Small, temporally dynamic, biologically diverse isolated wetlands are among the most imperiled ecosystems, yet their conservation
is hindered by lack of protective legislation and mapping. As part of an effort to better understand isolated wetland ecology
in an area undergoing dramatic land use change, we mapped isolated wetlands in South Carolina’s Piedmont and Blue Ridge regions
using remote sensing and local ecological knowledge (LEK). Remote detection of isolated wetlands was limited by digital resource
resolution, topography, and wetland size. LEK was the most useful tool for locating small isolated wetlands. We sampled 10%
of the study area using LEK and discovered 44 wetlands with “isolated” characteristics, none of which had been identified
by remote sensing. Only 8 of 44 wetlands found through LEK could be identified using remote sensing after their discovery.
LEK fills a gap in cryptic ecosystem detection when adequate remotely sensed data are not available. Though effective, using
LEK is neither as rapid nor as repeatable as remote sensing. We suggest a two-pronged approach for finding cryptic ecosystems:
remote sensing coupled with LEK where data resolution is inadequate. For remote detection of isolated wetlands, we suggest
a minimum resolution of 0.33 m for Color Infrared, leaf-off, high-water photography. Despite great advances in remote sensing,
data are not uniformly available worldwide and LEK may serve as an effective tool for locating cryptic resources for biodiversity
conservation. Mapping cryptic resources will allow for more accurate resource and biodiversity conservation planning under

AMP-activated protein kinase (AMPK) is a crucial cellular energy sensor. Once activated by falling energy status, it promotes ATP production by increasing the activity or expression of proteins involved in catabolism while conserving ATP by switching off biosynthetic pathways. AMPK also regulates metabolic energy balance at the whole-body level. For example, it mediates the effects of agents acting on the hypothalamus that promote feeding and entrains circadian rhythms of metabolism and feeding behaviour. We evaluated the association of red coloration with senescence in sugar maple (Acer saccharum Marsh.) leaves by assessing differences in leaf retention strength and the progression of the abscission layer through the
vascular bundle of green, yellow, and red leaves of 14 mature open-grown trees in October 2002. Computer image analysis confirmed
visual categorization of leaves as predominantly green, yellow or red, and chemical quantification of leaf pigment concentrations
verified that leaf color reflected underlying differences in leaf biochemistry. Significantly lower chlorophyll concentrations
within red and yellow leaves indicated that senescence was more advanced in leaves from these color categories relative to
green leaves. Among leaf types, only red leaves contained high concentrations of anthocyanins. There were significant differences
in leaf retention capacity among color categories, with the petioles of green leaves being the most firmly attached to twigs,
followed by red and then yellow leaves. Microscopic analysis indicated that yellow leaves had the most advanced extension
of the abscission layer through the vasculature, with green and red leaves having significantly less abscission layer progression
than yellow. A more limited progression of the abscission layer through vascular bundles may be evidence of delayed leaf senescence
that could extend resorption of mobile leaf constituents. Together, results from this study suggest an association between

The complexity of natural ecosystems makes it difficult to compare the relative importance of abiotic and biotic factors and to assess the effects of their interactions on ecosystem development. To improve our understanding of ecosystem complexity, we initiated an experiment designed to quantify the main effects and interactions of several factors that are thought to affect nutrient export from developing forest ecosystems. Using a replicated 2 × 2 × 4 factorial experiment, we quantified the main effects of these factors and the factor interactions on annual calcium, magnesium, and potassium export from field mesocosms over 4 years for two Vermont locations, two soils, and four different tree seedling communities. We found that the main effects explained 56%–97% of total variation in nutrient export. Abiotic factors (location and soil) accounted for a greater percentage of the total variation in nutrient export (47%–94%) than the biotic factor (plant community) (2%–15%). However, biotic control over nutrient export was significant, even when biomass was minimal. Factor interactions were often significant, but they explained less of the variation in nutrient export (1%–33%) than the main effects. Year-to-year fluctuations influenced the relative importance of the main effects in determining nutrient export and created factor interactions between most of the explanatory variables. Our study suggests that when research is focused on typically used main effects, such as location and soil, and interactions are aggregated into overall error terms, important information about the factors controlling ecosystem processes can be lost. 相似文献

Matrix metalloproteinases (MMPs) degrade components of the extracellular matrix of the disc, but the presence of MMP-19 has not been explored. In other tissues, MMP-19 is known to act in proteolysis of the insulin-like growth factor (IGF) binding protein-3, thereby exposing this protein to make it available to influence cell behavior. MMP-19 also has been shown to inhibit capillary-like formation and thus play a role in the avascular nature of the disc. Using immunohistochemistry, normal discs from six subjects aged newborn through 10 years and 20 disc specimens from control donors or surgical patients aged 15-76 (mean age 40.2 years) were examined for immunolocalization of MMP-19; six Thompson grade I discs, five Thompson grade II, eight Thompson grade III, five Thompson grade IV, and one Thompson grade V discs were analyzed. The results indicate that in discs from young subjects, MMP-19 was uniformly localized in the outer annulus. In discs from adult donors and surgical patients, outer and inner annulus cells only occasionally showed MMP-19 localization. The greatest expression of MMP-19 was observed in young discs, and little expression was seen in older or degenerating discs. Because MMP-19 has been shown to regulate IGF-mediated proliferation in other tissues, its decline in the aging/degenerating disc may contribute to the age-related decrease in disc cell numbers. 相似文献

The AMP-activated protein kinase cascade is a sensor of cellular energy charge, and its existence provides strong support for the energy charge hypothesis first proposed by Daniel Atkinson in the 1960s. The system is activated in an ultrasensitive manner by cellular stresses that deplete ATP (and consequently elevate AMP), either by inhibiting ATP production (e.g., hypoxia), or by accelerating ATP consumption (e.g., exercise in muscle). Once activated, it switches on catabolic pathways, both acutely by phosphorylation of metabolic enzymes and chronically by effects on gene expression, and switches off many ATP-consuming processes. Recent work suggests that activation of AMPK is responsible for many of the effects of physical exercise, both the rapid metabolic effects and the adaptations that occur during training. Dominant mutations in regulatory subunit isoforms (gamma2 and gamma3) of AMPK, which appear to increase the basal activity in the absence of AMP, lead to hypertrophy of cardiac and skeletal muscle respectively. 相似文献

We investigated the effect of carbohydrate (CHO) ingestion before and during exercise and in combination on glucose kinetics, metabolism and performance in seven trained men, who cycled for 120 min (SS) at approximately 63% of peak power output, followed by a 7 kJ/kg body wt time trial (TT). On four separate occasions, subjects received either a placebo beverage before and during SS (PP); placebo 30 min before and 2 g/kg body wt of CHO in a 6.4% CHO solution throughout SS (PC); 2 g/kg body wt of CHO in a 25.7% CHO beverage 30 min before and placebo throughout SS (CP); or 2 g/kg body wt of CHO in a 25.7% CHO beverage 30 min before and 2 g/kg of CHO in a 6.4% CHO solution throughout SS (CC). Ingestion of CC and CP markedly (>8 mM) increased plasma glucose concentration ([glucose]) compared with PP and PC (5 mM). However, plasma [glucose] fell rapidly at the onset of SS so that after 80 min it was similar (6 mM) between all treatments. After this time, plasma [glucose] declined in both PP and CP (P < 0.05) but was well maintained in both CC and PC. Ingestion of CC and CP increased rates of glucose appearance (R(a)) and disappearance (R(d)) compared with PP and PC at the onset of, and early during, SS (P < 0.05). However, late in SS, both glucose R(a) and R(d) were higher in CC and PC compared with other trials (P < 0.05). Although calculated rates of glucose oxidation were different when comparing the four trials (P < 0.05), total CHO oxidation and total fat oxidation were similar. Despite this, TT was improved in CC and PC compared with PP (P < 0.05). We conclude that 1) preexercise ingestion of CHO improves performance only when CHO ingestion is maintained throughout exercise, and 2) ingestion of CHO during 120 min of cycling improves subsequent TT performance. 相似文献

In contrast to the more typical mock grant proposals or literature reviews, we describe the use of the creative essay as a novel tool for teaching human genetics at the college level. This method has worked well for both nonmajor and advanced courses for biology majors. The 10- to 15-page essay is written in storylike form and represents a student's response to the choice of 6-8 scenarios describing human beings coping with various genetic dilemmas. We have found this tool to be invaluable both in developing students' ability to express genetic concepts in lay terms and in promoting student awareness of genetic issues outside of the classroom. Examples from student essays are presented to illustrate these points, and guidelines are suggested regarding instructor expectations of student creativity and scientific accuracy. Methods of grading this assignment are also discussed. 相似文献
