A 31 kDa polypeptide that accumulates in the thylakoids when maize leaves are chilled to 5°C in the light is characterized using monoclonal antibodies and analyses of chlorophyll-protein complexes. This polypeptide reacted with a monoclonal antibody, MLH2, that was specific for the 28 kDa polypeptide of the light-harvesting complex (LHCII) of pea leaves. On chilling leaves the appearance of a chlorophyll-protein complex having an apparent molecular weight of 31 kDa coincided with the appearance of a 31 kDa polypeptide and a decrease in the 29 kDa chlorophyll-protein, CP29. Returning the leaves to 25°C for 1 h produced a loss of both the 31 kDa chlorophyll-protein and 31 kDa polypeptide from the thylakoids, and an increase in the amount of CP29. Breakdown of the 31 kDa polypeptide in vitro was Mg2+-dependent and inhibited by EDTA and transition metal ions. It is suggested that the 31 kDa polypeptide may be a precursor of the apoprotein of CP29 and can bind chlorophyll. The appearance of the 31 kDa polypeptide correlated with a marked change in the 77 K fluorescence emission spectra of isolated LHCII particles, which did not revert with the disappearance of the 31 kDa on returning the leaves to 25°C for 1 h. The physiological significance of this spectral perturbation is discussed. 相似文献

A method of two-colour immunofluorescence staining has been developed to allow the simultaneous analysis of both surface and cytoplasmic antigens. This involves the use of direct fluorochrome antibody conjugates for cell-surface antigen staining, followed by cell permeabilization and the staining of cytoplasmic antigens with biotinylated antibodies and streptavidin-fluorochrome conjugates. Fluorochrome-antibody conjugates bound to cell-surface epitopes were found not to be affected by the subsequent permeabilisation and cytoplasmic staining. This method was used to examine the surface phenotype of T cells expressing a cytoplasmic antigen, STA. STA is a unique determinant detected in activated human T cells by the monoclonal antibody K-1-21, which also recognizes a cross-reactive conformation-dependent epitope on human free kappa light chains. Cytometric analysis showed that STA is found in both Leu 2a+ cytotoxic/suppressor T cells and Leu 3a+ helper/inducer T cells but is not induced in the Leu 15+ population which contains suppressor T cells. We used a simplified probe detection system for positron-emitting radionuclides in order to measure blood-brain barrier transport of amino acids in anesthetized dogs. Plasma and brain time-activity curves were recorded after intravenous bolus injection of L-[11C]methionine before and after administration of 1 microgram of vasopressin. Three-compartment models with three or four transfer coefficients were used to derive the kinetics of L-[11C]methionine uptake in brain. The blood-brain clearance of the tracer (K1) was 0.075 ml ml-1 min-1 before and 0.041 ml ml-1 min-1 after injection of vasopressin. The partition volume and the initial distribution (plasma) volume of methionine were unchanged and within the expected limits. The net accumulation rate of methionine (K), estimated by both the four-parameter (kinetic) and three-parameter (graphic) approaches, decreased after vasopressin injection in all six studies. 相似文献

The gene for Huntington disease (HD) has been localized close to the telomere on the short arm of chromosome 4. However, refined mapping using recombinant HD chromosomes has resulted in conflicting findings and mutually exclusive candidate regions. Previously reported significant nonrandom allelic association between D4S95 and HD provided support for a more proximal location for the defective gene. In this paper, we have analyzed 17 markers, spanning approximately 6 Mb of DNA distal to locus D4S62, for nonrandom association to HD. We confirm the previous findings of nonrandom allelic association between D4S95 and HD. In addition, we provide new data showing significant nonrandom association between HD and 3 markers at D4S133 and D4S228, which are approximately 3 Mb telomeric to D4S95. 相似文献

Metabolic syndrome, insulin resistance, prediabetes, and overt type 2 diabetes mellitus are associated with an accelerated atherosclerosis (atheroscleropathy). This quartet is also associated with multiple metabolic toxicities resulting in the production of reactive oxygen species. The redox stress associated with these reactive oxygen species contribute to the development, progression, and the final fate of the arterial vessel wall in prediabetic and diabetic atheroscleropathy.
