Mast cells (MCs) are responsible for the innate immune response. Rat MCs are more suitable than mouse MCs as models of specific parasite infection processes and ovalbumin-induced asthma. Rat peritoneum-derived MCs and RBL-2H3 cells (an MC cell line) are widely used in disease studies. However, the application of rat bone marrow-derived MCs (BMMCs) are poorly documented in terms of the methodology of rat BMMC isolation. Here, we describe a relatively rapid, efficient, and simple method for the cultivation of rat BMMCs. As compared to previous protocols, rat BMMCs produced with the proposed protocol exhibited advantages in differentiation, proliferation, lifespan, and functionality, which should prove useful for studies of mucosal MC diseases in specific rat models. 相似文献

Zinc plays a role in alleviating oxidative stress. However, the related mechanisms remain to be further elucidated. The present study was conducted to investigate whether the recovery of oxidative stress in high-fat-diet (HFD)-pretreated mice was affected by zinc. Male mice received either an HFD or a low-fat-diet (LFD) for 8 weeks. Then, the mice fed with HFD and LFD were both assigned to either a control diet (30 mg zinc, ZD) or a no-added zinc diet (NZD) for an additional 4 weeks. The results showed that after feeding with NZD for 4 weeks, the HFD-pretreated mice had the highest plasma glucose and insulin concentrations, while had the lowest CuZn-SOD and glutathione concentrations. Moreover, after feeding with NZD for 4 weeks, the HFD-pretreated mice had the highest hepatic ROS and homocysteine concentrations, while had the lowest glutathione and methionine concentrations. Furthermore, the HFD-pretreated mice fed with NZD for 4 weeks had the lowest gene and protein expression of betaine homocysteine-S-methyltransferase (BHMT), cystathionine β-synthase, and Sp1. The results suggested that zinc was critical for oxidative stress alleviation and homocysteine clearance in HFD-pretreated mice. Alzheimer’s disease (AD), a complex chronic progressive central nervous system degenerative disease and a public health problem of the world, often characters cognitive dysfunction accompaning aggression and depression, and may lead to death. More attentions should be paid on it because there is no modified strategy against AD till now. AD is featured with the loss of cholinergic neurons, the amyloid-beta peptide (Aβ) plaques and the neurofibrillary tangles and several hypotheses were established to explain the pathogenesis of AD. Hydroxycinnamic acids, including caffeic acid (CA) and ferulic acid (FA) are widely distributed in natural plants and fruits. CA and FA exert various pharmacological activities, including anti-inflammatory, antioxidant, neuroprotection, anti-amyloid aggregation and so on. All these pharmacological activities are associated with the treatment of AD. Here we summarized the pharmacological activities of CA and FA, and their hybrids as multi-target-directed ligands (MTDLs) against AD. The future application of CA and FA was also discussed, hoping to provide beneficial information for the development of CA- and FA-based MTDLs against AD. 相似文献

Bocaviruses are associated with many human infectious diseases, such as respiratory tract infections, gastroenteritis, and hepatitis. Rats are known to be reservoirs of bocaviruses, including rodent bocavirus and rat bocavirus. Recently, ungulate bocaparvovirus 4, a known porcine bocavirus, has also been found in rats. Thus, investigating bocaviruses in rats is important for determining the origin of the viruses and preventing and controlling their transmission. To the best of our knowledge, no study to date has investigated bocaviruses in the livers of rats. In this report, a total of 624 rats were trapped in southern China between 2014 and 2017. Liver and serum samples from rats were tested for the prevalence of bocaviruses using PCR. Sequences related to ungulate bocaparvovirus 4 and rodent bocavirus were detected in both liver and serum samples. Interestingly, the prevalence of ungulate bocaparvovirus 4 (reference strain:KJ622366.1) was higher than that of rodent bocavirus (reference strain:KY927868.1) in both liver (2.24% and 0.64%, respectively) and serum samples (2.19% and 0.44%, respectively). The NS1 regions of ungulate bocaparvovirus 4 and rodent bocavirus related sequences displayed over 84% and 88% identity at the nucleic acid and amino acid levels, respectively. Furthermore, these sequences had similar genomic structure, genomic features, and codon usage bias, and shared a common ancestor. These viruses also displayed greater adaptability to rats than pigs. Our results suggested that ungulate bocaparvovirus 4 and rodent bocavirus may originate from rats and may be different genotypes of the same bocavirus species. 相似文献

The photophysical properties of five blue light-emitting polymers based on spirobifluorene applied in polymer light-emitting diodes (PLED) materials have been studied by quantum chemistry. In order to understand the intrinsic reasons for the different performances displayed by the polymers, we carried out density functional theory (DFT) and Marcus theory investigations on their oligomers in terms of structure and properties stability, absorption and emission properties, and carrier injection and transport properties. Especially, some important parameters which had not been reported to our knowledge were given in this contribution, such as the ionization potentials (IPs), electron affinities (EAs), reorganization energies (λ), k e /k h (the ratio between the electron transfer rate (k e ) and hole transfer rate (k h )), and the radiative lifetimes (τ). The main results indicate that the co-oligomers of PCC-1, PCC-2, and PCC-3 with push-pull interactions produced by the existing D-A segments have better carrier injection and transport properties than the oligomers of PSF and PCF. Especially PCC-2 co-oligomer, its large radiation lifetime (7.46 ns) and well balanced and adequate carrier transport guarantee its champion performance for PLED. The calculated results coincide with the experimental ones. Besides, PNF structurally similar to PCC-2 has similar photoelectric properties to PCC-2 in theory, and the fluorescence emission of PNF co-oligomer is superior to PCC-2 co-oligomer. In this communication, the effect of acute treatment with lamotrigine (LTG) was investigated on release of main excitatory amino acids (EAA) such as glutamate (Glu) and aspartate (Asp) in the hippocampus of pentylenetetrazol (PTZ)-induced and PTZ-kindled freely moving rats using micro dialysis. The results show that, levels of Glu and Asp significantly increased in the rat hippocampus during the seizure/interical periods for PTZ-status epilepticus (SE) and PTZ-kindled epileptic (EP) rats. The levels of Glu and Asp increased more in EP rat hippocampus than in SE rat hippocampus. After administration of 20 mg/kg LTG, the levels of Glu and Asp significantly decreased in the SE and EP rat hippocampus. The results indicate that: (a) excitability of the PTZ-kindled epileptogenic model is higher than that of the status epilepticus model; (b) the modulation of LTG on the EAA neurotransmitters certainly plays an important role in antiepileptic efficacy, especially in PTZ-kindled epileptic model where the release of EAA was influenced more markedly by acute application of 20 mg/kg LTG. 相似文献

目的:探寻军医大学学员内隐层面对自我和他人坚毅性评价的特点及脑电特征,为全面、客观的评估个体的坚毅性提供理论依据和客观指标。方法:使用E-Prime2.0参照经典内隐联想范式编制内隐联想-坚毅测验,对100名军医大学学员施测坚毅量表(Grit O),选取高、低坚毅水平被试(各20名)进行内隐联想-坚毅测验,并记录脑电,分析两组被试的内隐效应及主要脑电成分。结果:计算内隐效应D值,t检验显示高坚毅组(0.55±0.36)显著低于低坚毅组(0.87±0.49),t=-2.257,P0.05,Cohen'd=0.74。两组被试均诱发明显的N400和LPP,高坚毅组中N400在任务状态下主效应显著,F(1,17)=8.528,P0.05,η2=0.334,且在电极位置上主效应显著,F(10,170)=8.207,P0.001,η2=0.326。LPP在任务状态下主效应显著,F(1,17)=5.471,P0.05,η2=0.243,且在电极位置上主效应显著,F(10,170)=18.479,P0.001,η2=0.521;低坚毅组中N400在任务状态下主效应显著,F(1,17)=10.051,P0.05,η2=0.372,且在电极位置上主效应显著,F(10,170)=8.223,P0.001,η2=0.326,LPP在任务状态下主效应不显著。结论:1.军医大学学员坚毅性评价的内隐效应显著,即均倾向于认为自我的坚毅性高,他人的坚毅性低,通过问卷法评估坚毅性时应考虑坚毅评价的内隐效应。2.高、低坚毅性军医大学学员坚毅性内隐评价时的主要脑电成分N400、LPP存在差异,N400可作为坚毅性内隐评价符合程度的判断指标。3.内隐效应及N400可以作为对军医大学学员坚毅性评价时的客观指标。 相似文献
