Skim milk residues had a significant impact on the sensitivity to heat of a dairy isolate of the thermo-resistant, Streptococcus thermophilus. Cells of S. thermophilus (H) suspended in water or in milk had D values at 60°C of 2.0 and 14 min, respectively. Cells of S. thermophilus (H) attached to stainless steel in the presence of water or milk had D values at 60°C of 2.2 and 8.1 min, respectively. The attached cells in both experiments were heat-treated in the presence
of water. The increase in heat resistance could not be fully attributed to individual components (caseinate or whey) in the
milk. The potential for thermo-resistant streptococci to survive heat treatment in a dairy manufacturing plant is therefore

To determine extant patterns of population genetic structure in common ash and gain insight into postglacial recolonization processes, we applied multilocus-based Bayesian approaches to data from 36 European populations genotyped at five nuclear microsatellite loci. We identified two contrasting patterns in terms of population genetic structure: (1) a large area from the British Isles to Lithuania throughout central Europe constituted effectively a single deme, whereas (2) strong genetic differentiation occurred over short distances in Sweden and southeastern Europe. Concomitant geographical variation was observed in estimates of allelic richness and genetic diversity, which were lowest in populations from southeastern Europe, that is, in regions close to putative ice age refuges, but high in western and central Europe, that is, in more recently recolonized areas. We suggest that in southeastern Europe, restricted postglacial gene flow caused by a rapid expansion of refuge populations in a mountainous topography is responsible for the observed strong genetic structure. In contrast, admixture of previously differentiated gene pools and high gene flow at the onset of postglacial recolonization of western and central Europe would have homogenized the genetic structure and raised the levels of genetic diversity above values in the refuges. 相似文献

Soil microbial communities have a profound influence on soil chemical processes and subsequently influence tree nutrition and growth. This study examined how the addition of a commercial inoculum or forest‐collected soils influenced nitrogen (N) and phosphorus (P) dynamics, soil microbial community structure, and growth in Liriodendron tulipifera and Prunus serotina tree saplings. Inoculation method was an important determinant of arbuscular mycorrhizal fungi (AMF) community structure in both species and altered soil N dynamics in Prunus and soil P dynamics in Liriodendron. Prunus saplings receiving whole forest soil transfers had a higher rhizosphere soil carbon/nitrogen ratio and ammonia content at the end of the first growing season when compared to unmanipulated control saplings. Inoculation with whole forest soil transfers resulted in increased inorganic phosphorus in Liriodendron rhizosphere soils. The number of AMF terminal restriction fragments was significantly greater in rhizosphere soils of Liriodendron saplings inoculated with whole forest soil transfers and Prunus saplings receiving either inoculum source than control saplings. Forest soil inoculation also increased AMF colonization and suppressed stem elongation in Liriodendron after 16 months; conversely, Prunus AMF colonization was unchanged and stem elongation was significantly greater when saplings were inoculated with whole forest soil transfers. Longer term monitoring of tree response to inoculation will be essential to assess whether early costs of AMF colonization may provide long‐term benefits. This study provides insight into how practitioners can use microbial inoculation to alter AMF community structure and functioning, subsequently influencing tree growth and nutrient cycling during the restoration of degraded lands. 相似文献

The search for longevity‐determining genes in human has largely neglected the operation of genetic interactions. We have identified a novel combination of common variants of three genes that has a marked association with human lifespan and healthy aging. Subjects were recruited and stratified according to their genetically inferred ethnic affiliation to account for population structure. Haplotype analysis was performed in three candidate genes, and the haplotype combinations were tested for association with exceptional longevity. An HRAS1 haplotype enhanced the effect of an APOE haplotype on exceptional survival, and a LASS1 haplotype further augmented its magnitude. These results were replicated in a second population. A profile of healthy aging was developed using a deficit accumulation index, which showed that this combination of gene variants is associated with healthy aging. The variation in LASS1 is functional, causing enhanced expression of the gene, and it contributes to healthy aging and greater survival in the tenth decade of life. Thus, rare gene variants need not be invoked to explain complex traits such as aging; instead rare congruence of common gene variants readily fulfills this role. The interaction between the three genes described here suggests new models for cellular and molecular mechanisms underlying exceptional survival and healthy aging that involve lipotoxicity. 相似文献

Polyphenols represent a large family of plant secondary metabolites implicated in the prevention of various diseases such as cancers and cardiovascular diseases. The potato is a significant source of polyphenols in the human diet. In this study, we examined the expression of thirteen genes involved in the biosynthesis of polyphenols in potato tubers using real-time RT-PCR. A selection of five field grown native Andean cultivars, presenting contrasting polyphenol profiles, was used. Moreover, we investigated the expression of the genes after a drought exposure. We concluded that the diverse polyphenolic profiles are correlated to variations in gene expression profiles. The drought-induced variations of the gene expression was highly cultivar-specific. In the three anthocyanin-containing cultivars, gene expression was coordinated and reflected at the metabolite level supporting a hypothesis that regulation of gene expression plays an essential role in the potato polyphenol production. We proposed that the altered sucrose flux induced by the drought stress is partly responsible for the changes in gene expression. This study provides information on key polyphenol biosynthetic and regulatory genes, which could be useful in the development of potato varieties with enhanced health and nutritional benefits. 相似文献

The events which make possible the characteristic fusion of the cell membranes of embryonic myoblasts are known to involve modification of the cell membrane (Hausman, R.E., Dobi, E.T., Woodford, E.J., Petrides, S., Ernst, M. and Nichols E.B. (1986) Dev. Biol. 113, 40-48). Myoblasts from chick embryos were allowed to differentiate in gyrotory aggregate culture and the order of their membranes was measured by EPR. Two spin-labels which insert at different depths into the lipid bilayer were used. Measurement with the 5-nitroxystearate label showed an increase in myoblast membrane order (2T' parallel) from 0-15 h of culture and again from 26-38 h of culture. Measurement with the 12-nitroxystearate label showed the 0-15 h increase in order but the second increase was greatly reduced and shifted in time. While the specific sources of these changes in membrane order cannot yet be identified, the changes observed correlated well with known events of myogenic differentiation in vitro. The initial increase in membrane order occurred while the myoblasts were recovering from the effects of trypsin dissociation and undergoing gyrotory aggregation. The second increase in membrane order occurred during the known period of prostaglandin receptor activity and increased cell-cell adhesion. 相似文献
