign lagoviruses. In Australia, growing reports of rising rabbit numbers mean that there is a clear need for improved strategies to monitor and control rabbit populations. In addition, because RCV-A1 does not cause any fitness decrease in its rabbit host (37), it is possible that this virus could be used as a population genetic marker to track movements and changes in the distribution of the European rabbit across the Australian continent. Indeed, other microorganisms have proven to be useful indicators of the population dynamics of their hosts (3, 42). Microbial markers are particularly informative for the study of population processes in the very recent past since genetic changes will not have had sufficient time to be recorded in the more slowly evolving host genome (2).The host-pathogen interaction of Australian rabbits and their viruses is unique in that both were introduced only once (or possibly a few times in a limited period of time). In addition, 150 years of historical records are available documenting the introduction, spread, and control efforts of rabbits in Australia. Herein, we explore the evolutionary history and dynamics of RCV-A1 in Australia, with a particular focus on revealing the phylogeographic distribution of the virus across this continent, the evolutionary processes that have shaped its diversity, and its suitability as a marker to study host distribution and migration patterns.  相似文献

The effect of oxygen on squalene epoxidase activity in Saccharomyces cerevisiae was investigated. In cells grown in standing cultures, the epoxidase was localized mainly in the "mitochondrial" fraction. Upon aeration, enzyme activity increased and the newly formed enzyme was associated with the "microsomal" fraction. At 0.03% (vol/vol) oxygen, epoxidase levels doubled, whereas the ergosterol level was only slightly increased. Cycloheximide inhibited the increase in epoxidase under these conditions. An apparent Km for oxygen of 0.38% (vol/vol) was determined from a crude particulate preparation for the epoxidase.  相似文献

The inhibition patterns of rabbit sera (RS1 & RS2) from two different rabbits on the photosynthetic electron transport of isolated spinach thylakoids were studied. Fifty l of RSI were required for 100% inhibition of a H2O MV/O2 reaction, while only 10 l of a 1:10 dilution of RS2 were needed for 100% inhibition. The RS2 serum was greatly hemolyzed. The -globulin fraction from purified rabbit serum (RS1) did not inhibit photosynthetic electron transport, indicating that the antibody fraction of the rabbit serum does not contain the inhibitor. It appears that the inhibitor is from the hemolyzed red blood cells. Rabbit sera added prior to chloroplast illumination caused no inhibition, while addition of rabbit sera during illumination inhibited a H2O MV/O2 reaction within 1–3s. Aminotriazole, a catalase inhibitor, did not affect the efficacy of the rabbit sera indicating that the unknown rabbit serum inhibitor is not catalase. Various Hill reactions were employed to determine the site of inhibition. Rabbit sera inhibited the following reactions: DHQ/DCMU MV/O2, DAD/Asc/DBMIB MV/O2, and DCIP/Asc/DBMIB MV/O2. Rabbit sera did not inhibit a H2O DADox reaction indicating that inhibition is on the reducing side of PSI. However, a H2O Fd/NADP+ reaction was not inhibited by rabbit sera. NADP did not interfere with the ability of RS2 to inhibit a MV-mediated Mehler reaction. In simultaneously measured assays of Fd-mediated O2 and NADP+ reductions, RS2 serum inhibited the reduction of O2 by ferredoxin without inhibiting the reduction of NADP+. Two oxazolone-derived potential ligands with enethioether substituents have been synthesized that differ by the terminal thioether moiety (S-Et in L1, S-C6H4(OMe)-2 in L2). Both L1 and L2 behave as bidentate {NS} chelate ligands to form stable complexes with copper(I) triflate that crystallize as dimeric complexes [L2Cu2(OTf)2] (4 and 5) featuring a central {Cu2S2} diamond core with distinctly different Cu-S bonds. L1 as well as 4 and 5 have been characterized by single crystal X-ray diffraction. NMR spectroscopy including 1H and 19F DOSY experiments reveals that 4 and 5 dissociate into monomeric species [LCu(OTf)] (4′ and 5′) in CDCl3 solutions. 4′ and 5′ retain the {NS} binding motif of the oxazolone-derived ligands, but are in slow equilibrium with their {OS} isomers 4″ and 5″ that result from E/Z isomerization of the exocyclic enethioether double bond.  相似文献
