Background

Acute myocardial infarction (AMI) launches an inflammatory response and a repair process to compensate cardiac function. During this process, the balance between proinflammatory and anti-inflammatory cytokines is important for optimal cardiac repair. Stem cell transplantation after AMI improves tissue repair and increases the ventricular ejection fraction. Here, we studied in detail the acute effect of bone marrow mononuclear cell (BMMNC) transplantation on proinflammatory and anti-inflammatory cytokines in patients with ST segment elevation myocardial infarction (STEMI).

Methods

Patients with STEMI treated with thrombolysis followed by percutaneous coronary intervention (PCI) were randomly assigned to receive either BMMNC or saline as an intracoronary injection. Cardiac function was evaluated by left ventricle angiogram during the PCI and again after 6 months. The concentrations of 27 cytokines were measured from plasma samples up to 4 days after the PCI and the intracoronary injection.

Results

Twenty-six patients (control group, n = 12; BMMNC group, n = 14) from the previously reported FINCELL study (n = 80) were included to this study. At day 2, the change in the proinflammatory cytokines correlated with the change in the anti-inflammatory cytokines in both groups (Kendall’s tau, control 0.6; BMMNC 0.7). At day 4, the correlation had completely disappeared in the control group but was preserved in the BMMNC group (Kendall’s tau, control 0.3; BMMNC 0.7).

Conclusions

BMMNC transplantation is associated with preserved balance between pro- and anti-inflammatory cytokines after STEMI in PCI-treated patients. This may partly explain the favorable effect of stem cell transplantation after AMI.  相似文献

The effects of dynamic and intermittent isometric knee extension exercises on skeletal muscle blood flow and flow heterogeneity were studied in seven healthy endurance-trained men. Regional muscle blood flow was measured using positron emission tomography (PET) and an [(15)O]H(2)O tracer, and electromyographic (EMG) activity was recorded in the quadriceps femoris (QF) muscle during submaximal intermittent isometric and dynamic exercises. QF blood flow was 61% (P = 0.002) higher during dynamic exercise. Interestingly, flow heterogeneity was 13% (P = 0.024) lower during dynamic compared with intermittent isometric exercise. EMG activity was significantly higher (P < 0.001) during dynamic exercise, and the change in EMG activity from isometric to dynamic exercise was tightly related to the change in blood flow in the vastus lateralis muscle (r = 0.98, P < 0.001) but not in the rectus femoris muscle (r = -0.09, P = 0.942). In conclusion, dynamic exercise causes higher and less heterogeneous blood flow than intermittent isometric exercise at the same exercise intensity. These responses are, at least partly, related to the increased EMG activity.  相似文献

Conventional spectral analyses of heart rate variability (HRV) have been limited to stationary signals and have not allowed the obtainment of information during transient autonomic cardiac responses. In the present study, we evaluated the ability of the short-time Fourier transform (STFT) method to detect transient changes in vagal effects on the heart. We derived high-frequency power (HFP, 0.20-0.40 Hz) as a function of time during active orthostatic task (AOT) from the sitting to standing posture before and after selective vagal (atropine sulfate 0.04 mg/kg) and sympathetic (metoprolol 0.20 mg/kg) blockades. The HFP minimum point during the first 30 s after standing up was calculated and compared with sitting and standing values. Reactivity scores describing the fast and slow HFP responses to AOT were calculated by subtracting the minimum and standing values from the sitting value, respectively. The present results, obtained without controlled respiration, showed that in the drug-free condition, HFP decreased immediately after standing up (P < 0.001) and then gradually increased toward the level characteristic for the standing posture (P < 0.001), remaining lower than in the sitting baseline posture (P < 0.001). The magnitudes of the fast and slow HFP responses to AOT were abolished by the vagal blockade (P < 0.001) and unaffected by the sympathetic blockade. These findings indicate that HFP derived by the STFT method provided a tool for monitoring the magnitude and time course of transient changes in vagal effects on the heart without the need to interfere with normal control by using blocking drugs.  相似文献

The kin selection theory has recently been criticised on the basis of claiming that genetic relatedness does not play a causal role in the social evolution among individuals of insect societies. We outline here a line of criticism of this view by demonstrating two things. First, there are strong conceptual, theoretical and empirical reasons to think that close genetic relatedness has been necessary for the rise of the helper castes of social insects. And second, once we understand how causal explanation itself results from an interplay of two logically distinct elements, necessity and sufficiency, we can also understand the scenarios in which relatedness does not seem to play a causal role for evolution of helper castes. The result of this analysis is that we should be more careful about the way we frame the empirical data on the evolution of social behaviour.  相似文献

Thin-layer chromatography (TLC), gas chromatography (GC), and liquid chromatography (LC) in combination with mass spectrometry (MS) have been adopted for the isolation and identification of oxolipids and for determining their functionality. TLC provides a rapid separation and access to most oxolipids as intact molecules and has recently been effectively interfaced with time-of-flight (TOF) MS (TOF-MS). GC with flame ionization (FI) (GC/FI) and electron impact (EI) MS (GC/EI-MS) has been extensively utilized in the analysis of isoprostanes and other low-molecular-weight oxolipids, although these methods require derivatization of the analytes. In contrast, LC with ultraviolet (UV) absorption (LC/UV) or evaporate light scattering detection (ELSD) (LC/ELSD) as well as electrospray ionization (ESI) or atmospheric pressure chemical ionization (APCI) MS (LC/ESI-MS) or LC/APCI-MS has proven to be well suited for the analysis of intact oxolipids and their conjugates without or with minimal derivatization. Nevertheless, kit-based colorimetric and fluorescent procedures continue to serve as sensitive indicators of the presence of hydroperoxides and aldehydes.

The efficacy of two Finnish strains of Metarhizium anisopliaeagainst the rape blossom beetle Meligethes aeneus (Coleoptera:Nitidulidae) and the effect on its parasitoids Phradismorionellus (Hymenoptera: Ichneumonidae) and Diospilus capito(Hymenoptera: Braconidae) were studied in laboratory and semi-fieldexperiments. Several bio-assays were performed using either directexposure of the host beetle to the pathogens, or pots containing soilinoculated with M. anisopliae conidia at a rate of 2 ×108 per ml soil. Parasitised and unparasitised M. aeneuslarvae were collected in the field and were placed for pupation in thetest soil. The effect of treatment on M. aeneus andD. capito was estimated after adult emergence, and the effecton P. morionellus was estimated by dissecting hibernating pupae.While both of the M. anisopliae strains were highly pathogenicto M. aeneus adults and larvae upon direct exposure, soiltreatment resulted in no significant difference between the number ofinsects emerging (or found) from the treated and untreatedpots. However, the treated insects appeared to be latently infectedand the actual infection rate had to be estimated after their deathand incubation in a moist chamber. The mortality of the beetles causedby direct exposure to the fungus was 85% (range 70–88%). The rateof latent infection following indirect exposure via treated soil indeep pots in laboratory was 80% (range 49–100%), while that inP. morionellus was only 17% (range 0–85%), and inD. capito significantly higher, 76% (range 72–100%).The results indicate that M. anisopliae is a potentially usefulcandidate to be used as a bio-insecticide to control the pollenbeetle, and that at least the most abundant parasitoid, Phradismorionellus, is less affected than the target pest itself. Thepossible ecological role of latent infections in the host as well asin some of the parasitoids needs to be clarified.  相似文献

Selective feeding by herbivores on establishing seedlings has been suggested to affect genotype frequencies in several plant populations. The existence of genotypes susceptible to herbivores calls for an explanation in such populations. In the present study we assessed the choice of multiple herbivores, field voles (Microtus agrestis) and insects, among genotypes of silver birch (Betula pendula) representing variation occurring in a naturally regenerated stand. We examined how food choice of voles and insects is related to each other, competitive ability among the seedling genotypes and variation in soil fertility. We set up a field experiment and randomly assigned seedling populations, composed of mixed genotypes, to fully crossed insect exposure and fertilization treatments. After the first growing season we exposed a half of the seedling populations to vole herbivory. Voles selected clearly among the genotypes: they preferred the fastest growing seedlings as well as those with a low density of resin droplets on their stems. The preference of voles and insects among the genotypes was tightly correlated. We conclude that the effects of herbivory compensate those of intraspecific competition in this system and thus favor coexistence of genotypes differing in their susceptibility to herbivores.  相似文献
