The purpose of this study was to investigate whether 6-min walk test (6MWT) would improve the forearm skin blood flow (FSBF) response to acetylcholine (ACh), an endothelium-dependent vasodilator, in Tunisian women over a wide range of BMI. The FSBF was measured noninvasively using a laser Doppler flowmeter in response to local infusion of a cumulative dose of ACh, before and after the 6MWT for 102 healthy women; the results were expressed as percentage of baseline. The 6MWT was monitored and recorded. The mean response of FSBF to ACh was significantly greater before as well as after the 6MWT in lean (1,235 ± 123% vs. 1,644 ± 140%) than in overweight (630 ± 62% vs. 1,080 ± 66%) and obese subjects (402 ± 38% vs. 795 ± 40%) (P < 0.0001). Our regression analysis also revealed that the maximal FSBF response to ACh (i.e., its efficacy) was inversely correlated with BMI both before as well as after the 6MWT (r = -0.828, P < 0.0001; r = -0.859, P < 0.0001, respectively), and the efficacies of ACh in the three groups were all significantly elevated following the 6MWT (P < 0.0001). As indicated by ANOVA test, the 6MWT improved the FSBF responses of the lean, overweight, and obese subjects, by 33, 71, and 98%, respectively. We confirm that obesity induced a reduction of skin vasodilatory reserve and altered both endothelial-dependent relaxation and wall compliance. However, our new data clearly demonstrated that the 6MWT not only improved significantly the FSBF responses in the three groups of women, but the obese patients appeared to benefit more from the 6MWT than the overweight and the lean subjects. 相似文献

Ingestion of rosemary (Rosmarinus officinalis L.) by two groups of adult Sprague-Dawley rats at levels of 250 and 500 mg/kg body wt for 63 days was investigated for its effects on fertility. Body weight and absolute and relative testes weights were not affected, but the average weights of epididymides, ventral prostates, seminal vesicles, and preputial glands decreased significantly. A significant decline in spermatogenesis in testes due to a decrease in the number of primary and secondary spermatocytes and spermatids in treatment group 2 (500 mg/kg) is attributed to a significant decrease in testosterone. Sperm motility and density were also significantly decreased in the cauda epididymis and in the testes of rosemary-treated male rats in group 2. In addition, the treatment markedly increased the number of fetal resorptions in female rats impregnated by group 2 males, thereby reducing their fertility. 相似文献

The fossil record contains abundant evidence for the activity of microorganisms in the form of characteristic decay structures within fossil plant remains. Despite an abundance of charcoal in many sedimentary environments, there is little published evidence of such decay structures within charcoal from pre-Quaternary clastic deposits. The present contribution presents some examples of pre-Quaternary charcoal from clastic sediments which exhibit pre-charring decay structures, stratigraphically reaching from the Permian up to the Oligocene. Examples include specimens affected by the principle types of wood rot known from modern ecosystems (i.e. brown-rot, white rot and soft-rot) as well as a peculiar decay pattern resembling an atypical type of white-rot, which is only rarely known from modern wood. Theoretically there are different, so far hypothetical, scenarios which could be used to explain the scarcity of published reports on such material. Besides taphonomical biases directly influencing the sedimentary record of charcoal towards material not affected by microbial decay, it is conceivable that the lack of reports of such material from pre-Quaternary clastic deposits represents a, maybe unintentional, bias introduced by scientists working on pre-Quaternary charcoal. 相似文献

This study explored the influence of triclosan (TCS) in the absence and presence of sodium fluoride (NaF) on estrogenic activity and thyroid function of adolescent female rats. The results indicated that the individual exposure to TCS evoked a significant decline in T3 and T4 but the levels of estradiol, FSH, and LH were significantly elevated beside marked up regulation of calbindin‐D9k and estrogen α mRNA expression. On the other hand, the single exposure to NaF causes insignificant changes in thyroid hormones, but evoked a trend toward an increase in both estradiol and LH levels. No significant differences in the TSH level were recorded among the experimental groups. The joint exposure to TCS and NaF induced a significant improvement in thyroid and reproductive hormone levels. Overall, these findings revealed that exposure to TCS resulted in significant endocrine and reproductive alterations in immature female rats, while TCS + NaF coexposure resulted in lessening most effects. 相似文献

In 9 healthy and 14 asthmatic subjects before and after astandard bronchial challenge and a modified [deep inspiration (DI), inhibited] bronchial challenge and after albuterol, we tracked airwaycaliber by synthesizing a method to measure airway resistance (Raw;i.e., lung resistance at 8 Hz) in real time. We determined the minimumRaw achievable during a DI to total lung capacity and the subsequentdynamics of Raw after exhalation and resumption of tidal breathing.Results showed that even after a bronchial challenge healthy subjectscan dilate airways maximally, and the dilation caused by a single DItakes several breaths to return to baseline. In contrast, at baseline,asthmatic subjects cannot maximally dilate their airways, and thisworsens considerably postconstriction. Moreover, after a DI, thedilation that does occur in airway caliber in asthmatic subjectsconstricts back to baseline much faster (often after a single breath).After albuterol, asthmatic subjects could dilate airways much closer tolevels of those of healthy subjects. These data suggest that theasthmatic smooth muscle resides in a stiffer biological state comparedwith the stimulated healthy smooth muscle, and inhibiting a DI inhealthy subjects cannot mimic this. 相似文献

SUMO proteins are important regulators of many key cellular functions in part through their ability to form interactions with other proteins containing SUMO interacting motifs (SIMs). One characteristic feature of all SUMO proteins is the presence of a highly divergent intrinsically disordered region at their N-terminus. In this study, we examine the role of this N-terminal region of SUMO proteins in SUMO–SIM interactions required for the formation of nuclear bodies by the promyelocytic leukemia (PML) protein (PML-NBs). We demonstrate that the N-terminal region of SUMO1 functions in a paralog specific manner as an auto-inhibition domain by blocking its binding to the phosphorylated SIMs of PML and Daxx. Interestingly, we find that this auto-inhibition in SUMO1 is relieved by zinc, and structurally show that zinc stabilizes the complex between SUMO1 and a phospho-mimetic form of the SIM of PML. In addition, we demonstrate that increasing cellular zinc levels enhances PML-NB formation in senescent cells. Taken together, these results provide important insights into a paralog specific function of SUMO1, and suggest that zinc levels could play a crucial role in regulating SUMO1-SIM interactions required for PML-NB formation and function. 相似文献
