The dependence of downstream waters on upstream ecosystems necessitates an improved understanding of watershed-scale hydrological interactions including connections between wetlands and streams. An evaluation of such connections is challenging when, (1) accurate and complete datasets of wetland and stream locations are often not available and (2) natural variability in surface-water extent influences the frequency and duration of wetland/stream connectivity. The Upper Choptank River watershed on the Delmarva Peninsula in eastern Maryland and Delaware is dominated by a high density of small, forested wetlands. In this analysis, wetland/stream surface water connections were quantified using multiple wetland and stream datasets, including headwater streams and depressions mapped from a lidar-derived digital elevation model. Surface-water extent was mapped across the watershed for spring 2015 using Landsat-8, Radarsat-2 and Worldview-3 imagery. The frequency of wetland/stream connections increased as a more complete and accurate stream dataset was used and surface-water extent was included, in particular when the spatial resolution of the imagery was finer (i.e., <10 m). Depending on the datasets used, 12–60% of wetlands by count (21–93% of wetlands by area) experienced surface-water interactions with streams during spring 2015. This translated into a range of 50–94% of the watershed contributing direct surface water runoff to streamflow. This finding suggests that our interpretation of the frequency and duration of wetland/stream connections will be influenced not only by the spatial and temporal characteristics of wetlands, streams and potential flowpaths, but also by the completeness, accuracy and resolution of input datasets.  相似文献

Leptospermum scoparium or mānuka is a New Zealand native medicinal plant that produces essential oils with antimicrobial properties. This study investigated the arbuscular mycorrhizal fungi (AMF) community in mānuka by culture dependent (trap culture) and independent (denaturing gradient gel electrophoresis) approaches. Furthermore, to assess whether mycorrhizal inoculation could alter growth and essential oil composition of mānuka, plants of a single regional chemotype were grown in unsterilized soil and inoculated with five AMF isolates. Leaf essential oil compositions and yields were determined by microscale solvent extraction and gas chromatography-mass spectrometry (GC-MS) analysis. AMF inoculation significantly increased growth compared to uninoculated plants. Qualitative i.e. different relative proportions of compounds, which are distinctive in chemotypes and quantitative (i.e. absolute concentrations of compounds, expressed as mg/g of dry leaf or equivalent) effects of AMF inoculation on mānuka essential oil composition depended on the isolate. AMF inoculation modified the Gammaproteobacterial community on roots and this may have contributed to changes in essential oil composition. Overall, these results demonstrated that AMF can improve the growth of mānuka and affect plant secondary metabolites in leaves, which would be valuable in commercial essential oil production from plantation-grown mānuka.  相似文献

But for decades, philosophers of biology have regarded “de-extinction” as conceptually incoherent. Once a species is gone, it is gone forever. We argue that a range of metaphysical, biological, and ethical grounds for opposing de-extinction are at best inconclusive and that a pragmatic stance that allows for its possibility is more appealing.  相似文献

The fraction of IgG antibodies with anti-oligomeric Aβ affinity and surface sialic acid was compared between Octagam and Gammagard intravenous immunoglobulin (IVIG) using two complementary surface plasmon resonance methods. These comparisons were performed to identify if an elevated fraction existed in Gammagard, which reported small putative benefits in a recent Phase III clinical trial for Alzheimer’s Disease. The fraction of anti-oligomeric Aβ IgG was found to be higher in Octagam, for which no cognitive benefits were reported. The fraction and location of surface-accessible sialic acid in the Fab domain was found to be similar between Gammagard and Octagam. These findings indicate that anti-oligomeric Aβ IgG and total surface sialic acid alone cannot account for reported clinical differences in the two IVIG products. A combined analysis of sialic acid in anti-oligomeric Aβ IgG did reveal a notable finding that this subgroup exhibited a high degree of surface sialic acid lacking the conventional α2,6 linkage. These results demonstrate that the IVIG antibodies used to engage oligomeric Aβ in both Gammagard and Octagam clinical trials did not possess α2,6-linked surface sialic acid at the time of administration. Anti-oligomeric Aβ IgG with α2,6 linkages remains untested as an AD treatment.  相似文献

In Escherichia coli and the majority of β- and γ-proteobacteria, the fourth step of lipid A biosynthesis, i.e. cleavage of the pyrophosphate group of UDP-2,3-diacyl-GlcN, is carried out by LpxH. LpxH has been previously suggested to contain signature motifs found in the calcineurin-like phosphoesterase (CLP) family of metalloenzymes; however, it cleaves a pyrophosphate bond instead of a phosphoester bond, and its substrate contains nucleoside diphosphate moieties more common to the Nudix family rather than to the CLP family. Furthermore, the extent of biochemical data fails to demonstrate a significant level of metal activation in enzymatic assays, which is inconsistent with the behavior of a metalloenzyme. Here, we report cloning, purification, and detailed enzymatic characterization of Haemophilus influenzae LpxH (HiLpxH). HiLpxH shows over 600-fold stimulation of hydrolase activity in the presence of Mn2+. EPR studies reveal the presence of a Mn2+ cluster in LpxH. Finally, point mutants of residues in the conserved metal-binding motifs of the CLP family greatly inhibit HiLpxH activity, highlighting their importance in enzyme function. Contrary to previous analyses of LpxH, we find HiLpxH does not obey surface dilution kinetics. Overall, our work unambiguously establishes LpxH as a calcineurin-like phosphoesterase containing a Mn2+ cluster coordinated by conserved residues. These results set the scene for further structural investigation of the enzyme and for design of novel antibiotics targeting lipid A biosynthesis.  相似文献

Early environment can have a major impact on development, with family life known to play an important role. Longitudinal studies can therefore help increase our understanding of variance in cognitive abilities in young animals, as well as over time. We followed 22 marmosets (11 male and 11 female) from infancy through to early adolescence. At 3 months old, the marmosets were trained to reliably touch a rewarded stimulus. At 5 months, behavior was observed within the natal group. At 9 months, the marmosets were given a visual discrimination task to assess learning ability. Mann–Whitney U tests found no sex or family size differences in number of errors at 3 or 9 months. While no significant relationships were found between behavior in the family and learning at 3 months, significant negative correlations were found between duration spent in locomotion and learning errors (p = .05), as well as between frequency of calm vocalizations and learning errors (p = .001) at 9 months. A U-shape curve was found between amount of social play and learning at 9 months. Positive family interactions, including moderate amounts of play, as well as calm individual behavior, may therefore be important in learning. This study sheds light on cognitive development in much younger marmosets than previously studied, and helps increase understanding of how individual differences in learning may arise.  相似文献

ObjectiveThe aims of this study were to assess participatory methods for obtaining community views on child health research.BackgroundCommunity participation in research is recognised as an important part of the research process; however, there has been inconsistency in its implementation and application in Australia. The Western Australian Telethon Kids Institute Participation Program employs a range of methods for fostering active involvement of community members in its research. These include public discussion forums, called Community Conversations. While participation levels are good, the attendees represent only a sub-section of the Western Australian population. Therefore, we conducted a telephone survey of randomly selected households to evaluate its effectiveness in eliciting views from a broader cross-section of the community about our research agenda and community participation in research, and whether the participants would be representative of the general population. We also conducted two Conversations, comparing the survey as a recruitment tool and normal methods using the Participation Program.ResultsWhile the telephone survey was a good method for eliciting community views about research, there were marked differences in the profile of study participants compared to the general population (e.g. 78% vs 50% females). With a 26% response rate, the telephone survey was also more expensive than a Community Conversation. The cold calling approach proved an unsuccessful recruitment method, with only two out of a possible 816 telephone respondents attending a Conversation.ConclusionWhile the results showed that both of the methods produced useful input for our research program, we could not conclude that either method gained input that was representative of the entire community. The Conversations were relatively low-cost and provided more in-depth information about one subject, whereas the telephone survey provided information across a greater range of subjects, and allowed more quantitative analysis.  相似文献

Aim  Pleistocene climatic cycles have left marked signatures in the spatial and historical genetic structure of high‐latitude organisms. We examine the mitochondrial (cytochrome b) genetic structure of the singing vole, Microtus miurus (Rodentia: Cricetidae: Arvicolinae), a member of the Pleistocene Beringian fauna, and of the insular vole, Microtus abbreviatus, its putative sister species found only on the St Matthew Archipelago. We reconstruct the phylogenetic and phylogeographical structure of these taxa, characterize their geographical partitioning and date coalescent and cladogenetic events in these species. Finally, we compare the recovered results with the phylogenetic, coalescent and spatial genetic patterns of other eastern Beringian mammals and high‐latitude arvicoline rodents. Location  Continental Alaska (alpine and arctic tundra) and the St Matthew Archipelago (Bering Sea). Methods  We generated and analysed cytochrome b sequences of 97 singing and insular voles (M. miurus and M. abbreviatus) from Alaska. Deep evolutionary structure was inferred by phylogenetic analysis using parsimony, maximum likelihood and Bayesian approaches; the geographical structure of genetic diversity was assessed using analysis of molecular variance and network analysis; ages of cladogenetic and coalescent events were estimated using a relaxed molecular clock model with Bayesian approximation. Results  Regional nucleotide diversity in singing voles is higher than in other high‐latitude arvicoline species, but intra‐population diversity is within the observed range of values for arvicolines. Microtus abbreviatus specimens are phylogenetically nested within M. miurus. Molecular divergence date estimates indicate that current genetic diversity was formed in the last glacial (Wisconsinan) and previous interglacial (Sangamonian) periods, with the exception of a Middle Pleistocene split found between samples collected in the Wrangell Mountains region and all other singing vole samples. Main conclusions  High levels of phylogenetic and spatial structure are observed among analysed populations. This pattern is consistent with that expected for a taxon with a long history in Beringia. The spatial genetic structure of continental singing voles differs in its northern and southern ranges, possibly reflecting differences in habitat distribution between arctic and alpine tundra. Our phylogenetic results support the taxonomic inclusion of M. miurus in its senior synonym, M. abbreviatus.  相似文献
