Vegetative cells of Pelagomonas calceolata Andersen & Saunders were confirmed to possess a reduced flagellar apparatus, consisting of a single basal body/flagellum that is not accompanied by either flagellar roots or a barren basal body. Just prior to division, the parental flagellum retracts (or is abscised) as two new basal bodies/flagella arise de novo. During cytokinesis the parental basal body segregates with a new basal body/flagellum, briefly producing a progeny cell typical of other known uniflagellates (i.e. containing a basal body/flagellum and accompanying barren basal body). The parental basal body then disintegrates or "transforms" out of existence, leaving both progeny cells with a single basal body/flagellum (i.e. neither progeny cell possesses any vestige of a mature flagellum/basal body ). Pelagomonas calceolata belongs to a lineage of chromophyte algae characterized by having a reduced flagellar apparatus, but it is the only known species, not only in this lineage but among all eukaryotes, to have undergone the complete elimination of the mature flagellum /basal body .  相似文献

The triorganotin compounds triphenyltin chloride and tributyltin chloride have been known as inhibitors of the transmembrane proton channel forming F0-domain of ATPases at micromolar concentrations. We show that these compounds at higher concentrations (10–100 µM) also inhibit uncoupled electron transport in chloroplasts within the low potential chain of the cytochrome bf complex. They cause high levels of transiently reduced cytochrome b563 as they decelerate the reoxidation process in flash illuminated chloroplasts. At the same time they slow down the flash induced slow electrogenic step generated at the cytochrome bf complex. The inhibitory effect of triphenyltin chloride on cytochrome b563 turnover in chloroplasts is comparable to that of the Qn-inhibitor MOA-stilbene, with even less side effects on the high potential chain. Studies on the isolated bf complex suggest different binding sites for triorganotins and the quinone analogue type Qn-inhibitors. The results are interpreted within the framework of the modified Q-cycle model by a putative organotin sensitive proton translocating site which enables proton transfer from the outer aqueous face of the membrane to the hydrophobic quinone reduction site within the complex. Hence, cytochrome b563 oxidation and plastoquinone reduction may be inhibited as a consequence of proton transfer being suppressed by triorganotins. In analogy, the previously described inhibitory effect of Val/K+ at the n-side of the cytochrome bf complex [Klughammer and Schreiber (1993) FEBS 336: 491–495] may be rationalised by binding of the cyclic depsipeptide at the entrance of the proton path to the Qn-site.  相似文献

The silkmoth chorion has provided a stimulating model for the study of evolution and developmental regulation of gene families. Previous attempts at inferring relationships among chorion sequences have been based on pairwise comparisons of overall similarity, a potentially problematic approach. To remedy this, we identified the alignable regions of low sequence variability and then analyzed this restricted database by parsimony and neighbor-joining methods. At the deepest level, the chorion sequence tree is split into two branches, called "alpha" and "beta." Within each branch, early- and late-expressing genes each constitute monophyletic groups, while the situation with middle-expressing genes remains uncertain. The HcB gene family appears to be the most basal beta-branch group, but this conclusion is qualified because the effect of gene conversion on branching order is unknown. Previous studies by Eickbush and colleagues have strongly suggested that ErA, HcA, and HcB families undergo gene conversion within a gene family, whereas the ErB family does not. The occurrence of conversion correlates with a particular tree structure; namely, branch lengths are much greater at the base of the family than at higher internodes and terminal branches. These observations raise the possibility that chorion gene families are defined by gene conversion events (reticulate evolution) rather than by descent with modification (synapomorphy).   相似文献

Slow cortical potentials (SCPs) are considered to reflect the regulation of attention resources and cortical excitability in cortical neuronal networks. Impaired attentional functioning, as found in patients with schizophrenic disorders, may covary with impaired SCP regulation. This hypothesis was tested using a self-regulation paradigm. Twelve medicated male schizophrenic inpatients and 12 healthy male controls received continuous feedback of their SCPs, during intervals of 8 s each, by means of a visual stimulus (a stylized rocket) moving horizontally across a TV screen. The position of the feedback stimulus was a linear function of the integrated SCP at each point in time during the feedback interval. Subjects were required to increase or reduce negative SCPs (referred to pretrial baseline) depending on the presentation of a discriminative stimulus. The correct response was indicated by the amount of forward movement of the feedback stimulus and by monetary rewards. Schizophrenics participated in 20 sessions (each comprising 110 trials), while controls participated in 5 sessions. Compared with the healthy controls, schizophrenics showed no significant differentiation between negativity increase and negativity suppression during the first sessions. However, in the last 3 sessions, patients achieved differentiation similar to controls, demonstrating the acquisition of SCP control after extensive training.  相似文献

Water quality and algal populations in the Vaal River Barrage Reservoir have been monitored extensively for many decades, because of its importance as a water source for the most densely populated area in South Africa. Although Aulacoseira granulata (Ehrenberg) Simonsen is frequently found and at it times dominates algal assemblages in the Barrage, Aulacoseira ambigua (F.Meister) Tuji & D.M.Williams has never before been recorded at this locality. During a countrywide proficiency-testing scheme for algae counting, coordinated by Rand Water, spiral-shaped diatom colonies with distinctly curved cells were detected during May 2015. Upon investigation, it was found that the spiral colonies were Aulacoseira ambigua f. japonica, its presence in the fresh waters of South Africa being recorded for the first time. Since their first appearance during May 2015, colonies of Aulacoseira ambigua f. japonica have been constantly present in the surface waters of the Barrage. Their presence at this site can be linked to increasing eutrophication in the Vaal River, because the spiral form is known to prefer eutrophic conditions. Centric diatoms rarely form spiral colonies and there is some confusion in the literature as to the identity of this particular form, therefore in this paper we discuss this taxon, its nomenclature and ecological significance.  相似文献

Plasma membrane vesicles (PMVs) were isolated from Cyanophora paradoxa Korshikov (Claucocystophyta) and purified by differential and sucrose density gradient centrifugation. PMVs were identified ultrastructurally and biochemically using marker enzymes. Analysis of monosaccharides revealed a sugar content of 37.3% of dry weight of delipidated PMVs with high amounts of fucose (28 mol% of total carbohydrate), galactose (23%), mannose (21%) and the minor constituents xylose, rhamnose, glucose and N-acetylglucosamine. Proteins were analyzed by SDS-PACE. More than 20 polypeptides were resolved and several were identified as glycopeptides, as shown by PAS staining and lectin blots. Fucose linked α-1→6 to N-acetylglucosamine (AAA-binding), mannose linked α-1→2,3,6 to mannose (CNA-binding) and terminally bound galactose (RCA-binding) were identified on several polypeptides. Sterols (β-sitosterol and an additional unknown sterol, 40% of total lipids) and diacylglycerol (43%) are the major lipid components in the PMVs of C. paradoxa. Minor components are PE, PC, PC and an unknown glycolipid. Fatty acid analysis revealed 12 different fatty acids, predominantly 16:0,18:0 and the polyunsaturated acids 20:4 and 20:5 (together 69% of total fatty acids), α-Linolenic acid, common in higher plant PMs, could only be detected as a minor constituent (< 1 %) of PMVs of C. paradoxa.  相似文献

Very long-chain polyunsaturated fatty acids (VLC-PUFAs) are important dietary requirements for maintaining human health. Many marine microalgae are naturally high in ω − 3 VLC-PUFAs, however, the molecular mechanisms underpinning fatty acid (FA) desaturation and elongation in algae are poorly understood. An advanced molecular understanding would facilitate improvements of this nascent industry. We aimed to investigate expression responses of four front-end fatty acid desaturase genes and downstream effects on FA profiles to nitrogen limitation and cultivation growth stage in Isochrysis aff. galbana (TISO). Cultures were grown in nitrogen-replete and -deplete medium; samples were harvested during logarithmic, late logarithmic and stationary growth phases to analyse FA content/composition and gene expression of ?6-, ?8-, ?5- and ?4-desaturases (d6FAD (putative), d8FAD, d5FAD and d4FAD, respectively). d6FAD (putative) exhibited no differential expression, while d8FAD, d5FAD and d4FAD were significantly upregulated during logarithmic growth of nutrient-replete cultures, coinciding with rapid cell division. In conclusion, it is demonstrated that expression of some FADs in I. aff. galbana varies with culture age and nitrogen status which has downstream consequences on FA desaturation levels. This has implications for the commercial production of VLC-PUFAs where a trade-off between total lipid yield and VLC-PUFAs has to be made.  相似文献

The resistance of a plant community against herbivore attack may depend on plant species richness, with monocultures often much more severely affected than mixtures of plant species. Here, we used a plant–herbivore system to study the effects of selective herbivory on consumption resistance and recovery after herbivory in 81 experimental grassland plots. Communities were established from seed in 2002 and contained 1, 2, 4, 8, 16 or 60 plant species of 1, 2, 3 or 4 functional groups. In 2004, pairs of enclosure cages (1 m tall, 0.5 m diameter) were set up on all 81 plots. One randomly selected cage of each pair was stocked with 10 male and 10 female nymphs of the meadow grasshopper, Chorthippus parallelus. The grasshoppers fed for 2 months, and the vegetation was monitored over 1 year. Consumption resistance and recovery of vegetation were calculated as proportional changes in vegetation biomass. Overall, grasshopper herbivory averaged 6.8%. Herbivory resistance and recovery were influenced by plant functional group identity, but independent of plant species richness and number of functional groups. However, herbivory induced shifts in vegetation composition that depended on plant species richness. Grasshopper herbivory led to increases in herb cover at the expense of grasses. Herb cover increased more strongly in species-rich mixtures. We conclude that selective herbivory changes the functional composition of plant communities and that compositional changes due to selective herbivory depend on plant species richness.  相似文献

The core protein of pestiviruses is released from the polyprotein by viral and cellular proteinases. Here we report on an additional intramembrane proteolytic step that generates the C terminus of the core protein. C-terminal processing of the core protein of classical swine fever virus (CSFV) was blocked by the inhibitor (Z-LL)(2)-ketone, which is specific for signal peptide peptidase (SPP). The same effect was obtained by overexpression of the dominant-negative SPP D(265)A mutant. The presence of (Z-LL)(2)-ketone reduced the viability of CSFV almost 100-fold in a concentration-dependent manner. Reduction of virus viability was also observed in infection experiments using a cell line that inducibly expressed SPP D(265)A. The position of SPP cleavage was determined by C-terminal sequencing of core protein purified from virions. The C terminus of CSFV core protein is alanine(255) and is located in the hydrophobic center of the signal peptide. The intramembrane generation of the C terminus of the CSFV core protein is almost identical to the processing scheme of the core protein of hepatitis C viruses.  相似文献
