Five-week-old wheat plants were exposed, under controlled environmental conditions, to 60 nl 1?115NO2 or to purified air. After 48 and 96 h of exposure, leaves, stalks and roots were analysed for 15N-enrichment in α-amino nitrogen of soluble, free amino acids. In addition, the in vitro nitrate reductase (NR, EC 1.6.6.1) and nitrite reductase (NIR, EC 1.7.7.1) activities were determined in the leaves. NR activity in the leaves decreased continously during the 96-h exposure to purified air. In the leaves exposed to 15NO2, NR activity increased within the first 24 h, then decreased, and reached the level of controls after 96 h. NiR activity in leaves exposed to purified air was almost constant during the 96-h exposure. In leaves exposed to 15NO2, NiR activity increased within the first 48 h, then decreased, and reached the level of controls after 72 h, Exposure to 15NO2 enhanced the total content of soluble, free amino acids in all tissues analysed. Most of this increase was attributed to Glu in the leaves and to Asn plus Gln the α-amino group of soluble, free amino acids was observed in the leaves, the lowest enrichment in the roots. The main labelled amino compounds were Glu (with 8.0%15N enrichment compared to the control), γ-aminobutyric acid (GABA; 7.9%), Ala (7.2%). Ser (6.8%), Asp (5.5%) and Gln (4.6%). Appreciable incorporation of 15 into Asn was not found. After 96 h exposure to 15NO2 the 15N enrichment in the α-amino group of soluble, free amino acids in the leaves declined as compared to the values obtained after 48 h fumigation. The possible pathway and the time course of 15N incorporation into soluble, free amino acids from the 15NO2 absorbed are discussed.  相似文献

Abstract The gene coding for the β-subunit of the sodium ion translocating ATP synthase from Propionigenium modestum was cloned and sequenced. The predicted amino acid sequence resembles that of the β-subunits from proton-translocating ATP synthases. The same conserved regions are found in both types of ATP synthases. This is a good indication that the β-subunits of the proton and sodium ion translocating ATP synthases have evolved from a common ancestor.  相似文献

参照植物根尖细胞学研究的方法标准,对香薷属3种(5个居群)植物进行核形态学分析。结果表明:(1)从染色体数目看,密花香薷2居群染色体数目2n=16;野苏子2居群染色体数目2n=20,染色体数目和倍性与前人报道的一致;毛穗香薷染色体数目2n=10为首次报道。(2)聚类分析结果显示,3种(5居群)植物中野苏子和密花香薷亲缘关系较近;结合现有报道数据分析表明,该属植物仅有2种倍性(二倍体和四倍体),且二倍体占主导地位。(3)核型参数分析表明:密花香薷的稻城无名山居群1核型公式为2n=2x=16=14m+2sm,居群2为2n=2x=16=16m,着丝粒指数(CI)分别为39.57和42.32,不对称系数AI值分别为2.75和2.87,核型不对称性都为1A型;毛穗香薷的核型公式为2n=2x=10=10m,着丝粒指数(CI)为41.76,不对称系数AI值为5.25,核型不对称性为1B型;野苏子的昆明西山居群核型公式为2n=2x=20=14m+6sm,聂拉木樟木沟居群为2n=2x=20=16m+4sm,着丝粒指数(CI)分别为38.49和40.97,不对称系数AI值为4.20和4.30,核型不对称性为1B型和2B型。  相似文献

Multiple sclerosis is a chronic neuroinflammatory demyelinating disorder of the central nervous system with a strong neurodegenerative component. While the exact etiology of the disease is yet unclear, autoreactive T lymphocytes are thought to play a central role in its pathophysiology. MS therapy is only partially effective so far and research efforts continue to expand our knowledge on the pathophysiology of the disease and to develop novel treatment strategies. Experimental autoimmune encephalomyelitis (EAE) is the most common animal model for MS sharing many clinical and pathophysiological features. There is a broad diversity of EAE models which reflect different clinical, immunological and histological aspects of human MS. Actively-induced EAE in mice is the easiest inducible model with robust and replicable results. It is especially suited for investigating the effects of drugs or of particular genes by using transgenic mice challenged by autoimmune neuroinflammation. Therefore, mice are immunized with CNS homogenates or peptides of myelin proteins. Due to the low immunogenic potential of these peptides, strong adjuvants are used. EAE susceptibility and phenotype depends on the chosen antigen and rodent strain. C57BL/6 mice are the commonly used strain for transgenic mouse construction and respond among others to myelin oligodendrocyte glycoprotein (MOG). The immunogenic epitope MOG35-55 is suspended in complete Freund''s adjuvant (CFA) prior to immunization and pertussis toxin is applied on the day of immunization and two days later. Mice develop a "classic" self-limited monophasic EAE with ascending flaccid paralysis within 9-14 days after immunization. Mice are evaluated daily using a clinical scoring system for 25-50 days. Special considerations for care taking of animals with EAE as well as potential applications and limitations of this model are discussed.  相似文献

A series of three α‐ and three β‐fluorinated representatives of the family of cinnamate‐derived odorants (cinnamaldehyde ( 1 ), cinnamyl alcohol ( 2 ), and ethyl cinnamate ( 3 )) as used as fragrance ingredients is described. Olfactive evaluation shows that the fluorinated compounds exhibit a similar odor profile to their parent compounds, but the olfactive detection thresholds are clearly higher. In vitro evaluation of the skin sensitizing properties with three different assays indicates that α‐fluorination of Michael acceptor systems 1 and 3 slightly improves the skin sensitization profile. α‐Fluorocinnamyl alcohol 2b is a weaker skin sensitizer than cinnamyl alcohol 2a by in vitro tests and the fluorinated product drops below the sensitization threshold of the KeratinoSens® assay. On the other hand, β‐fluorination of compounds 1  –  3 results in highly reactive products which display a worsened in vitro skin sensitization profile.  相似文献

In contrast to 16:3 plants like rapeseed (Brassica napus), which contain alpha-linolenic acid (18:3(Delta9,12,15)) and hexadecatrienoic acid (16:3(Delta7,10,13)) as major polyunsaturated fatty acids in leaves, the silica-less diatom Phaeodactylum tricornutum contains eicosapentaenoic acid (EPA; 20:5(Delta5,8,11,14,17)) and a different isomer of hexadecatrienoic acid (16:3(Delta6,9,12)). In this report, we describe the characterization of two cDNAs having sequence homology to Delta12-fatty acid desaturases from higher plants. These cDNAs were shown to code for a microsomal and a plastidial Delta12-desaturase (PtFAD2 and PtFAD6, respectively) by heterologous expression in yeast (Saccharomyces cerevisiae) and Synechococcus, respectively. Using these systems in the presence of exogenously supplied fatty acids, the substrate specificities of the two desaturases were determined and compared with those of the corresponding rapeseed enzymes (BnFAD2 and BnFAD6). The microsomal desaturases were similarly specific for oleic acid (18:1(Delta9)), suggesting that PtFAD2 is involved in the biosynthesis of EPA. In contrast, the plastidial desaturase from the higher plant and the diatom clearly differed. Although the rapeseed plastidial desaturase showed high activity toward the omega9-fatty acids 18:1(Delta9) and 16:1(Delta7), in line with the fatty acid composition of rapeseed leaves, the enzyme of P. tricornutum was highly specific for 16:1(Delta9). Our results indicate that in contrast to EPA, which is synthesized in the microsomes, the hexadecatrienoic acid isomer found in P. tricornutum (16:3(Delta6,9,12)) is of plastidial origin.  相似文献

HER2-testing in breast and gastric cancers is mandatory for the treatment with trastuzumab. We hypothesized that imaging mass spectrometry (IMS) of breast cancers may be useful for generating a classifier that may determine HER2-status in other cancer entities irrespective of primary tumor site. A total of 107 breast (n = 48) and gastric (n = 59) cryo tissue samples was analyzed by IMS (HER2 was present in 29 cases). The obtained proteomic profiles were used to create HER2 prediction models using different classification algorithms. A breast cancer proteome derived classifier, with HER2 present in 15 cases, correctly predicted HER2-status in gastric cancers with a sensitivity of 65% and a specificity of 92%. To create a universal classifier for HER2-status, breast and nonbreast cancer samples were combined, which increased sensitivity to 78%, and specificity was 88%. Our proof of principle study provides evidence that HER2-status can be identified on a proteomic level across different cancer types suggesting that HER2 overexpression may constitute a unique molecular event independent of the tumor site. Furthermore, these results indicate that IMS may be useful for the determination of potential drugable targets, as it offers a quicker, cheaper, and more objective analysis than the standard HER2-testing procedures immunohistochemistry and fluorescence in situ hybridization.  相似文献

The small GTPases Rho and Rac regulate actin filament assembly and the formation of integrin adhesion complexes to produce stress fibers and lamellipodia, respectively, in mammalian cells. Although numerous candidate effectors that might mediate these responses have been identified using the yeast two-hybrid and affinity purification techniques, their cellular roles remain unclear. We now describe a biological assay that allows components of the Rho and Rac signaling pathways to be identified. Permeabilization of serum-starved Swiss 3T3 cells with digitonin in the presence of guanosine 5′-O-(3-thiotriphosphate) (GTPγS) induces both actin filament and focal adhesion complex assembly through activation of endogenous Rho and Rac. These responses are lost when GTPγS is added 6 min after permeabilization, but can be reconstituted using concentrated cytosolic extracts. We have achieved a 10,000-fold purification of the activity present in pig brain cytosol and protein sequence analysis shows it to contain moesin. Using recombinant proteins, we show that moesin and its close relatives ezrin and radixin can reconstitute stress fiber assembly, cortical actin polymerization and focal complex formation in response to activation of Rho and Rac.  相似文献

PbTe alloys have been the most efficient materials for thermoelectric power generation since the 1950s. In addition to the observed lattice thermal conductivity reduction by alloy scattering, recent efforts targeting electronic‐transport enhancement through band structure engineering have demonstrated a design route to achieve extraordinary thermoelectric performance. Guided by these recent results, the current work discusses the influence of alloying CdTe in p‐type PbTe where the solubility is strongly temperature‐dependent. Beneficial changes to the band structure and effective scattering of phonons are concurrently inferred when the PbTe–CdTe precipitate composite is expected to become a solid solution at high temperature, enhancing the peak thermoelectric figure of merit to ~1.7. This work emphasizes the importance of solid solution alloys of PbTe for thermoelectric power‐generation applications and also provides an upper limit of about 1 micrometer to the average distance between interfaces above which phonon scattering by composite interfaces is ineffectual.  相似文献
