为了解茶褐牛肝菌(Neoboletus brunneissimus)的化学成分,共分离鉴定了18个化合物,经理化性质和波谱分析,分别鉴定为5α,8α-环二氧-(22E,24R)-麦角甾-6,22-二烯-3β-醇(1)、5α,8α-环二氧-(22E,24R)-麦角甾-6,9(11),22-三烯-3β-醇(2)、(22E,24R)-麦角甾-7,22-二烯-3β-醇(3)、(24S)-麦角甾-7-烯-3β-醇(4)、(22E,24R)-麦角甾-7,22-二烯-3β,5α,6β-三醇(5)、(24S)-乙基胆甾烯-7-烯-3β,5α,6β-三醇(6),(22E,24R)-麦角甾-7,22-二烯-3β,5α,6β,9α-四烯(7)、(22E,24R)-麦角甾-5,7,2-三烯-3β-醇(8)、3β-O-吡喃葡萄糖基-5α,8α-环二氧-(22E,24R)-麦角-6,22-二烯(9)、富马酸单甲酯(10)、富马酸(11)、琥珀酸(12)、反-2-癸烯二酸(13)、烟酸(14)、烟酰胺(15)、莽草酸(16)、亚油酸-α-甘油酯(17)和1-O-吡喃葡萄糖基-(2S,3R,4E,8E,2′R)-2-N-(2′-羟基棕榈酰)-9-甲基-4,8-鞘氨醇(18)。以上化合物均为首次从茶褐牛肝菌中分离得到,其中化合物6、9、13和17为首次从牛肝菌科高等真菌中分离得到。  相似文献

本工作用12%乙醇麻醉的大鼠,观察了下丘脑室旁核(PVH)微量注射K型阿片受体激动剂U-50,488H对大鼠肾水钠钾排出的影响,以及第三脑室注射U-50,488H对PVH中多巴胺神经元活性的影响。结果如下:(1)PVH微量注射U-50,488H(5μn/ul)后20min内大鼠尿量开始增加(P<0.01),持续约100min,41—60min尿量增加达峰值(P<0.001)。(2)PVH预先(10min)注射K型阿片受体阻断剂NBT(Nor-BinaltorphimineTetrahydrate)(5μg/pl)可以阻断U-50,488H所产生的利尿效应(P<0.01)。(3)第三脑室注射U-50,488H(10μg/10ul)20min后,PVH中酪氨酸羟化酶免疫反应阳性(Tyrosinehydroxylase-immunoreactivity,TH-IR)神经元数量减少,染色强度减弱,于注药后50min变化最为显著,100min时已恢复正常。上述结果表明:PVH微量注射U-50,488H可作用于K型阿片受体引起利尿效应;第三脑室微量注射U-50,488H可抑制PVH中TH-IR神经元的免疫活性。  相似文献

Calmodulin (CAM) is recognized as a major intermediary in intracellular calcium signaling, but as yet little is known of its role in developmental and behavioral processes. We have generated and studied mutations to the endogenous Cam gene of Drosophila melanogaster that change single amino acids within the protein coding region. One of these mutations produces a striking pupal lethal phenotype involving failure of head eversion. Various mutant combinations produce specific patterns of ectopic wing vein formation or melanotic scabs on the cuticle. Anaphase chromosome bridging is also seen as a maternal effect during the early embryonic nuclear divisions. In addition, specific behavioral defects such as poor climbing and flightlessness are detected among these mutants. Comparisons with other Drosophila mutant phenotypes suggests potential CAM targets that may mediate these developmental and behavioral effects, and analysis of the CAM crystal structure suggests the structural consequences of the individual mutations.  相似文献

The erythroid- and developmental stage-specific expression of the human ε-globin gene is controlled,in part,by the 5‘-flanking DNA sequence of this gene.In the present study,we have used DNA-protein binding assays to identify trans-acting factors which regulate the temporal expression of the human ε-globin gene during development.Using gel mobility shift assays and DNaseI footprinting assays,a nuclear protein factor (termed ε-SSF1) in the nuclear extracts from mouse haematopoietic tissues at d 11 and d 13 of gestation was identified.It could specifically bind to the positive control region (between-535 and -453bp) of the human ε-globin gene.We speculated that the ε-SSF1 might be an erythroid-and developmental stage-specific activator.In addition,we found another nuclear protein factor (terned ε-R1) in the nuclear extract from mouse fetal liver at d18 of gestation,which could strongly bind to the silencer region (between-392 and -177bp) of this gene.Therefore,we speculated that the ε-R1 might be an erythroid-and developmental stagespecific repressor.Our data suggest that both ε-SSF1 and ε-R1 might play important roles in developmental regulation of the human ε-globin gene expression during the early embryonic life.On the hand,we observed that the binding patterns of nuclear proteins from three cell lines (K562,HEL and Raji) to these regulatory regions were partially different.These results suggest that different trans-acting factors in K562,HEL and Raji cells might be responsible for activating or silencing the human ε-globin gene in three different cell lines.  相似文献

Leptin, the product of the ob gene, reduces body fat in genetically obese animals and circulates in elevated concentrations in the blood of obese patients. Polymorphic markers situated in the proximity of the human ob gene have recently been suggested to be linked to morbid obesity. We have studied the possible association between the microsatellite markers near the ob gene and morbid obesity in 252 morbidly obese patients with a mean body mass index (BMI) of 43 ± 7 kg/m2, and 151 lean controls with a mean BMI of 22 ± 2 kg/m2, and searched for linkage of these gene markers to obesity in 76 affected sib-pairs (BMI ≥ 32). No significant association was observed between any of the eight microsatellite markers and morbid obesity, and affected-sib-pair analysis failed to show linkage of three selected ob gene markers to obesity in the sibships. There was a strong positive correlation between serum leptin levels and BMI in morbidly obese patients; a carrier status for either of the two most prevalent alleles of the microsatellite marker D7S530 in the vicinity of the ob gene was associated with serum leptin levels in the obese subjects. Two of the markers (D7S2519, D7S649) showed a significant relation to the weight-losing response to a 16-week very-low-calorie dietary intervention. We have thus been able to confirm a tight relationship between serum leptin and body mass but have found no evidence for genetic linkage of the ob gene markers to morbid obesity in a population considered to represent a genetic isolate and to be an ideal model for studies of complex disorders. Abstract:  This paper describes a new genus and species of dyrosaurid, Chenanisuchus lateroculi gen. et sp. nov. (Crocodyliformes, Mesoeucrocodylia) from the Thanetien (Late Palaeocene) of the Oulad Abdoun Basin, Morocco. This new taxon has a particularly short snout, as well as widely separated and laterally facing orbits. In the holotype, the mandible exhibits a retroarticular process that is strongly depressed posterior to the glenoid fossa, bringing the ventral margin of the medial wing of the articular to the same level as the ventral margin of the retroarticular process. This feature is shared with Congosaurus bequaerti , Dyrosaurus and isolated dyrosaurid material from Mali and Niger, but is absent in the putative closely related crocodyliforms, such as pholidosaurids and Terminonaris , and could be a dyrosaurid character.  相似文献

We investigated concentration-dependent primary and secondary substrate relationships in the simultaneous metabolism of the ubiquitous pollutant phenol and the naturally occurring substrate acetate by a Pseudomonas sp. soil isolate capable of utilizing either substance as a sole source of carbon and energy. In addition to conventional analytical techniques, solid-state 13C nuclear magnetic resonance spectroscopy was used to follow the cellular distribution of [1-13C]acetate in the presence of unlabeled phenol. With 5 mM acetate as the primary substrate, Pseudomonas sp. 9S8D2 removed 1 mM phenol (secondary substrate) at a rate of 2 nmol/mg of total cell protein. Although extensive acetate metabolism was indicated by a significant redistribution of the carboxyl label, this redistribution was not affected by the presence of phenol as a secondary substrate. When the primary and secondary substrate roles were reversed, however, the presence of 1 mM phenol altered the metabolism of 0.1 mM acetate, as evidenced by both the two- to fourfold increases in carboxyl label that appeared in terminal methyl and acyl chain methylene carbon resonances and the decrease in label that occurred in the carbohydrate spectral region. These results suggest that, when phenol is present as the primary substrate, acetate is preferentially shuttled into fatty acyl chain synthesis, whereas phenol carbon is funnelled into the tricarboxylic acid cycle. Thus, simultaneous use of a xenobiotic compound and a natural substrate apparently does occur, and the relative concentrations of the two substrates do influence the rate and manner in which the compounds are utilized.(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献
